[Effects of light and plant growth regulators on growth of normal and hairy root of Lycium barbarum in vitro].

OBJECTIVE: To study effects of the cycle of light and exogenous plant growth regulators on growth of normal root and hairy root of Lycium barbarum and establish a long-term system of in vitro culture of L. barbarum roots. METHOD: Using normal roots from aseptic seedlings and hairy roots originated from Agrobacterium rhizogenes A4-mediated genetic transformation of L. barbarum, the growth of root segments was examined after 20 days of culture under different culture conditions. RESULT AND CONCLUSION: When cultured on the MS medium with different plant growth regulators (IAA, IBA, NAA), normal root segments of L. barbarum could elongate and branch, and the addition of 1 mg x L(-1) IBA into the medium was the most suitable for long-term in vitro culture of L. barbarum root. When cultured on MS medium without hormone, light promoted the elongation and lateral root formation of type II hairy root, but not of type I hairy root. Among three tested hormones, IAA at low concentration was the best for growth of type I hairy root, while 1 mg x L(-1) IBA was the most suitable for growth of type II hairy root. With the increase of hormone level added into MS medium, callusing frequencies of type I, II hairy roots were increased, at the same time, the formation of lateral root from hairy root or callus was inhibited.

[Construction of RNA interference expression vectors of human neuropathy target esterase and its inhibition for expression of NTE in mammalian cells].

OBJECTIVE: To construct the RNA interference expression vector for expression of human neuropathy target esterase (NTE) gene in mammalian cells. METHODS: Spe I and Xho I-digested insert from pSUPER, which comprised H1 RNA polymerase III promoter and the multiple cloning sites, were cloned into the compatible in the pcDNA3.1 (+) to generate pSUPER/neo that could express small interfering RNA in mammalian cells. The annealed oligos targeting the expression of NTE were ligated into pSUPER/neo vector digested with Bgl II and Hind III to generate pSUPER/neo-NTE, which was transfected into COS7 and SH-SY5Y cells. The inhibitory effect of the expression of NTE was detected by western blot analysis and the enzyme activity assay. RESULTS: pSUPER/neo-NTE could stably express double-stranded RNA of NTE. The expression of pSUPER/neo-NTE in COS7 and SH-SY5Y cells could efficiently inhibit the activity of NTE in the mammalian cells. CONCLUSION: Stable eukaryotic expression vector of double-stranded RNA of NTE, pSUPER/neo-NTE, has been constructed successfully with promoter substitution strategy.

Cloning and sequencing of junction fragment with exons 45-54 deletion of dystrophin gene.

OBJECTIVE: To study the mechanisms of dystrophin gene deletion, the junction fragment with exons 45-54 deletion were cloned and sequenced. METHODS: A Duchenne muscular dystrophy (DMD) patient with exons 45-54 deletion has been substantiated by PCR amplification of the exons. Then we used a PCR-based genome-walking method for localizing the breakpoints in introns 44 and 54. At last, the deletion-junction fragment was directly amplified by PCR approach with forward and reverse primers annealing to a DNA sequence as close as possible to the breakpoints in introns 45 and 54. The sequencing result of the deletion-junction fragment was compared with the normal intronic sequences. RESULTS: A total of 2716 bp sequence containing the junction fragment was obtained. The 5' breakpoint was located in LINE/L1 element of intron 44 and close to a matrix attachment region (MAR). The 3' breakpoint was located in the minor potential MAR with topoisomerase II cleavage sites around. Beside the 3' breakpoint there was a 6 bp palindromic sequence. A 4 bp microhomologous sequence (AGAG) was in the joint of the deletion-junction fragment. CONCLUSION: The nonhomologous recombination caused by L1 repeated element, topoisomerase II cleavage sites, MARs and the nonhomologous end joining of microhomologous sequence may be the important factors in this huge gene fragment deletion.

[Prognostic value of N-terminal-pro-brain natriuretic peptide on admission in patients with chronic heart failure].

