[Peroral immunization--a method for enhancing the safety of the recombinant vector (the vaccinia virus)].

Oral and dermal administrations of vaccinia virus into rabbits, guinea pigs, and monkeys demonstrated a milder (without homeostasis disturbance) course of the vaccinal process with oral immunization, intensive immunity forming in minimal sensitization of the body was compared with dermal one. The results of revaccination in adults with oral smallpox vaccine and primary immunization in Ethiopia showed that oral immunization with vaccinia virus was safe, effective and lowly reactogenic. The comparative study of the preparation in remote revaccination (5 or more years later) proved its advantage over dermal vaccine. It consists in ecological safety (vaccinia virus excretion into the environment during 1 and 11 days, respectively, by 10-fold concentration reduction), reactogenicity (5 and 33% of common reactions, respectively) in the same immunogenicity.

[A method of fluorescent immunoenzyme analysis in the complex evaluation of viral and rickettsial vaccines].

With the use of the unified indirect solid-phase fluorescent enzyme immunoassay the combined evaluation of the antigenic and immunogenic properties of experimental whole-virion inactivated virus vaccines against Venezuelan and eastern equine encephalomyelitides, as well as of dried chemical typhus vaccine, has been made; their safety was determined indirectly by the content of ovalbumin. The protective role of antibodies evaluated by the solid-phase fluorescent enzyme immunoassay in typhus immunity has been shown.

[The evaluation in hamadryas baboons of the possibility for the specific prevention of Ebola fever].

The protective role of virus-neutralizing antibody was demonstrated in Ebola virus-infected animals (Papio hamadryas) used in experiments on the development of passive humoral immunity by using specific immunoglobulin. Two immunizations of the monkeys with purified concentrated Ebola virus antigen with complete Freund adjuvant was shown to confirm intensive immunity to subsequent challenge protecting 80% of the immunized animals. It is concluded that effective preparations may be developed for specific prophylaxis of Ebola fever.

[A comparative evaluation of an immunoenzyme analytical method using a fluorogenic or chromogenic substrate in determining the Marburg virus antigen].

A comparative evaluation of the developed variants of indirect solid-phase enzyme immunoassay using fluorogenic and chromogenic substrates for the determination of Marburg virus antigen was carried out. The resolving capacity of this method was 3.8 x 10(-9) g of protein for the former and 3.1 x 10(-8) g of protein for the latter substrate. Cross titrations demonstrated the lack of common antigens with Ebola virus.

[A trial to produce an inactivated Lassa fever vaccine].

A single and double immunization of Papio hamadryas monkeys with an inactivated concentrated purified adsorbed preparation of Lassa virus in doses of 0.5-1.0 and 1.1-2.1 mg by protein, respectively, protected the animals against the development of the disease and occurrence of viremia after an intramuscular challenge with 0.4 PFU dose. Upon aerogenic infection of monkeys protection of 50% of the animals was achieved by a single immunization with the inactivated preparation in a dose 1.1 mg by protein.

[Development and study of the properties of immunoglobulin against Ebola fever].

Immunoglobulin to Ebola fever has been for the first time prepared from hyperimmune equine blood sera by alcohol fractionation after Cohn. Preclinical study of the physicochemical and immunobiological properties of immunoglobulin showed that it protects up to 100% Papio hamadryas infected intramuscularly at doses of 110 to 29 LD50 Ebola virus. Scheme for the use of Ebola immunoglobulin has been experimentally validated.

[An experimental study of the immunogenic and protective properties of Coxiella burnetii strain M-44 and a live enteral vaccine against Q fever].

High effectiveness of C.burnetii strain M-44 was confirmed in experimental studies on subcutaneously immunized guinea pigs ¿correction of pig] after their intratracheal infection with the virulent culture of the infective agent. The study revealed that the dose necessary for the enteral immunization of guinea pigs and monkeys was 10(4.8) -- 10(5.0) times less than that needed for the oral immunization of the animals. Live enteral vaccine obtained on the basis of this strain possessed sufficiently pronounced immunogenic and protective properties, protecting 75% of immunized monkeys from subcutaneous infection with C.burnetii virulent culture.

[Development of the immunoenzyme test-system for detection of Ebola virus antigen].

An enzyme immunoassay system has been developed for the detection of Ebola virus antigen. It permits a highly accurate and sensitive rapid detection of the antigen. Optimal dilutions of specific immunoglobulin (1:500, corresponding to protein concentration of 50 micrograms/ml) and conjugate were found. The resolving capacity of the new test system is 1.9 x 10(-7) g protein.

[Developing principles for emergency prevention and treatment of Ebola fever].

The authors validate the efficiency of pathogenetic approach to the development of urgent measures for the prevention and therapy of Ebola fever. The virus circulating in the body is to be blocked as soon as possible and the impaired functions and systems repaired. Therapy of Ebola fever should be based on the earliest possible and sufficiently prolonged administration of specific immunoglobulins in combination with pathogenetic drugs.

[Development and study of properties of immunoglobulins against Lassa fever].

A horse may serve the producer of immune antiserum to Lassa virus. Specific immunoglobulin with at least 1:512 titer of virus-neutralizing antibodies to Lassa fever was obtained by alcohol sedimentation after Cohn from the blood serum of immunized horses. The preparation does not differ from heterologous commercial immunoglobulins. Preclinical studies of immunoglobulin to Lassa fever demonstrated its safety and a high specific activity. The agent can be injected both alone and in combination with virasole.
