Adaptive health behaviors among ethnic minorities.

Race, ethnicity, and cultural attitudes and practices are among the variables that influence health behaviors, including adaptive health behaviors. The following discussions highlight the important role of social conditions in shaping health behaviors and the central role of family in promoting health across the Asian, Hispanic, Native American, and African American ethnic groups. Factors that may lead to health-damaging behaviors are also discussed. The need for additional research that identifies correlations among physiological, social, and behavioral factors and health behaviors, as well as underlying mechanisms, is called for.

In human fetal astrocytes exposure to interleukin-1 beta stimulates acquisition of the GD3+ phenotype and inhibits cell division.

In human astrocyte cultures established from second-trimester fetal brain tissue, approximately 5-10% of total astrocyte population in unstimulated cultures were GD3+/glial fibrillary acidic protein (GFAP)+. The GD3+ cells were always GFAP+ and grew as flat, highly spread cells but changed to process-bearing cells after interleukin-1 beta (IL-1 beta) stimulation. It is interesting that IL-1 beta, a known mitogen for rat astrocytes, suppressed human fetal astrocyte proliferation as determined by [3H]thymidine incorporation, bromodeoxyuridine (BrdU) labeling, and cell counting. The GD3+ population, however, consistently increased in absolute number after IL-1 beta stimulation, in a dose- and time-dependent manner. The IL-1 beta-mediated increase in number of GD3+ astrocytes was independent of initial cell density or serum concentration. By flow cytometry, IL-1 beta enhanced both the mean fluorescence intensity and the percentage of GD3+ cells. To investigate whether the increase in GD3+ astrocyte cell number was due to proliferation of preexisting GD3+ astrocytes or due to conversion of GD3- to GD3+ cells, we performed BrdU/GD3 double immunocytochemistry. BrdU/GD3 double-positive cells were extremely rare in both control and IL-1 beta-stimulated cultures. Moreover, an increase in number of GD3+ astrocytes was still observed in control and IL-1 beta-stimulated cultures were GD3+ cells had been initially eliminated by cell sorting. These results indicate that GD3+ astrocytes in human fetal culture may represent a postmitotic, differentiated, distinct phenotype.

Epidermal growth factor precursor is present in a variety of human renal cyst fluids.

Progressive human renal cystic diseases are characterized by proliferation of the epithelial cells lining the cyst. The kidney synthesizes epidermal growth factor and its presence in renal cyst fluid might contribute to renal epithelial cell proliferation. We screened autosomal dominant polycystic kidney disease, acquired renal cystic disease, the von Hippel-Lindau syndrome, multilocular cystic nephroma, multicystic dysplastic kidney disease and simple cyst fluids for the presence of epidermal growth factor by radioreceptor assay, specific radioimmunoassay, immunoprecipitation and immunoblotting. Multiple epidermal growth factor immunoreactive species of approximately 180 kD. and lower molecular weights were present in almost all cyst fluids examined, suggesting endogenous synthesis and limited proteolysis of epidermal growth factor precursor protein in cyst fluid. Tamm-Horsfall protein was detected by enzyme-linked immunosorbent assay in most cysts (for example 55 of 59 autosomal dominant polycystic kidney disease samples). The majority of simple and autosomal dominant polycystic kidney disease renal cysts contained high sodium ion concentration, epidermal growth factor precursor protein and Tamm-Horsfall protein, characteristic of the early thick ascending limb. Rather than the mere presence of epidermal growth factor in renal cyst fluids, increased sensitivity to epidermal growth factor or other mitogens present in renal cyst fluid may be pathogenic in progressive renal cystic disease.

Cytokinin action is coupled to ethylene in its effects on the inhibition of root and hypocotyl elongation in Arabidopsis thaliana seedlings.