OBJECTIVE: Heart failure is responsible for a huge burden in hospital care. Our goal was to evaluate the value of N-terminal-pro-brain natriuretic peptide (Nt-proBNP) on predicting death or hospital readmission after hospital discharge in patients with chronic heart failure (CHF). METHODS: From March 2003 to April 2005, 135 consecutive patients (97 male and 38 female, mean age 60.7 years +/- 13.1 years) with chronic heart failure [dilated cardiomyopathy (44%) and coronary heart disease (35%)] were included in this study. Plasma concentrations of the Nt-proBNP were measured by ELISA on admission. All patients received conventional therapy and were followed up for 24 months. The primary end point was death or readmission. RESULTS: (1) During the follow up period (640 days +/- 100 days), 11 patients died and 39 patients rehospitalized, the median Nt-proBNP level on admission was significantly higher in patients died during the follow up period (5908 ng/L) than that of rehospitalized patients (2768 ng/L, P = 0.038). Plasma Nt-proBNP level on admission were significantly higher in primary end point group (n = 50, 2947 ng/L) than that in non-primary end point group (n = 85, 917 ng/L, P < 0.01). (2) Variables associated with an increased hazard of death and/or rehospitalization were Nt-proBNP and NYHA degree when analyzed by logistic regression models. Increased Log Nt-proBNP was the strongest independent predictor of an adverse outcome of CHF (odds ratio 13.8, 95% confidence interval 2.29 to 2.78, P < 0.01). (3) Area under the curve for Nt-proBNP in evaluating prognosis of CHF patients was 0.885 (positive predictive value 88.5%, negative predictive value 11.5%). CONCLUSION: Nt-proBNP level on admission is a strong predictor of rehospitalization and death within 24 months after hospital discharge in patients with chronic heart failure.

[Experimental study of repairing damaged human amniotic epithelial cells with formulated fibrin clot and cell growth factor].

OBJECTIVE: To investigate whether damaged human amniotic epithelial cells (HAEC) could be repaired on the matrix of formulated fibrin clot in vitro and the effects of epidermal growth factor (EGF), basic fibroblast growth factor (bFGF) and transforming growth factor beta(1) (TGF-beta(1)) on the proliferation of HAEC. METHODS: Ring drill was used to drill the HAEC layer on culture sheets to make quantified models of damaged HAEC, on which the lacks were then covered with fibrin clot. Subsequently, EGF (EGF group), bFGF (bFGF group) and TGF-beta(1) (TGF-beta(1) group) of different concentration were added into the sheets respectively. After the predesigned culturing time, the growing and transiting conditions of HAEC were observed under inverted microscope after Giemsa stain. Also, the proliferating conditions of HAEC were detected by using 5-bromodeoxyuridine (BrdU). RESULTS: In all groups, HAEC could transit toward damaged area on fibrin clot and grow there. Higher transiting speed and larger cell numbers were observed in the EGF and bFGF groups followed by the control group, while the TGF-beta(1) group showed the relatively poorer results. Proliferating rates of HAEC were 17.8%, 28.0%, 35.3%, 51.6%, 34.1%, 34.2% and 26.0% respectively by EGF of different cultured concentration (1.0 ng/ml, 5.0 ng/ml, 10.0 ng/ml, 20.0 ng/ml, 40.0 ng/ml, 80.0 ng/ml and 160.0 ng/ml). Proliferating rates of HAEC were 18.0%, 35.7%, 43.0%, 52.7%, 67.4%, 43.6% and 30.5% respectively by bFGF of different cultured concentration (1.0 ng/ml, 5.0 ng/ml, 10.0 ng/ml, 20.0 ng/ml, 40.0 ng/ml, 80.0 ng/ml and 160.0 ng/ml). Compared with the control group, EGF groups (EGF concentration ranging from 10 ng/ml to 80 ng/ml) and bFGF groups (bFGF concentration ranging from 5 ng/ml to 80 ng/ml) showed better proliferating effects of HAEC (P < 0.05), especially the 20 ng/ml EGF group and 40 ng/ml bFGF group had the best proliferating results among their own respective groups (P < 0.05). Proliferating rates of HAEC were 17.1%, 15.1%, 9.3%, 6.2%, 4.8%, 3.6%, 2.0% and 1.2% respectively by TGF-beta(1) of different cultured concentration (0.1 ng/ml, 0.2 ng/ml, 0.4 ng/ml, 0.8 ng/ml, 1.6 ng/ml, 3.2 ng/ml, 6.4 ng/ml and 12.8 ng/ml). Proliferating rates of HAEC in TGF-beta(1) groups (TGF-beta(1) concentration ranging from 0.8 ng/ml to 12.8 ng/ml) were significantly lower than that in the control group (P < 0.05). CONCLUSIONS: HAEC could transit and grow on the matrix of fibrin clot and repair the damaged area. EGF and bFGF could obviously stimulate HAEC proliferation, while TGF-beta(1) might have the inhibitive effects.

[Analysis of the different proteomes between the acute leukemia cells and normal white blood cells].

