Histoelectrofocussing is a combination of the usual histochemical methods of wound age estimation with biochemical procedure. Histological sections from skin wounds were directly subjected to isoelectric focussing. The enzymes being extracted and separated were detected by means of histochemical approach. The alterations of the activity of different enzyme fractions can be calculated quantitatively by densitometry. It is possible to differentiate between enzymes of the common skin and those with a topographic connection to the wound edge.

This paper presents the reconstruction of an unusual case of suicide. After raiding a branch-bank a robber fled shooting with his Sauer-Western revolver caliber .44 magnum at the pursuing policemen and succeeded in wrestling a pistol Walther caliber 7,65 mm from them. Under the fire of sub-machine guns he destroyed himself by a shot to the neck. Our investigations concerned a textile damage at the front of the sweater of the deceased surrounded by primer residue, showing characteristics of a close-up shot. The damage was identified as effect of explosion gases exhausting far-off the muzzle. The distance between this injury and the bullet hole corresponded with the length of the barrel of the Sauer-Western revolver and could be used for identification; it confirmed the diagnosis of a close-up shot at the neck, too. Collateral experiments with shots from distant ranges developed spadiceous melt figures of textile fibers around the bullet hole, the appearance of which is considered proof for a close-up shot commonly.

To elucidate the post-mortem proteolysis we investigated the behaviour of the free amino acids in rotting liver homogenates by means of two-dimensional thin-layer chromatography and reflection densitometry. The post-mortem alterations of the amino acid levels are characterized mostly by a two-phase increase with maxima at the end of the first and after the third week. A few amino acids showed only little changes hardly above the physiological intra-cellular levels. Anaerobic rat experiments and experiences with antibiotic treatment give rise to the supposition that increasing amino acid concentrations indicate a predominance of proteolytic activities over aminoacid catabolism. In the beginning autoenzymes are prevalent, while afterwards bacterial proteases probably predominate. The transient regression about the second and thrid week is interpreted as the effect of a temporarily intensified amino acid degradation by foreign enzymes (maximal bacterial growth).
The identification of the implement used is an important factor in the explantion of the causes of violent deaths. Hitherto existing knowledge has been almost exclusively confined to the damages to skull bones. This paper draws attention to tool marks in the area of pierced rib cartilage and considers the possibilities of their analysis. The evaluation of saw marks is further discussed on the basis of sawing experiments. Also, there are features and peculiarities which can help to solve the problems posed by criminal dismemberment.

Samples of thoracic fluid were obtained at regular intervals from the putrefying bodies of dead dogs. For quantitative estimation of the proteins two-dimensional immunoelectrophoresis was employed and for the amino acids and amines two-dimensional thin-layer chromatography. The results lead to speculation that the initially lagging proteolytic changes are due to catalysis of autoenzymes, whereas bacteriological processes are responsible for a distinct acceleration from the 20th day after death. At this time the amino-acid concentrations showed a marked increase, which is followed by a maximum of bacterial growth, that ultimately leads to a sudden regression of amino acids. Simultaneously amines appear. The drying up of the thoracic cavity after 35 to 45 days marks the change from liquefaction to the terminal decay.
A method for the identification and quantitation of hemoglobin derivatives is described. Samples diluted 1 in 100 were applied to precoated polyacrylamide-gel plates containing an Ampholine gradient of pH 3.5 - 9.5 and focused for 1.5 h at a constant 40 W. All hemoglobins under investigation wee characterized by specific patterns which allow them to be differentiated. Electrofocusing is recommended for screening tests, but quantitative evaluation using densitometry is possible as well.
Estimation of the age of skeletal remains is usually based on subjective evaluation of the morphological grade of decomposition. This assumes, that the investigator is provided with pertinent experience. Experimentally elaborated physical and chemical methods proposed for objectification of the statements did not yet assest until today due to lacking temporal correlation. Our investigations were stimulated by observations on fossil bones indicating that the collagenbound amino acids diminish very slowly dependent on geological age. Samples of 53 skeletons of five to 2500 years were pulverized and subjected to acid hydrolysis. The amino acids released from bone collagen were separated by means of thin-layer chromatography and detected by isatin and ninhydrin reagents. The dye complexes of alanine, hydroxyproline, proline, tyrosine, and valine were measured densitometrically and quantified through calibration curves. We established, that the concentrations of the amino acids exhibited a time dependent reduction as equivalent of a loss collagen. The temporal correlation is very narrow (r less than -- 0,8) and sufficient for datings within plus/minus ten years of the more recent bones. Environmental conditions during post-mortem decay seem to have little influence on this process.
UNLABELLED: No reliable data are available on cases of lethal child abuse (by active force) in the area of Federal Republic of Germany prior to reunification (the former West Germany). In a multicenter study we therefore examined the police and court records for such cases occurring in the period 1 January 1985 to 2 October 1990 in nearly the entire area of Federal Republic of Germany. RESULTS: The study center received information on 58 cases of lethal child abuse. Extrapolated to all institutes of legal medicine, this corresponds to 62 cases in all of West Germany in the period studied. An approximately equal number of unreported cases should be added to this figure. Including unreported cases, at least 20 cases of lethal child abuse occurred per year; thus only one in every two cases ever came to light. Almost two thirds of the victims were younger than one year old. At autopsy 59% exhibited signs of repeated abuse at autopsy. By far the most common cause of death was direct impact from a blunt object, usually to the head. Mostly, the male person to whom the victim relates most closely (father, stepfather, partner of the mother) has killed the child. Twenty-one of the 74 persons charged saw the charges against them dropped or were acquitted due to lack of evidence; 51 received sentences ranging from one year probation to life. In the remaining two cases the outcome of the trial was unknown. Signs of abuse were readily apparent at autopsy in almost all cases. The high number of unreported cases underscores the need to educate medical students and practicing physicians to be on the look-out for signs of abuse and argues for an increase in the rate of autopsy.
