Malonate was tested for its effects on rat hepatic metabolism of [1,4-14C]-succinic acid and [1-14C]-glycerol. The administration of malonic acid did not limit [1,4-14C]-succinate metabolism and change the rate of radioactive labelling from [1-14C]-glycerol to the total lipid fraction, as well as the fraction of phospholipids and triacylglycerides. However, there was a reduction in the use of glycerol as a precursor of higher fatty acids and cholesterol. It is suggested that the detected effect of malonate is due to its action on oxidative decarboxylation of pyruvic acid.
The studies have demonstrated that the inhibitory effect of urea, peroxidase, catalase, its combination with superoxide dismutase on HADPH-dependent lipid peroxidation in the old rat heart is higher than that in young animals. Exposure of young rats to stress is followed by enhanced antioxidative effects of all the parameters under study. This effect is not found in the old animals exposed to stress.
The investigations conducted have shown that in the myocardium of aged rats the intensity of lipid peroxidation prevails over its value in young animals. This leads to formation of the age-related peculiarities of the lipid peroxidation response to the action of stressor factors and to the change of the antioxidants' effect on induced lipid peroxidation reactions in the heart.
Malonic acid is one of the intermediates of cell metabolism. Malonic dialdehyde and malonyl coenzyme A may be important precursors of malonate. The correlation of biosynthesis intensity and utilization of malonate determines capacity of its intracellular pool. It is supposed that pool capacity is related both to metabolism regulation and to functional condition of organs and tissues.
Effect of malonic acid on the activity of pyruvate dehydrogenase was studied in rat brain hemispheres. Malonate was shown to regulate the oxidative decarboxylation of pyruvic acid. The rate of the reaction depended on the concentration of malonic acid as well as on the content of pyruvate dehydrogenase substrates in the experimental samples.
Clinico-experimental studies were made of the level of cortizol and corticosterone in the tissue fluid of burn vesicles and the blood plasma flowing from the burned and intact body areas; the character and the extent of entrance into the tissues and utilization by them of these hormones was assessed. Along with the increase in the blood cortizol and corticosterone concentrations, and, particularly, in the exudate of the burn vesicles the burned humans and rabbits displayed a different dynamics of the changes in the hormone level in the biological media under study. In the patients and the experimental animals cortizol content in the burn exudate was several times greater than corticosterone concentration. Cortizol level in the edema fluid directly correlated with the degree of edema. It appeared that the burned tissues had a different capacity to the uptake and utilization of cortizol and corticosterone. Features of similarity and differences were noted in the dynamics of the hormonal shifts in the sick and experimental animals. In difference from the intact animals, in the burned rabbits the corticosterone type of the adrenal secretion changed to preponderantly cortizol one.
The present investigation reveals increase of diene conjugate accumulation, decrease of the level of TBA-positively reacting substances and the constant concentrations of Schiff's bases in the incubation medium during the stimulation of NADPH-dependent lipid peroxidation in the heart homogenates of stressed rats as compared with native animals. The intensity of diene conjugate accumulation and the decrease of thiobarbiturate-reacting substances were less expressed in the aged rats than in young ones. It is supposed that stress in the aged rats in accompanied by the decreased effectiveness of initiation processes of free radical reactions and the emergence of certain specific features of conversion of myocardial lipid peroxidation intermediates during stress.
The complementary system is activated in response to development of the reperfusion syndrome in patients subjected to bilateral aortofemoral shunting for Leriche's syndrome. These changes result in the development of respiratory disorders presenting as constriction of the small and medium-sized bronchi, increase of pulmonary shunting of the blood, and manifest hypoxia.
