[Expression of androgen receptor and estrogen receptor in carcinoma of larynx].

OBJECTIVE: To study the expressions of androgen receptor (AR) and estrogen receptor (ER) in the pathogenesis of laryngeal carcinoma. METHOD: The method of in situ hybridization was employed to detect the expressions of AR and ER in 63 specimens of laryngeal carcinoma, 20 specimens of normal tissue beside carcinoma and 20 specimens of laryngeal benign lesion. RESULT: The expressions of AR and ER in specimens of normal tissue and laryngeal benign lesion were negative. The expressions of AR in laryngeal carcinomas was 68.3%, the expressions of ER was 69.8%, which was not correlated with sex and clinical classification (P > 0.05). With the differentiation of pathologic-classify and clinical stage, the expressions of AR in carcinoma of larynx were significantly increased, but ER was opposite (P < 0.05). The expression of AR was observed more frequently in patients with lymph node metastasis, but the expression of ER was degraded (P < 0.05). Compared with carcinoma of larynx, the expressions of AR and ER in specimens of normal tissue laryngeal benign lesion were significantly different (P < 0.01). CONCLUSION: The expressions of sex hormone acceptor are concerned with the incidence of carcinoma of larynx. The expressions of AR and ER are closely related to pathologic-classify, clinical stage and lymph node metastasis. AR promoted the growth of laryngeal carcinoma, but ER restrained it. AR and ER could be marked to understand malignant degree and prognosis, but they were not correlated with sex and clinical classification. The difference in expression of AR and ER does not explain the difference of sex and incidence of laryngeal carcinoma difference in expression of AR and ER, it shows that the significant difference of sex in the incidence of carcinoma of larynx may be concerned with other factors.

[Preparation of graded zirconia-hydroxyapatite composite bioceramic and its immunocompatibility in vitro].

Graded Zirconia-hydroxyapatite composite bioceramic and simplex Zirconia-hydroxyapatite composite bioceramic were produced, and the extractes of these two of materials were made to evaluate their immunocompatibility. Scanning electron microscope (SEM) was used to detect the character of the surface of the graded composite bioceramic. Holding and without holding phytohemagglutinin (PHA), proliferation and activation of peripheralblood monocytes(PBMCs) cultured in the two extracts were studied. Cultured in PHA after 72 hours, the proliforation rate of the graded composite group was significantly higher than the simplex composite group (P < 0.01). There was no difference of apoptosis of PBMCs of the two groups (P > 0.05). Cultured in PHA after 24 hours, the ratio of CD3/CD69 positive PBMCs of the simple composite group was significantly higher than that of the graded composite material group (P < 0.01). The numbers of PBMCs activated by the graded composite material group were less than that of the simply composite material group and the technique of graded composite will be helpful to improve its immunocompatibility.

[cDNA microarray-based screening of differentially expressed genes in macrophages in the spleen of patients with portal hypertension and hypersplenism].

OBJECTIVE: To identify the differentially expressed genes associated with hypersplenism in patients with portal hypertension. METHODS: The total RNA were extracted from the macrophages isolated from normal spleen and the spleen of patients with portal hypertension and reversely transcribed to cDNA with the incorporation of fluorescent (cy3 and cy5)-labeled dCTP to prepare the hybridization probes. After hybridization of Biostar-H140s chip containing 14,112 spots of cDNAs with the prepared probes, the gene chip was scanned for fluorescence intensity to screen the differently expressed genes. Three gene chips were used for hybridization and only the genes with differential expression in all the three chips were considered to associate with hypersplenism in patients with portal hypertension. RESULTS: Totaling 896, 1330 and 898 genes were identified to be differentially expressed by the three chips, respectively, and 121 genes (0.86%) showed differential expression in all the three chips, including 21 up-regulated known genes and 73 down-regulated known genes. The differently expressed genes were functionally related with ion channels and transport proteins, cyclins, cytoskeleton, cell receptors, cell signal transduction, metabolism, immunity, and so forth. These genes might be involved in hypersplenism in the condition of portal hypertension. CONCLUSION: cDNA microarray-based screening of differentially expressed genes in the macrophages in the spleen may provide new insights into the pathogenesis of hypersplenism in patients with portal hypertension.

[The proteomics study of apoptotic NB4 cells induced by sodium butyrate].

OBJECTIVE: To explore the molecular mechanisms of apoptotic NB4 cells induced by the histone deacetylase inhibitor, sodium butyrate(SB). METHODS: SB was exposed to NB4 cells at a final concentration of 1.0 mmol/L. The untreated and treated cells were analysed with FACS and 2-dimensional electrophoresis (2-DE) at 0, 12, 24, 48 and 72 h. The changed protein spots were identified with MALDI-TOF-MS and ESI-TOF-MS/MS. RESULTS: SB induced apoptosis of NB4 cells. Twenty-one changed proteins involving apoptotic signal transduction, immunological regulation, transcriptional control, cellular metabolism, molecular transport and so on were identified. Thirteen of them had been reported to be related to apoptosis. CONCLUSION: SB can induce apoptosis and many functional protein changes in tumor cells. These results pave the way to further explore the anti-tumor mechanism of SB.

[Molecular mechanism of anti-apoptotic action of survivin in NCI-H446 lung cancer cells].

