A procedure for fast screening of urine sample is described. TLC and GC equipped with NPD enable detection of therapeutic doses of drugs to be made in a short time. Tables of retention values for TLC and GC are also presented. TLC detects drugs with a sequence of three reagents. GC screening is carried out with a temperature programmed run of 30 degrees C/min up to 290 degreesC. For the determination of retention indices new polar phases with suitable thermal stabilities such as OV-225 and OV-275 are used.
Extraction procedure and detection of bis(2-chloroethyl)sulfide from urine samples of two patients are described using gas chromatography-mass spectrometry. Concentrations are found in the low ppb-range. The result is discussed in connection with the reactivity of this substance and the corresponding metabolites.
One of the basic aims of forensic toxicology is the identification of previously unknown drugs and poisons. This is frequently achieved using the combination of gas chromatography and benchtop quadrupole or ion trap mass spectrometers. The influence of matrix loading on the mass spectral quality was tested, and it was found that a realistic amount of matrix changed the pattern of the spectra obtained by the ion trap mass spectrometer. Disturbed mass spectra led to unsuitable suggestions from the library search and thus rendered the identification of the "general unknown" more difficult. On the other hand, higher selectivity and lower detection limits favored the ion trap technology for target analysis because of the capability of MS-MS.
Morphine and 6-acetylmorphine in illicit heroin samples can be easily determined by propionylation in aqueous solution (disodium hydrogen phosphate buffer). Heroin, 3-propionylmorphine and 6-acetyl-3-propionylmorphine are separated on methyl-phenylpolysiloxane liquid phases (5% phenyl content). The analytical procedure and the corresponding statistical data are presented together with the retention indices of typical street heroin concomitant drugs. The procedure can also be applied to the analysis of opium preparations and very dilute heroin (morphine) samples.
We investigated antigenic changes of murine glomerular basement membrane (GBM) collagen type IV after oral feedings of 3-cis- or 4-cis-hydroxyproline. As shown by thin layer chromatography and gas chromatographic-mass spectrometric studies, 3-cis- and 4-cis-hydroxyproline were incorporated into the GBM collagen instead of the normal trans isomer and led to a considerable reduction of the collagen antibody reactivity on indirect immunofluorescence, passive hemagglutination and radioimmunoassay when compared to GBM collagen of control animals. We speculate that antigen modification due to the incorporation of stereoisomers could serve as a model for basement membrane immunopathology.
The body of a 76-year old female pensioner was recovered from the Danube as adipoceratous torso after the person had been missing for almost two years. Identification first of all based on comparing postmortem and radiological findings which revealed correspondence of numerous features. The missing woman had been receiving long-term drug therapy of melperone (active substance of the neuroleptic Buronil) in doses of 25 mg three times daily. Combined gas chromatography and mass spectrometry succeeded in detecting the characteristic mass fragment of melperone in the adipoceratously transformed bone marrow of the water-logged corpse; in the remaining soft tissues outside the bones the concentration was below detection range. The reported case exemplifies that demonstration of low drug levels resulting from therapeutic ingestion can contribute to the identification of extensively decomposed bodies and to the correct relating of severed parts.
