Suppression of tumorigenesis by transcription units expressing the antisense E6 and E7 messenger RNA (mRNA) for the transforming proteins of the human papilloma virus and the sense mRNA for the retinoblastoma gene in cervical carcinoma cells.

Human cervical carcinoma cell lines that harbor human papilloma virus (HPV) have been reported to express HPV E6 and E7 proteins at least in the beginning stages if not at all stages of the disease. The HPV E6 and E7 proteins bind to and inactivate the products of the p53 and retinoblastoma (Rb) tumor suppressor genes, which thereby allow the cervical carcinoma cells to circumvent the action of these tumor suppressor genes. We observed that the introduction of the antisense HPV 18 E6 and E7 sequences, as well as a sense cDNA for the human wild-type Rb gene into a human cervical carcinoma cell line (HeLa), which is positive for the HPV 18 provirus, decreased the in vitro and in vivo growth rate of the transfected cells if both antisense transcripts for the HPV 18 E6 and E7 and sense transcripts for human Rb were expressed. In addition, overexpression of a complementary DNA (cDNA) for the Rb messenger RNA was sufficient to slow the proliferation of HeLa cells, and the level of Rb cDNA expression was correlated with the degree to which the rate of growth of the tumor was slowed. The results of our experiments show that the presence of HPV E6 and E7 proteins and the resultant inactivation of Rb in cervical carcinoma cells contributes to the neoplastic phenotype even in highly evolved cervical carcinoma cell lines such as HeLa, which have been derived from a cervical carcinoma patient at an advanced stage of the disease process. These data suggest that the HPV proteins play a role not only at the beginning of cervical cancer, but also at advanced stages of this disease. These experiments may lead to genetic approaches to the control of this disease that involve antisense sequences that downregulate the E6 and E7 genes or lead to expression of the Rb gene.

[Promoting effects of rhizoma Drynariae on the calcification of cultivated chick embryo bone primordium].

It has been discovered by tissue cultivation and isotope tracing that the injection of Rhizoma Drynariae significantly promotes the calcification of chick embryo bone primordium in vitro, increases the ALP activity of cultivated tissues and accelerates the synthesis of proteoglycan. It has also been proved that the promotion of the synthesis of proteoglycan is an important factor of the promotion of calcification.

[Examination on extraction technology for radix Sophorae Flavescentis by means of orthogonal experiment design].

The extraction and purificatory crafts for Radix Sophorae Flavescentis were observed through the orthogonal experiment design using matrine as the fixed indicatrix for TLC-scanning. Quantitative analyses of matrine were made on the momentum parameters of the different craft conditions. The results of the three different extraction methods show that the best way is to use 2% gelatin to remove tannin with the least loss of matrine, and that the ethanol extract from water precitation is better than the water extract from ethanol precipitation.

Comparison of four methods to generate immunoreactive fragments of a murine monoclonal antibody OC859 against human ovarian epithelial cancer antigen.

In the present study, four different proteases (pepsin, papain, bromelain and ficin) were screened with a murine monoclonal antibody OC859, in order to verify whether different digestion procedures could improve yield and stability of the F(ab')2 or Fab fragments. The yields of F(ab')2 or Fab fragments from digestion with pepsin, papain, bromelain and ficin were respectively 20.3 +/- 2.0%, 50.5 +/- 5.0%, 74.4 +/- 2.7% and 82.8 +/- 10.2% of the theoretical maximum. Immunoreactivity in a noncompetitive solid-phase radioimmunoassay (SPRIA) of the fragments generated by the four proteases were respectively 10 +/- 5%, 36 +/- 5%, 60 +/- 6% and 75 +/- 6% of the intact OC859 IgG. These results suggested that the fragmentation of OC859 with ficin gave a higher yield of superior immunoreactive fragments.

[The role of sympathetic mechanism in the effect of electroacupuncture on immunoregulation].

1. In normal control mice (only injected SRBC) after EA the lymphocyte transformation test (LTT, P < 0.05) and interlukin-2 (IL-2) activity (P < 0.05) were significantly increased, but the serum lysozyme (IZM, P < 0.01) was apparantly decreased. 2. After injection of 6-OHDA in mice, the spleen weight, spleen index, thymus weight, thymusindex, spleen lymphocyte population and serum IZM, igG contents were markedly decreased, whereas LTT, IL-2 and plaque forming cell (PFC) were umchanged, In contrast, the thymus lymphocyte population was markedly increased. 3. After EA of mice axotomized sympathetic nerve with 6-OHDA the above mentioned immune parameters deceased by 6-OHDA could be returned to normal or nearing normal levels. However, the unchanged LTT, IL-2 and the increased thymus lymphocyte population could be enhanced further, especially IL-2. These results suggest that peripheral sympathetic nerve may be participation in the EA-mediating immunological responses.

[Serum lipid-associated sialic acid (LSA) in diagnosing and monitoring ovarian cancer].