OBJECTIVE: To investigate cell apoptosis induced by survivin ASODN and clarify the precise mechanism of anti-apoptotic action of survivin. METHODS: Cells of lung cancer cell line NCI-H446 were treated with survivin ASODN at different concentrations. The changes of survivin mRNA and protein expression were assessed by RT-PCR and Western blot assay. The apoptosis index (AI) and proliferation index (PI) were determined by flow cytometry (FCM). After 500 mmol/L survivin ASODN treatment, cells were stained with Rh123 to detect changes of mitochondrial membrane potential (deltapsim) by FCM. The concentration of cytoplasmic cytochrome c (cyt-c) was continuously determined by ELISA. Relative activities of caspase-9 and caspase-3 were assessed by colorimetric assay. The expression of caspase-8 protein was measured by Western blot assay. The apoptotic rates of lung cancer cells induced by survivin ASODN with or without mitochondrial permeability transition pole (MPTP) inhibitor CsA treatment were assessed by FCM. RESULTS: Down-regulated survivin mRNA was shown to be in dose-dependent and time-dependent manners. Its maximal effect was achieved at a concentration of 500 nmol/L for 72 h, at which mRNA was down-regulated by 62.7%, the expression of survivin protein in NCI-H446 cells was also obviously decreased. After treatment with survivin ASODN at concentration of 500 mmol/L for 72 h, AI was 48.35%, higher than that of control, lipofectin, NSODN, survivin ASODN 100 mmol/L and 300 mmol/L groups (3.75%, 3.41%, 4.69%, 19.85% and 34.39%, respectively). PI was 24.38%, lower than that of control, lipofectin, NSODN, survivin ASODN100 and 300 mmol/L groups (75.74%, 73.12%, 71.76%, 51.03% and 38.94%, respectively). Deltapsim was decreased in 9.54% of NCI-H446 cells treated with survivin ASODN for 3 h and 97.06% for 24 h. Following it, release of cyt-c from mitochondria to cytosol and activation of caspase-9 and caspase-3 increased significantly. The above mentioned indicators changed with a time-dependent and time diversity relationship. In the presence of CsA, the apoptotic rate of lung cancer cells induced by survivin ASODN was decreased significantly. No up-regrulation and activation in caspase-8 protein was observed. CONCLUSION: Survivin inhibits apoptosis via regulation of mitochondrial-dependent pathway. survivin ASODN can not only induce apoptosis but also inhibit cell proliferation through blocking the expression of survivin mRNA and protein.

[Influence of neoadjuvant chemotherapy on microsatellite instability in gastric carcinoma].

BACKGROUND & OBJECTIVE: Most results from clinical trials of neoadjuvant chemotherapy for gastric carcinoma are certain. Meanwhile, microsatellite instability (MSI) is thought to be a new mechanism of cell canceration. This study was to investigate the influence of neoadjuvant chemotherapy on MSI in gastric carcinoma. METHODS: Two microsatellite loci D2S123 and D3S1298 in tumor samples from 48 patients with gastric carcinoma, received preoperative chemotherapy of 5-fluorouracil (5-FU) and leucovorin (CF), were analyzed by polymerase chain reaction (PCR), electrophoresis on 8% denatured polyacrylamide gel, and silver staining. The microsatellite statuses of preoperative and postoperative specimens were compared. Chi-square test was used to analyze the influence of neoadjuvant chemotherapy on MSI in gastric carcinoma. RESULTS: The detection rate of MSI was significantly higher in preoperative samples than in postoperative samples (31.3% vs. 12.5%, P=0.026). After neoadjuvant chemotherapy, 9 cases changed from MSI-positive to MSI-negative, while no MSI-negative cases became MSI-positive. CONCLUSION: The neoadjuvant chemotherapy regimen of 5-FU/CF can induce some MSI-positive gastric carcinoma change to MSI-negative, but has no effect on MSI-negative cases.

[Investigation and clinical significance of the micrometastasis of prelaryngeal lymph nodes in the patients with glottic carcinoma].

OBJECTIVE: To investigate the clinical significance of the micrometastasis of prelaryngeal lymph nodes in the patients with glottic carcinoma. METHOD: In prelaryngeal lymph nodes from 26 patients with glottic carcinoma, cytokeratin 19 (CK19) mRNA was detected with reverse transcription polymerase chain reaction (RT-PCR), comparing with the routine histopathologic methods (HE staining). RESULT: The positive expression of CK19 mRNA in prelaryngeal lymph nodes was detected in 8 patients (30.77%). While there was no positive finding by routine methods. The difference was significant (P < 0.05). In 8 cases with stage I glottic carcinoma, no micrometastasis in the prelaryngeal lymph node was found, and in 18 cases with stage II - VA glottic carcinoma, micrometastasis was found in 8 cases (44.44%) (P < 0.05). CONCLUSION: The detection of prelaryngeal lymph nodes micrometastasis with RT-PCR is more sensitive than with the routine histopathologic methods. The study of micrometastasis of prelaryngeal lymph nodes may provide information for the clinical management of the patients with glottic carcinoma.

[Photosynthetic and growth characteristics of different ecotype capsicum under weak light].

This paper studied the effects of weak light on the photosynthetic characteristics, assimilates allocation, yield formation, and seedling growth of different ecotype capsicum (hot pepper and sweet pepper). The results showed that under weak light, the net photosynthetic rate (Pn) , dark respiration rate (Rd ) , night respiration rate (Rn) , transpiration rate (Tr) , water use efficiency (WUE) , light compensation point (LCP) , CO2 compensation point (CCP) and carboxylation efficiency ( CE) of test capsicums decreased, while their apparent quantum yield (AQY) increased. The decrease of Pn was considered as a result of non-stomata restriction. Weak light increased plant height/stem diameter ratio and specific leaf area, but decreased root/shoot ratio, total dry mass, and seedling' s healthy index. Under the effect of weak light, the formation of biological and economic yields was inhibited significantly, and the allocation of assimilates to stem and leaf was increased while that to fruit decreased. Hot pepper under weak light had lower light compensation point, higher relative healthy index, higher relative yield per plant, and higher relative setting rate, showing a stronger capability of weak light tolerance than sweet pepper.
