Fertility control in vas occluded rats and the biochemical effects of ascorbic acid feeding.

A new approach to rapid male sterilization has been studied by giving a single injection of 95% ethanol directly into the vas deferens. It produced an effective block in the lumen. The mating exposure test showed that the males were sterile. Vas occlusion caused an atrophy of the Cauda epididymis. Extensive necrosis and exfoliation of the seminiferous elements were conspicuous. Reduced succinate dehydrogenase, acid phosphatase, vesicular fructose and altered ascorbate turnover pattern could be restored to normal by simultaneous ascorbic acid feeding. In conclusion ascorbic acid feeding plays a definite beneficial role in vas occluded rats. The antiandrogenic effects of vas occlusion could be reversed by simultaneous ascorbic acid feeding.

Antispermatogenic/antiandrogenic properties of solasodine (C27H43O2N) obtained from solanum xanthocarpum berries on the male genital tract of dog (Canis-familiaris). A histophysiological approach.

Chronic administration of solasodine (20 mg/kg alt. day for 30 days) caused testicular lesions resulting in a severe impairment of spermatogenic elements. The epididymides were devoid of spermatozoa. Total protein, sialic acid and glycogen contents of the testis and epididymis were reduced significantly whereas the testicular cholesterol was elevated. Acid Phosphatase enzyme activity of the testes was low after solasodine treatment. Serum enzymes (SGPT, alkaline phosphatase) serum protein, triglycerides, non esterified fatty acid levels were in normal range when compared with their own controls. Cholesterol and phospholipid levels were elevated after solasodine treatment to intact dogs. Reduced androgen production was reflected in low levels of sialic acid in the testes and epididymides and reduced Leydig cell nuclei. Castration alone brought about reduction in size of the epididymis. Castration followed by solasodine treatment caused epididymal degeneration. Simultaneous administration of TP to solasodine treated castrated dogs failed to stimulate the epididymal growth. Antispermatogenic/antiandrogenic activity of the compound solasodine is discussed. Solasodine administration in dogs definitely rendered the male infertile as evidenced by the absence of sperms in the cauda epididymis and ductus deferens.

Dose dependent alteration in lipid and carbohydrate metabolites in streptozotocin induced diabetic rats.

1. A relationship is established between different doses of intraperitoneally injected streptozotocin (SZ) and the degree of hyperglycemia and hyperlipemia. 2. Changes in serum level of glucose, cholesterol, triglycerides and NEFA were determined after intraperitoneal administration of three different doses (55 mg, 85 mg and 125 mg/kg body weight) of SZ. Cholesterol level was significantly (P less than 0.001) elevated after 72 hrs only in the animals which received 85 mg/kg and 125 mg/kg SZ. 3. Dose dependent changes in NEFA and triglycerides could be observed after 24 hrs of SZ administration. 4. Diabetic animals consistently showed triphasic blood sugar response, initial hyperglycemia at 5-7 hrs, profound hypoglycemia in between 8-12 hrs and finally an irreversible hyperglycemic state by 24 hrs and onwards. 5. Liver and muscle glycogen were continuously decreased except a significant rise at 12 hrs coinciding with hypoglycemic phase. During the experimental period SZ treated rats continuously lost weight, while control animals progressively gained weight. In summary these changes indicated that diabetogenicity is dose dependent and the severity can be judged by elevated lipid metabolites.

Male contraception properties of a new synthetic steroid derivative (danazol) in Rattus rattus rufescens.

Chronic administration of Danazol (25 mg/kg body wt) caused lesions in the testes of Rattus rattus Rufescens. Depletion of spermatocytes, spermatids and spermatozoa was conspicuous. Impairment of Leydig cell function was correlated with reduced cell size and depressed accessory sex organ weights. Epididymal cell height was greatly reduced. The lumen was devoid of spermatozoa. Danazol administration inhibited the synthesis of RNA, protein, sialic acid in the testes and accessory sex organs. Total cholesterol of the testes was increased, whereas the acid phosphatase enzyme activity was reduced. Testosterone propionate did not enhance the growth of accessory sex organs in castrated rats receiving Danazol. In conclusion, Danazol inhibits the system of steroidogenesis and spermatogenesis in Rattus rattus, when treated chronically for a period of 40 days. These effects are reversible after 60 days of cessation of drug administration.

Hypolipidemic activity of guggal resin (Commiphora mukul) and garlic (Alium sativum linn.) in dogs (Canis familiaris) and monkeys (Presbytis entellus entellus Dufresne).

1. The identification of cholesterol as a constituent in the genesis of coronary artery disease in man and experimental animals are well documented. 2. The hypolipidemic effects of Commiphora mukul (guggulu) and Alium sativum (Garlic powder) were screened in dog and Presbytis monkeys. 3. Progressive decrease in the mean values of cholesterol, triglycerides and phospholipids were conspicuous for forty eight hours following the administration of guggulu/garlic powder. 4. 25 mg/kg body weight garlic powder was more effective in lowering the serum cholesterol and triglycerides as compared with that of guggulu. 5. A comparative hypolipidemic action of the two compounds is discussed.

Effect of cyproterone acetate on the reproductive system of the female rat. A histological review.

Effects of cyproterone acetate, a synthetic steroidal compound, on the reproductive organs of female rats have been investigated. This agent caused reduction of ovarian weights indicative of suppression of pituitary gonadotrophins. Oestrogenic nature of cyproterone acetate was investigated in intact and ovariectomized rats taking uterine weight and vaginal keratinization as an index of oestrogenicity. Cyproterone acetate in ovariectomized animals induced vaginal keratinization and increased the uterine weights. These effects were parallel to the effect of oestradiol dipropionate in ovariectomized animals, thus indicating oestrogenic activity of cyproterone acetate. We may conclude that the above compound caused antifertility effects due to its oestrogenic nature at the dose level of 2 mg/alternate day in rats when the compound was administered subcutaneously.