This study was aimed to analyze the different proteomes between human acute leukemia (AL) cells and normal white blood cells by proteomic technology in order to lay the basis for diagnosing AL and understanding the mechanism of leukemogenesis. The proteins from AL cells of 40 AL patients identified by FAB classification and proteins from normal lymphocytes and granulocytes of 20 normal volunteers were separated by two-dimensional electrophoresis (2-DE), and the differentially expressed proteins between the two groups were identified by both matrix assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF-MS) and electronspray ionization (ESI)-MS/MS. The results showed that among the differentially expressed proteins between AL cells and normal lymphocytes and granulocytes, some proteins involved in the process of malignant transformation (such as Op18, NM23-H1), cell proliferation (such as PCNA) and apoptosis inhibition (such as tumor necrosis factor inhibitor protein) were found to be up regulated in AL cells. However, some proteins involved in differentiation and physiological functions of normal cells were down regulated in AL cells. It is concluded that there are many events involved in the process of leukemogenesis, expression of some proteins relating to the malignant transformation, cell proliferation and apoptosis inhibition are up-regulated in AL cells. The proteome analysis may provide a new approach to explaining the molecular mechanism underlying the pathogenesis of AL.

[Lysosome of Eisenia fetida as biomarker of soil pollution].

The lysosome of Eisenia fetida can accumulate neutral red dye rapidly. When exposed to environment pollution, the membrane of the lysosome is easily to be cracked, and the dye shall leak to the cytoplasm gradually, making the cell red. Therefore, neutral red retention time (NRRT) can reflect the damaged degree of lysosome. In this paper, the NRRT of the lysosome in coelomocytes of E. fetida was determined after exposed to different concentrations of acetochlor and selenium. The results showed that with increasing exposure time and increasing concentration of acetochlor and Se, the NRRT was decreased significantly, and had significant correlations with the concentrations of acetochlor in soil and of accumulated Se in E. fetida's body. NRRT could be served as a biomarker of soil pollution, and a promising tool for the early warning of potential danger.

[The relationship between hypermethylation of the PTEN promoter and laryngeal squamous cell carcinoma].

OBJECTIVE: To explore the relationship between hypermethylation of the PTEN gene promoter and laryngeal squamous cell carcinoma (LSCC). METHOD: Promoter hypermethylation and mRNA expression were detected by methylation specific PCR and RT-PCR in LSCC and normal laryngeal tissue. RESULT: Among the 40 patients with LSCC, hypermethylation of PTEN gene promoter was detected in 16 cases (40.0%). Among the nine normal laryngeal tissues, hypermethylation of PTEN gene promoter was detected in no case. The high and moderate expression rates of PTEN gene mRNA in nine normal laryngeal tissues were 77.8% (7/9) and 22.2% (2/9), respectively; The high, moderate, low and negative expression rates of PTEN mRNA in 15 laryngeal cancers with lymph node metastasis were 26.7% (4/15), 33.3% (5/15), 26.7% (4/15) and 13.3% (2/15), respectively. The PTEN mRNA expression in normal laryngeal tissues was higher than that in the laryngeal cancers with lymph node metastasis (P < 0.05). The high, moderate and low expression rates of PTEN mRNA in 10 highly differentiated laryngeal cancers were 60.0% (6/10), 30.0% (3/10) and 10.0% (1/10), respectively; The high, moderate, low and negative expression rates of PTEN mRNA in 12 low differentiated laryngeal cancers were 16.7% (2/12), 25.0% (3/12), 41.7% (5/12) and 16.7% (2/12), respectively. The differences of PTEN mRNA expression between high and low differentiation laryngeal cancers were statistically significant (P < 0.05). The hypermethylation rate of PTEN gene promoter in the low mRNA expression laryngeal cancers was 70.0% (7/10), the hypermethylation rate of PTEN gene promoter in the high mRNA expression laryngeal cancers was 23.5% (4/17). The hypermethylation rate of PTEN gene promoter in the low mRNA expression laryngeal cancers was higher than that in the high mRNA expression laryngeal cancers (P < 0.05). CONCLUSION: Hypermethylation of PTEN gene promoter is associated with loss of its transcription in laryngeal squamous cell carcinoma, the decreasing of PTEN mRNA expression may be related to lymph node metastasis and low differentiation of laryngeal squamous cell carcinoma.

[Application of pulsed-field gel electrophoresis typing in tracing and carrying out surveillance programs on O139 cholera outbreaks].

