Hypolipidaemic and antiatherosclerotic effects of plumbagin in rabbits.

Plumbagin (2-methyl-5-hydroxy, 1:4 naphthoquinone) isolated from the roots of Plumbago zeylanica when administered to hyperlipidaemic rabbits, reduced serum cholesterol and LDL-Chol. by 53 to 86 percent and 61 to 91 percent respectively. It lowered cholesterol/phospholipid ratio by 45.8 percent and elevates the decreased HDL-Chol significantly. Further, Plumbagin treatment prevented the accumulation of cholesterol and triglycerides in liver and aorta and regressed atheromatous plaques of thoracic and abdominal aorta. Plumbagin treated hyperlipidaemic subjects excreted more fecal cholesterol and phospholipids. In conclusion-Plumbagin feeding brings about a definite regression of atheroma and prevents the accumulation of cholesterol and triglycerides in liver and aorta.

Testicular cell population dynamics following palmitine hydroxide treatment in male dogs.

Palmitine hydroxide isolated from the roots of Berberis chitria administered orally to dogs 30 mg/kg per day for 60 days brings about a consistent impairment of primary and secondary spermatocytes and elongated spermatids (Stages IV-VIII). The primary and secondary spermatocytes were reduced by 60 and 68%, respectively, and the elongated spermatids were decreased by 58%. The number of spermatogonia and Sertoli cells remained unaltered. The production of immature and mature Leydig cells decreased by 66% and 27%, respectively. Protein, sialic acid and glycogen content and acid phosphatase activity of testes and epididymides were lowered to varying extents. Testicular cholesterol was elevated significantly. Weights of the testes and epididymides were significantly reduced. The antispermatogenic action of palmitine hydroxide may be mediated by disturbances in Leydig cell function.

Effect of dietary zinc and copper on peripheral blood plasma cholesterol, testosterone and histomorphology of testes in rats.

Total plasma cholesterol (mg/dl) significantly (P less than 0.01) decreased from 70.8 to 54.01 as the dietary Cu levels increased from 2.5 to 5 ppm at 12 pm Zn concentrations in male weanling rats. A similar trend was observed in the blood peripheral testosterone concentration at 12 ppm Zn and 2.5 ppm Cu. Histological examination of testes revealed smaller seminiferous tubules with atrophy of germinal epithelium. Also a marked loss of spermatogenic cells was observed in Zn and Cu deficient rats.

Effect of dietary cholesterol on spermatogenesis.

Cholesterol fed to male rats and rabbits for 120 days impaired testicular function. The production of secondary spermatocytes in rats and rabbits was reduced by 26.1% and 46.9%, respectively. There was significant reduction in spermatid cell population, seminiferous tubules, and Leydig's cell nuclear dimensions. In rats aortic endothelium showed some alterations while in rabbits full-blown atherosclerotic plaque containing a necrotic core and a proliferative fibrous cap was found. Total cholesterol, triglycerides and phospholipids of the testes were increased whereas, testicular glycogen was significantly reduced. Three- and sevenfold increases were noticed in the serum cholesterol levels of rats and rabbits. The LDL cholesterol values were increased while the HDL cholesterol/Total cholesterol ratio was significantly decreased (P less than 0.001). It is concluded that hypercholesterolemia affects the testicular function and aortic endothelium.

Effect of cyproterone acetate on the reproductive system of the female rat. A histological review.

Effects of cyproterone acetate, a synthetic steroidal compound, on the reproductive organs of female rats have been investigated. This agent caused reduction of ovarian weights indicative of suppression of pituitary gonadotrophins. Oestrogenic nature of cyproterone acetate was investigated in intact and ovariectomized rats taking uterine weight and vaginal keratinization as an index of oestrogenicity. Cyproterone acetate in ovariectomized animals induced vaginal keratinization and increased the uterine weights. These effects were parallel to the effect of oestradiol dipropionate in ovariectomized animals, thus indicating oestrogenic activity of cyproterone acetate. We may conclude that the above compound caused antifertility effects due to its oestrogenic nature at the dose level of 2 mg/alternate day in rats when the compound was administered subcutaneously.