Cytokinins have profound effects on seedling development in Arabidopsis thaliana. Benzyladenine (BA) inhibits root elongation in light- or dark-grown seedlings, and in dark-grown seedlings BA inhibits hypocotyl elongation and exaggerates the curvature of apical hooks. The latter are characteristic ethylene responses and, therefore, the possible involvement of ethylene in BA responses was examined in seedlings. It was found that the inhibitory effects of BA on root and hypocotyl elongation were partially blocked by the action of ethylene inhibitors or ethylene-resistant mutations (ein1-1 and ein2-1). Ethylene production was stimulated by submicromolar concentrations of BA and could account, in part, for the inhibition of root and hypocotyl elongation. It was demonstrated further that BA did not affect the sensitivity of seedlings to ethylene. Thus, the effect of cytokinin on root and hypocotyl elongation in Arabidopsis appears to be mediated largely by the production of ethylene. The coupling between cytokinin and ethylene responses is further supported by the discovery that the cytokinin-resistant mutant ckr1 is resistant to ethylene and is allelic to the ethylene-resistant mutant ein2.

Pharmacokinetics of ofloxacin in serum and vitreous humor of albino and pigmented rabbits.

The purpose of this study was to evaluate the pharmacokinetics of ofloxacin in serum and vitreous humor samples from albino and pigmented rabbits by using a recently described animal model which permits robust estimation of parameter values. The drug was administered to rabbits intravenously, multiple vitreous humor and serum samples were taken from each rabbit, and the vitreous humor and serum samples were assayed by high-pressure liquid chromatography. The pharmacokinetic parameters were determined with RSTRIP, an iterative, nonlinear, weighted, least-squares regression program. Eight New Zealand White rabbits and eight Dutch Belted rabbits (split into single-dose and multiple-dose groups) were investigated in this study. The value of penetration into the vitreous humor of albino rabbits (n = 6) was 32.6% +/- 2.12%, with terminal-elimination half-life values of 3.21 and 2.39 h, respectively, for vitreous humor and serum. In pigmented rabbits after a single dose (n = 3) and with a steady-state concentration of drug in serum (n = 4), penetration values were similar, at 30.4% +/- 2.98% and 30.0% +/- 4.12%, respectively (P > 0.10). Following a single dose of ofloxacin, pigmented animals had elimination half-life values from serum and vitreous humor of 2.64 and 4.32 h, respectively. After steady state was achieved, half-life values for serum and vitreous humor were 3.12 and 6.05 h, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Determination of robust ocular pharmacokinetic parameters in serum and vitreous humor of albino rabbits following systemic administration of ciprofloxacin from sparse data sets by using IT2S, a population pharmacokinetic modeling program.

Robust determination of the concentration-time profile of anti-infective agents in certain specialized compartments is often limited by the inability to obtain more than a single sample from such a site in any one subject. Vitreous humor and cerebrospinal fluid are obvious examples for which the determination of concentrations of anti-infective agents is limited. Advances in pharmacodynamics have pointed out the importance of understanding the profiles of drugs in the plasma and in specialized compartments in order to dose the drugs to obtain the best patient outcomes. Advances in population pharmacokinetic modeling hold the promise of allowing proper estimation of drug penetration into the vitreous (or other specialized compartment) with only a single vitreous sample, in conjunction with plasma sampling. We have developed a rabbit model which allows multiple samples of vitreous to be obtained without breaking down the blood-vitreous barrier. We have employed this model to test the hypothesis that robust estimates of vitreous penetration by the fluoroquinolone ciprofloxacin can be obtained from a traditional intensive plasma sampling set plus a single vitreous sample. We studied 33 rabbits which were receiving 40 mg of ciprofloxacin per kg of body weight intravenously as short infusions and from which multiple plasma and vitreous samples were obtained and assayed for ciprofloxacin content by high-performance liquid chromatography. Data were analyzed by the iterative two-stage population modeling technique (IT2S), employing the iterative two-stage program of Forrest et al. (Antimicrob. Agents Chemother. 37:1065-1072, 1993). Two data sets were analyzed: all plasma and vitreous samples versus all plasma samples and the initially obtained single vitreous sample. The pharmacokinetic parameter values identified were used to calculate the percent vitreous penetration as the ratio of the area under the concentration-time curve for the vitreous to that for the plasma. The values identified, 4% penetration for the full data set versus 3% penetration for the single vitreous sample data set, and their corresponding estimates were not statistically significantly different. We conclude that population modeling holds promise for the analysis of penetration of antimicrobiol agents into specialized spaces from which only single samples can be obtained, particularly for patients with whom robust plasma sampling can be performed.