Serum from 161 patients with ovarian cancer, 28 patients with benign gynecologic disorders and 22 healthy women, was assayed for levels of tumor marker LSA, which were compared with CA125. The results showed that in the patients with ovarian cancer, the sensitivities of LSA and CA125 for the patients prior to surgery were 83.0% and 92.5%, respectively; the sensitivities for the recurrent patients after surgery were 73.7% and 82.5% respectively. A total sensitivity of 89.5% was obtained by combination of both markers. The positive predictive value of LSA and CA125 for the patients with suspected tumor recurrence were 89.4% and 100%, respectively, and their corroborative rate with the postoperative courses were 94.4% and 100%, respectively. Thus serum assay of LSA, can be used in monitoring patients with ovarian cancer. The technique for determination of serum level of LSA is much more simple and less expensive than the radioimmuno-assay of serum level of CA125.

The effects of supercooling chemicals on myocardial ultrastructure: a transmission electron microscopy case study.

Extended ischemia results in organ infarction which limits the availability of donor hearts. Hypothermic storage extends heart preservation by effectively stopping cellular metabolism, thereby preventing toxic accumulations of metabolic wastes and depletion of energy stores. However, cell swelling as a result of ion concentration changes and cell laceration due to ice crystal growth are consequences of hypothermic ischemia. Supercooling successfully preserves hearts for an extended time without associated myocardial necrosis. The efficacies of four supercooling preservative solutions, containing hypertonic glucose, polyethylene glycol, and or winter flounder antifreeze protein, are assessed using the Langendorff isolated organ perfusion apparatus and transmission electron microscopy. Polyethylene glycol seems the most effective in preventing myocardial necrosis possibly by dehydrating, minimizing cellular ice formation, protecting against cell swelling, and functioning as an antioxidant. Hypertonic glucose seems the most effective in reducing cell swelling; it may also depress solution freezing points, bind water, adjust both intra- and extracellular osmolarities, stabilize proteins, and assist in adenosine triphosphate (ATP) production. Antifreeze protein seems to bind effectively to ice and inhibit its growth; it may also reduce membrane permeabilities to Ca2+ and K+ ions.

Effect of warm ischemic injury on morphology and viability of aortic valves in rats.

Warm ischemia is one of the most important causes of valvular damage from death of donor to its valve harvesting. This study aimed at using qualitative and quantitative methods to characterize warm ischemic injury through models of SD rat's aortic valves and design to show the relationship among ultrastructural, biological and biochemical changes concerning with the length of warm ischemia time (WIT). 102 harvested SD rat's aortic valves were divided into 6 groups of different ischemic time for this study. 432 photomicrographs of transmission electron microscopy (TEM) were put into computer for analysis. The volume ratio of nucleus to plasma in cells (Vnp) and the ratio of extramembrane area to volume of mitochondria (S/V) were used to characterize the degree of valvular cell injuries. Valvular cells culture and biochemical metabolism including glucose degradation and 3H-TdR absorption rate were adopted. The valvular cells depicted a significant decrease and 3H-TdR taking-up also being inhibited under the influence of prolonged WIT.

A Gly1127Ser mutation in an EGF-like domain of the fibrillin-1 gene is a risk factor for ascending aortic aneurysm and dissection.

Ascending aortic disease, ranging from mild aortic root enlargement to aneurysm and/or dissection, has been identified in 10 individuals of a kindred, none of whom had classical Marfan syndrome (MFS). Single-strand conformation analysis of the entire fibrillin-1 (FBN1) cDNA of an affected family member revealed a G-to-A transition at nucleotide 3379, predicting a Gly1127Ser substitution. The glycine in this position is highly conserved in EGF-like domains of FBN1 and other proteins. This mutation was present in 9 of 10 affected family members and in 1 young unaffected member but was not found in other unaffected members, in 168 chromosomes from normal controls, and in 188 chromosomes from other individuals with MFS or related phenotypes. FBN1 intragenic marker haplotypes ruled out the possibility that the other allele played a significant role in modulating the phenotype in this family. Pulse-chase studies revealed normal fibrillin synthesis but reduced fibrillin deposition into the extracellular matrix in cultured fibroblasts from a Gly1127Ser carrier. We postulate that the Gly1127Ser FBN1 mutation is responsible for reduced matrix deposition. We suggest that mutations such as this one may disrupt EGF-like domain folding less drastically than do substitutions of cysteine or of other amino acids important for calcium-binding that cause classical MFS. The Gly1127Ser mutation, therefore, produces a mild form of autosomal dominantly inherited weakness of elastic tissue, which predisposes to ascending aortic aneurysm and dissection later in life.

[Endodermal sinus tumor of the testis in children: report of 9 cases].

Nine children with endodermal sinus tumor of testis have been treated since 1982. Their age ranged from 7 months to 5 years. Radical inguinal orchiectomy was carried out in all the patients. Four patients have received irregular chemotherapy. The follow-up showed that 4 died of the metastasis within 5 years and 5 (stage I clinically) survived without evidence of the recurrence for 1.5-7 years after operation. Serum AFP determination may give important information for the classification of the tumor and the evaluation of the follow-up results. For the cases of clinical stage I, radical orchiectomy, short course of chemotherapy, and frequent observations of AFP level are advocated.