OBJECTIVE: To study the characteristics of molecular typing and phylogenic relationship among the Vibrio cholerae serogroup O139 strains isolated from environment and sea food samples during cholera outbreaks, in Sichuan province in 2004 and to trace the source of infections so as to support the ascertainment of epidemic control strategy. METHODS: Cholera toxin gene was detected by polymerase chain reaction amplification. Pulsed-field gel electrophoresis (PFGE) was used for subtyping of isolates and clustering of patterns was analysed with the software BioNumerics. RESULTS: In all the 72 strains under analysis, 68 appeared to be toxigenic while 4 from river water derived isolates were toxin gene negative. Sixty-seven strains were clustered into 16 PFGE patterns when digested with Not I. The patterns of toxigeinc O139 strains isolated from turtles in the markets were identical with the patterns of strains appeared in the outbreaks respectively. The PFGE patterns of isolates from different outbreaks were inconsistent. CONCLUSION: The sources of infection causing these outbreaks were complicated. Contaminated turtles might also be one of the major sources of outbreaks when being served at the dinner parties in Sichuan in 2004.

[Gene expression profiling of diffuse-type gastric cancer by cDNA microarray].

OBJECTIVE: To identify cancer-related genes in diffuse-type gastric cancer and to explore its molecular mechanism by cDNA microarray analysis. METHODS: A total of 22 pairs of diffuse-type gastric cancer tissue and the corresponding normal mucosa were taken and freshly frozen. cDNA microarray with 14,592 genes/ESTs was used. Genes were considered to be up- or down-regulated when the fluorescent intensity ratio between tumor and normal mucosa was over 2-fold in over 50% of the samples (P < 0.05). Hierarchical clustering of regulated genes was performed as a measure to study expressional similarity. Validation of array results was carried out by real time quantitative PCR (QPCR). RESULTS: Compared with those of corresponding normal mucosa, there were a total of 153 genes/ESTs up-regulated and 204 down-regulated in diffuse-type gastric cancer. Hierarchical clustering demonstrated that the genes belonging to the same subgroup displayed similar function. Most of the overexpressed genes were those related to cell adhesion, cell motility, matrix reconstruction, cell proliferation and/or signal transduction; while genes related to defense response, toxicoid metabolism, DNA repairing, nuclear-cytoplasmic transport and/or anti-apoptosis made up the main list of the underexpressed genes. Seven genes showed higher expression in TNM (T I + T II) group than in (T III + T IV) group. QPCR confirmed the array analysis results. CONCLUSION: Gene expression profiling by cDNA microarray analysis provides not only molecular understanding of biological properties of cancer, but may also be helpful in discovering new diagnostic markers and therapeutic targets in gastric adenocarcinoma.

[Association of blood and cerebrospinal fluid IgG contents and severity of craniocerebral injury].

OBJECTIVE: To explore the association of blood and cerebrospinal fluid (CSF) IgG contents and the severity of craniocerebral injury. METHODS: Totalling 143 patients with craniocerebral injury were divided into 3 groups according Glasgow Coma Scale (GCS) scores, namely the mild injury group with GCS score of 12-15 (n=41), moderate injury group with GCS score of 9-11 (n=71) and severe injury group (GCS score 3-8, n=32). Another 9 patients with congenital hydrocephalus were also recruited as the control group. The CSF and blood samples were collected from these patients to measure the IgG contents 4 and 14 days and 1, 2, and 6 months after the injury, respectively. Physical disabilities of the patients were estimated with Rappaport's disability rating scale (DRS), whose correlations with CSF and blood IgG contents were analyzed. RESULTS: In the early stage of moderate to severe brain injury, the IgG content was lowered significantly in the blood but increased in CSF as compared with the control patients (P<0.05), and the changes in CSF and blood IgG displayed a significant correlation with the severity of the injury (r=0.950, P<0.01). During the recovery of severe brain injury, DRS score was in inverse correlation with blood IgG content but in positive correlation with CSF IgG content (Spearman's correlation coefficient of 0.800, P<0.05). CONCLUSION: In the early stage of brain injury, detection of blood IgG content may help with the assessment of the injury severity. During the recovery of the injury, dynamic monitoring of blood and CSF IgG contents provides clues of the outcome of the patients and benefit the modification of the treatment plan.

[Study on novel mutations of MEF2A gene in Chinese patients with coronary artery disease].