Chemical sterilization of males - successful inhibition of spermatogenesis in langurs (Presbytis entellus entellus dufresne) after metopiron (SU-4885, Ciba) administration.

UNLABELLED: 1. Daily administration of metopiron (SU-4885, Ciba) (10 mg/kg body wt. for 30 days) produced a massive atrophy of the spermatogenic elements. The pachytene primary spermatocytes have been implicated as the main site of damage. Seminiferous and Leydig cell shrinkage was conspicuous. Epididymal and vesicula seminalis structure and functions were severely affected. 2. RNA, protein and sialic acid contents of the testes, epididymides and seminal vesicles were reduced, whereas testicular cholesterol and enzyme phosphatase were elevated. 3. Marked reduction in the testicular glycogen was noticed. 4. Normal functioning of liver, kidney and general metabolic activities was revealed by clinical parameters (serum, transaminases, phosphatases, lipids, serum bilirubin, blood sugar and blood urea). Serum protein levels were low, whereas the serum cholesterol was elevated. 5. IN CONCLUSION: Metopiron (SU-4885, Ciba) at 10 mg/kg body wt. dose level did not cause severe damage to the vital organs but it produced an effective inhibition of spermatogenesis in male Presbytis in 30 days and thus induces an antifertility state.

Chemical sterilization of male langurs: synergistic action of alpha-chlorohydrin (U-5897) with methallibure (ICI, 33828) on the testes and epididymides of Presbytis entellus entellus Dufresne.

1. Synergistic action of alpha-chlorohydrin with methallibure (ICI, 33828) on the testicular function of Presbytis entellus entellus Dufresne has been studied. 2. Chronic administration of alpha-chlorohydrin alone (140 mg/day for 40 days) caused testicular lesion resulting in a massive atrophy of the spermatogenic elements. Epididymal epithelium was regressed and the lumen was devoid of spermatozoa. 3. alpha-Chlorohydrin inhibited the synthesis of RNA and sialic acid in the testes and epididymides. Total cholesterol per gram of testis and alkaline phosphatase activity were increased after alpha-chlorohydrin administration. 4. These effects could be achieved with a lower dose of alpha-chlorohydrin (1/4) when administered in combination with a gonadotrophin inhibitor, i. e. ICI, 33828 (Methallibure). Methallibure alone (200 mg/kg: total dose) has no damaging effects on the testes and epididymides. But it altered testicular cholesterol and enzyme activity. 5. In conclusion, an effective inhibition of spermatogenesis could be achieved by synergistic action of the two different drugs i. e. alpha-chlorohydrin and ICI, 33828 (Methallibure).

Alkaline Phosphatase

Effects of whole body CO60 irradiation on the male genital tract of gerbil (Meriones hurrianae), hedgehod (hemiechinus auratus Collaris), and house rat (Rattus rattus Rufescens).

1. The effects of 400 and 575 rads of whole body irradiation in gerbil (Meriones hurrianae), hedgehog (Hemiechinus auratus Collaris) and house rat have been studied with a view to compare the effects of irradiation on the desert mammalian species with those of domestic ones. 2. Typical signs and symptoms of radiation sickness, transient body loss and radiation mortality in gerbils and house-rats were apparent, whereas the hedgehogs were not affected at this dose level. 3. Extensive necrosis, exfoliation of seminiferous element, intertubular edema and shrinkage of the seminiferous tubule and Leydig cell nuclei in gerbils and house-rats were conspicuous in histological preparations. Mild regressive tubular lesions were also observed in hedgehogs treated with 575R. 4. Whole body irradiation depleted the RNA, protein and sialic acid contents of the testes. High cholesterol contens in the testes of irradiated gerbil, hedgehog and house rats identified this as the site of degenerating cells and fragments. 5. Reduced androgen production was confirmed in non active Leydig cells and decreased fructose contents in the seminal vesicles of gerbil and house rats. At 575 R dose level, hedgehog was comparatively radio-resistant. But at 875 R extensive damage to the reproductive organs could be noticed.