Hormone-independent repression of AP-1-inducible collagenase promoter activity by glucocorticoid receptors.

The role of the ligand in glucocorticoid receptor-mediated transactivation and transrepression of gene expression was investigated. Half-maximal transactivation of a mouse mammary tumor virus-chloramphenicol acetyltransferase reporter gene in transfected cells expressing the human glucocorticoid receptor mutant GRL753F, from which the rate of ligand dissociation is four to five times higher than the rate of dissociation from normal receptors, required a 200- to 300-fold-higher concentration of dexamethasone than was required in cells expressing the normal receptor. Immunocytochemical analysis demonstrated that this difference was not the result of a failure of the mutant receptor to accumulate in the nucleus after steroid treatment. In contrast, in cells cotransfected with a reporter gene containing the AP-1-inducible collagenase gene promoter, the concentration of dexamethasone required for 50% transrepression was the same for mutant and normal receptors. Efficient receptor-mediated transrepression was also observed with the double mutant GRL753F/C421Y, in which the first cysteine residue of the proximal zinc finger has been replaced by tyrosine, indicating that neither retention of the ligand nor direct binding of the receptor to DNA is required. RU38486 behaved as a full agonist with respect to transrepression. In addition, receptor-dependent transrepression, but not transactivation, was observed in transfected cells after heat shock in the absence of the ligand. Taken together, these results suggest that unlike transactivation, transrepression of AP-1 activity by the nuclear glucocorticoid receptor is ligand independent.

Familial ligand-defective apolipoprotein B. Identification of a new mutation that decreases LDL receptor binding affinity.

Detection of new ligand-defective mutations of apolipoprotein B (apoB) will enable identification of sequences involved in binding to the LDL receptor. Genomic DNA from patients attending a lipid clinic was screened by single-strand conformation polymorphism analysis for novel mutations in the putative LDL receptor-binding domain of apoB-100. A 46-yr-old woman of Celtic and Native American ancestry with primary hypercholesterolemia (total cholesterol [TC] 343 mg/dl; LDL cholesterol [LDL-C] 241 mg/dl) and pronounced peripheral vascular disease was found to be heterozygous for a novel Arg3531-->Cys mutation, caused by a C-->T transition at nucleotide 10800. One unrelated 59-yr-old man of Italian ancestry was found with the same mutation after screening 1,560 individuals. He had coronary heart disease, a TC of 310 mg/dl, and an LDL-C of 212 mg/dl. A total of eight individuals were found with the defect in the families of the two patients. They had an age- and sex-adjusted TC of 240 +/- 14 mg/dl and LDL-C of 169 +/- 10 mg/dl. This compares with eight unaffected family members with age- and sex-adjusted TC of 185 +/- 12 mg/dl and LDL-C of 124 +/- 12 mg/dl. In a dual-label fibroblast binding assay, LDL from the eight subjects with the mutation had an affinity for the LDL receptor that was 63% that of control LDL. LDL from eight unaffected family members had an affinity of 91%. By way of comparison, LDL from six patients heterozygous for the Arg3500-->Gln mutation had an affinity of 36%. The percentage mass ratio of the defective Cys3531 LDL to normal LDL was 59:41, as determined using the mAb MB19 and dynamic laser light scattering. Thus, the defective LDL had accumulated in the plasma of these patients. Using this mass ratio, it was calculated that the defective Cys3531 LDL particles bound with 27% of normal affinity. Deduced haplotypes using 10 apoB gene markers showed the Arg3531-->Cys alleles to be different in the two kindreds and indicates that the mutations arose independently. The Arg3531-->Cys mutation is the second reported cause of familial ligand-defective apoB.