OBJECTIVE: To explore the mutations of MEF2A gene in Chinese patients with coronary artery disease(CAD). METHODS: With polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and DNA direct sequencing, the mutation analysis of exon 11 of MEF2A gene was performed to 156 patients with CAD and 93 normal controls. RESULTS: By DNA sequence analyzing the samples of abnormal mobility shift of SSCP, the MEF2A gene mutations were found in three patients with CAD. One of mutations was 147130(C>A)(P431Q), and the second one was 21 bases deletion(147108-147128) which was leading to the absence of 7 amino acids (424QQQQQQQ430), and the third was 147191(G>T). Three mutations were all found in one patient, but meanwhile 21 bases deletion was found in the other two patients. CONCLUSION: Mutations in exon 11 of MEF2A gene exist in the patients with CAD, and the mutations may be pathological.

[Cloning and sequencing of the junction fragment of dystrophin gene with exons 3 to 5 deletion].

OBJECTIVE: To study the mechanisms of dystrophin gene deletion by cloning and sequencing the junction fragment of dystrophin gene with exons 3 to 5 deletion. METHODS: PCR was performed to verify dystrophin gene exons 3 to 5 deletion in a patient with Duchenne muscular dystrophy. A PCR-based genome-walking method was used to localize the breakpoint in introns 2 and 5, and the deletion-junction fragment was directly amplified by PCR approach with forward and reverse primers annealing to a DNA sequence as close as possible to the breakpoint in the introns 2 and 5. The sequencing result of the deletion-junction fragment was compared with the normal intron sequences. RESULTS: A sequence of 2113 bp containing the junction fragment was obtained. The 5' breakpoint was located in SINE/Alu element of intron 2, and the 3' breakpoint was located in the unique sequence near the sequence TTTAAA. The breakpoints were associated with a strong topoisomerase II cleavage site. A 26-bp fragment was inserted into the breakpoint and formed 3 duplications (GGCTTATATTTAA) of 13 bp around the deletion-junction fragment. CONCLUSION: Repeat sequence and strong topoisomerase II cleavage site around the breakpoint may predispose double-strand DNA breaks and recombination, which, in addition to the nonhomologous end-joining mechanism, may contribute as important factors to the gene deletion.

[Biological properties study and animal experiment of polylactide/polytrimethylene carbonate blends membrane].

Polylactide is a biodegradable and biocompatible biomaterial. Based on the PLA (Polylactide) membrane, we have produced a new PLA/PTMC (Polytrimethylene carbonate) blends membrane. The biological properties of this membrane were studied by cell toxicity experiment, acute toxicity experiment, skin irritant experiment, sensitization test, hemolytic test, micronucleus test and subcutaneous implantation test. The results demonstrated that the blends membrane has no toxicity and it does not cause skin irritation, hypersensitive reaction and hemolysis. The micronucleus ratio of the membrane is 1.3% +/- 1.0%, being less than 3%. The result of medullary micronucleus test was reported negative. The wounds were free from suppuration and necrosis after subcutaneous implantation in all periods. In the experimental application of this member to preventing adhesion after rabbit intestine operation, the membrane demonstrated good effect. In conclusion, PLA/PTMC blends membrane is a material with good biocompatibility.

[Clinical characteristic of optic neuritis and its relevancy with human leukocyte antigen].

OBJECTIVE: To investigate clinical characteristics of optic neuritis and its association with HLA (human leukocyte antigen). METHOD: The clinical data of 42 patients with optic neuritis were collected and flow polymerase chain reaction-reverse sequence specific oligonucleotide probe (PCR-rSSOP) was used to determine the genotype of HLA-DRB1. RESULTS: Two patients confirmed as Leber hereditary optic neuropathy by gene sequencing were excluded from the study. The complaint of pain was found in the patients (31/40) of optic neuritis. The visual field defect varied with patients and central and pericentral scotoma were demonstrated in 7 patients of 17. Of 40 patients with optic neuritis, 20 (50.0%) showed multifocal brain lesions in white matter by MR scanning. 26 of 38 patients had increased STIR signals. Inflammatory demyelinating changes were found in cerebral spinal fluid in 21 patients (80.8%). Out of demonstrated brain abnormal was revealed in 29 (72.5%) patients using MRI and/or CSF examination. The rate of HLA-DRB1(*)15 in the patients (35.0%) was much higher than control (19.4%), (chi(2) = 4.2328, P = 0.0397). Frequencies of HLA-DRB1(*)15 increased significantly in 26 female patients. The increased frequencies of HLA-DRB1(*)15 were associated with CSF abnormality and no relevancy was found with onset times, brain MR changes and increase of optic nerve signals. Acute optic neuritis responded well to intravenous megadose of methylprednisolone or immunoglobulin. CONCLUSIONS: Most of optic neuritis was characterized as clinical inflammatory demyelinating disease. HLA-DRB1(*)15 might correlated with genetic susceptibility of female patients with optic neuritis.