[Preparation and characterization of monoclonal antibodies against esterase of malathion-resistant Culex quinquefasciatus].

Five clones of monoclonal antibodies (McAbs) were generated from BALB/c mice which were activized with PPD and immunized with esterases extracted from malathion-resistant Culex quinquefasciatus. All 5 clones of McAbs belonged to the IgG1 subtype, additive index (A.I.) of every two McAbs did not exceeded 50%. After a 1 h of incubation, the McAbs at the titer of 1:1,000 completely inhibited the activity of the amplified esterases. Of 8 strains of adult mosquitoes studied by Western-blot assay, all exhibited the 64 kDa esterase band recognized by the McAbs, six (SP, FS, SH, CD, RD, RM) showed more bands of different MW (43-60 kDa), the higher the organophosphorus resistant level, the more and the denser the bands. The above McAbs had little or no effect against the esterases of dimethrin-resistant strain and the larva stage of malathion-resistant Culex quinquefasciatus.

[Effects of selenium and vitamin E on arachidonic acid metabolism in experimental myocardial ischaemia].

Wistar rats were fed with grain produced in the areas prevalent of Keshan Disease, and supplemented by selenium (Se) and vitamin E (VE) for ten weeks, and an acute myocardial ischaemia animal model was established by ligation of left anterior descending coronary arteries. Results indicated activities of phospholipase A2 (PLA2), creatine kinase (CK) and lactate dehydrogenase (LDH), and levels of arachidonic acid (AA), thromboxane A2 (TXA2), leucotriene C4 (LTC4) and lipid peroxides (LPO) increased, but level of prostacyclin (PC) and activities of glutathione peroxidase (GSH-PO), copper-zinc superoxide dismutase (Cu-Zn-SOD) and catalase (CAT) decreased 48 hours after ligation of the coronary vessels in rats without supplement of Se and VE. Supplement of Se or VE in their feed could reverse the above effects and the best results could be got by supplement of both. It suggested that supplement of adequate amount of Se and VE in their feed could affect metabolism of AA and levels of its metabolites, improve myocardial function and metabolic status and reduce myocardial damage in rats under the stress of acute myocardial ischaemia.

[Karyotype analysis of Candida species by PFGE and its use in classicification].

Electrophoretic karyotype in 7 medically important Candida species was analyzed by pulsed-field gel electrophoresis. The chromosomal DNAs of the Candida species were separated into 4-10 bamds ranging in size from 0.5-2.8 Mb. These patterns were species-specific, so the electrophoretic karyotype can be used as a genotype basis for classification and identification of Candida species.

[Angiotensin II increasing the inhibitory effects of electroacupuncture on the nociceptive discharge of neurons in the spinal dorsal horn].

Experiments were performed on 53 rats. The nociceptive discharges of spinal dorsal horn neurons were extracellularly recorded by glass microelectrodes and tested for the effects of angiotensin II (A II) on the nociceptive discharges of spinal dorsal horn neurons and AII on the inhibitory effects of electroacupuncture (EA). The results showed as follows: (a) The nociceptive discharges of spinal dorsal horn neurons might be inhibited (25/35) or facilitated (10) with A II (50-500 ng/20 ul) applied to the surface of dorsal spinal cord (L2-3); (b) The facilitory effects (5/5) were observed with A II (2 ug/20 ul) applied to the surface of dorsal spinal cord; (c) The inhibitory effects of EA (20 Hz, 2-4 V, 5 min) could be increased with A II (250 mg/20 ul, 8/9). Therefore, A II may modulate the nociceptive response of spinal dorsal horn neurons and influence the effects of EA, and different effects can be produced when the different dosage of A II is applied to the surface of dorsal horn.

[A study of relation of cardiac function and deformity degree in children with pectus excavatum].

40 children with pectus excavatum were divided into subgroups according to the FI (> 0.2 or < 0.2) and underwent M-ME examination for their cardiac function. 28 of 40 patients had their cardiac function and thoracic index re-examined after the operation. The results showed the cardiac function of children with pectus excavatum was damaged as FI of the patients was less than 0.2 and their cardiac function was getting well as their thoracic index improved.

[Intracerebroventricular injection of interleukin 1 receptor antagonist antagonises on the generation of immuno-suppressive factor in restraint mice].

Our previous work showed that a lymphocyte proliferation suppressing factor could be found in the serum of restraint mice. In the present work, it was found that intracerebroventricular (icv) injection of IL-1 receptor antagonist (IL-1Ra) was found capable of suppressing the production of such a serum protein under restraint stress. Nearly complete suppression could be achieved by 5.0 micrograms IL-1Ra. Intracerebroventricular injection of IL-1 beta (1 pg), however, increased the generation of the supressive protein. Neither intraperitoneal (ip) injection of IL-1Ra or IL-1 beta had any effect on the generation of the protein. The fact that icv. injection of a very small dose of IL-1 beta (0.06 fmol) was effective on the generation of the supressive protein led us to suggest that IL-1 in brain might act as an important mediator between CNS and the immune system.
