[Study of the membranes of pigment-free mutant of Staphylococcus aureus].

The membranes from pigmented strain of Staphyloccus aureus 209-P and its apigment mutant B-6 were isolated. The infrared spectra, the cytochrome content, and the activities of the dehydrogenases and oxidases of the membranes were studied. The apigment mutant membranes differed from those of the pigmented strain in all parameters studied. It is concluded that the carotenoid pigments affect the chemical composition and functional activity of the Staphylococcus membranes.

[Teichoic acids as possible acridine orange acceptors on the surface of staphylococcal cell walls].

A study was made of acridine orange adsorption by the surface structures of intact Staphylococcus aureus (209-P strain) cells and their cellular walls. Acridine orange sorption by the cells and the cellular wall of staphylococcus depended on the content of teichoic acids in the object under study. A conclusion was drawn that teichoic acids of the cellular walls of Staph. aureus were responsible for the binding of the acridine orange stain.

[The role of the carotenoid pigments of staphylococcus in the response of cells to temperature changes].

A study was made of the reaction of the membranous apparatus of the pigmented strains of staphylococcus 209-P and its four apigmented variants to the action of various temperatures measured by the exit from the cells of the low molecular components. Permeability of the cell membranes in case of the action of the extreme temperatures of the 209-P strain altered much more than that of the apigment variants. It is supposed that the carotinoid pigments of the apathogenic staphylococci took part in the formation of functional lability of the bacterial membranes.

[Acetyl-CoA-synthetase activity of pigmented staphylococci].

In 19 strains of staphylococci a study was made of the activity of acetyl-CoA-synthetase reaction. All the strains possessed an active enzymatic system transforming the acetate into an active form. The activity of acetyl-CoA-synthetase proved to be much greater in the pigmented staphyloccus strains than in the nonpigmented ones. It is supposed that there existed an association between the acetyl-CoA-synthetase and the biogenesis of carotinoid pigments in Staph. aureus.

[Study of acetyl-CoA-synthetase from staphylococcus aureus].

Acetyl-CoA-synthetase was isolated from cells of St. aureus 209-P. The method of isolation and partial purification of the enzyme is worked out. Km values of the enzyme for acetate, CoA and ATP are calculated. p-Chloromercuribenzoate and monoiodoacetate were shown to inhibit the enzyme activity. The enzyme activity is estimated depending on the age of the cell culture and on the presence of acetate in the culture medium.

[Respiration and dehydrogenase activity of pigmentless variants of Staphylococcus aureus].

Respiration and dehydrogenase activity were studied in four pigmentless mutants of Staphylococcus aureus 209-P. The rate of oxygen uptake from the incubation medium was higher in the mutants than in the parent strain. The rate of respiration and the activity of dehydrogenase in the pigmentless mutants were found to depend on the permeability of the cell membrane for substrates of respiration; carotenoids are presumed to be involved in this permeability.

[Interaction of acridine orange with teichoic acids of Staphylococcus aureus].

Teichoic acids of the cell wall of Staphylococcus aureus 209-P belong to the class of ribitol teichoic acids and can bind actively the molecules or acridine orange, changing its optical properties. The intravital fluorochromy of the cells with acridine orange may be caused by the sorption of its molecules by the teichoic acids of the cell wall.

[Fluorescent probe study of staphylococcal cell wall charges].

The effect of pH and MgCl2 concentration on the fluorescence of 1-anilinonaphthalene-8-sulfonate (ANS), an anion probe, was studied in suspensions of Staphylococcus aureus. The maximum of ANS fluorescence shifted towards the short wavelength region of the spectrum and its intensity increased when the cells were incubated in the medium with low pH and high ionic strength. The results are discussed in terms of electrostatic interactions between the anions of the probe and the negative space charge of the cell wall.

[Cell wall of pigmentless variants of Staphylococcus aureus].

Physico-chemical properties and chemical composition of the cell wall of pigmentless variants B-3 and B-6 differ from those of the parent strain of Staphylococcus aureus 209-P. As to morphology, the wall is more thick and loose. The mutants differ also by the phagotype, the ability for sorption of acridine orange, and the rate of sedimentation of cell suspensions. The properties of teichoic acids and the content of peptides in peptidoglycan change in the cell wall of the mutants.