Estimation of variance distribution in three-dimensional reconstruction. I. Theory.

A theory is developed for estimating the three-dimensional (3-D) variance of a 3-D image reconstructed from projections by weighted backprojection. The theory is applicable for any data-collection schemes that produce partially redundant sampling of the angular space. The particular data collection considered here, the single-exposure random-conical scheme, is used for the reconstruction of macromolecules in electron microscopy. In this context, the purpose of the 3-D variance estimation is to detect and localize the conformational variability, to assess the significance of structural differences between two experimentally related 3-D images, and to assess the significance of local features in a 3-D image. The 3-D variance estimate of each reconstruction voxel is obtained by (i) the comparison of closest points on Fourier sections associated with difference projections, (ii) the comparison of neighbor projections in real space, or (iii) the comparison of projections with reprojections of the reconstruction.

Estimation of variance distribution in three-dimensional reconstruction. II. Applications.

A previously developed theory of three-dimensional (3-D) variance estimation [J. Opt. Soc. Am. A 12, 2615-2627 (1995)] is applied to the structural study of a hemocyanin-Fab complex with the electron microscope. The precise locations of structurally variable regions of the macromolecule are determined from the 3-D variance maps. The structural differences among different classes of the macromolecular complex are assessed by the use of the statistical t-test, and the 3-D antibody binding sites are revealed. From a model analysis, a rule is demonstrated for visually identifying a 3-D conformational change by the inspection of the 3-D variance map. Our analysis lays the foundation for numerous practical applications of variance estimation in the 3-D imaging of macromolecules.

Topological requirements for recognition and cleavage of DNA by ribosome-inactivating proteins.

Ribosome-inactivating proteins (RIPs) were demonstrated to exhibit a unique enzymatic activity on cleaving supercoiled double-stranded DNA into the nicked or linear form. Although there is an interaction between supercoiled DNA and RIP, no sequence-specific recognition was involved. Instead, RIPs recognize supercoiled DNA by conformational specificity. Negatively supercoiled DNA is the preferential conformation in the action of RIPs. When double-stranded DNA occurs in the supercoiled form, even if with lower linking number, RIPs can still convert it into nicked or linear form. Terminal-labelling experiments indicated that radioactivity was incorporated into putative 5'-ends of nicked or linear DNA generated by RIPs. We conclude that RIPs act as a novel supercoil-dependent endonuclease when they cleavage supercoiled DNA. The impossibility that contaminating enzymes in the RIP preparations cleaved the supercoiled DNA is briefly discussed.

A group sequential procedure for all-pairwise comparisons of k treatments based on the range statistic.

In this paper, a group sequential procedure for all-private comparisons of the means of k independent normal populations with a common known variance is proposed. A repeated range test is defined and its critical points are tabulated. The power function is studied and minimum group size needed to achieve a desirable power level is provided. The test can also be converted into a set of repeated simultaneous confidence intervals from which all-pairwise comparisons can be made. An example of the application of this new interference method is given.

[Influence of the 2nd and 3rd grade abstracts from scorpion venom of Buthus martensii Karsch on action potentials of fast response myocardiocytes].

AIM: To screen the active components of sodium channel blockade action from Buthus martensii Karsch (BMK) scorpion venom. METHODS: Myocardiocytes of mice were cultured. Action potentials of fast response myocardiocytes were recorded. The effects of 22 contents of BMK scorpion venom in a concentration of 3 mg.L-1 were tested and compared with that of TTX 2.5 mg.L-1, nimodipine (Nim) 3 mg.L-1 and BaCl2 24.4 mg.L-1. RESULTS: BMK-1 and other 19 contents significantly decreased depolarization parameters Vmax, APA, OS, and MDP, which was similar to that of TTX and different from that of Nim and BaCl2. CONCLUSION: The variations in the action potentials of fast response myocardiolcytes indicated that 20 contents of BMK scorpion venom had sodium channel blockade action.
