Muraglitazar, a novel dual (alpha/gamma) peroxisome proliferator-activated receptor activator, improves diabetes and other metabolic abnormalities and preserves beta-cell function in db/db mice.

Muraglitazar, a novel dual (alpha/gamma) peroxisome proliferator-activated receptor (PPAR) activator, was investigated for its antidiabetic properties and its effects on metabolic abnormalities in genetically obese diabetic db/db mice. In db/db mice and normal mice, muraglitazar treatment modulates the expression of PPAR target genes in white adipose tissue and liver. In young hyperglycemic db/db mice, muraglitazar treatment (0.03-50 mg . kg(-1) . day(-1) for 2 weeks) results in dose-dependent reductions of glucose, insulin, triglycerides, free fatty acids, and cholesterol. In older hyperglycemic db/db mice, longer-term muraglitazar treatment (30 mg . kg(-1) . day(-1) for 4 weeks) prevents time-dependent deterioration of glycemic control and development of insulin deficiency. In severely hyperglycemic db/db mice, muraglitazar treatment (10 mg . kg(-1) . day(-1) for 2 weeks) improves oral glucose tolerance and reduces plasma glucose and insulin levels. In addition, treatment increases insulin content in the pancreas. Finally, muraglitazar treatment increases abnormally low plasma adiponectin levels, increases high-molecular weight adiponectin complex levels, reduces elevated plasma corticosterone levels, and lowers elevated liver lipid content in db/db mice. The overall conclusions are that in db/db mice, the novel dual (alpha/gamma) PPAR activator muraglitazar 1) exerts potent and efficacious antidiabetic effects, 2) preserves pancreatic insulin content, and 3) improves metabolic abnormalities such as hyperlipidemia, fatty liver, low adiponectin levels, and elevated corticosterone levels.

[Sequence analysis of a novel HLA-DRB1 allele, DRB1 * 1212].

OBJECTIVE: To investigate the molecular genetics basis for HLA novel allele HLA-DRB1*1212 in Chinese population. METHODS: Genomic DNA was extracted from whole blood by salting-out method. HLA-DRB1 gene exon 2 was amplified by PCR with group-specific primers from genomic DNA. PCR products were cut back from agarose gels and purified to sequence directly. The polymerase chain reaction-sequence specific oligonucleotide probes (PCR-SSO) was performed to confirm the mutations which were detected by sequencing in this study. RESULTS: The sequencing results showed HLA-DRB1 alleles of the proband as DRB1*090102 and the novel allele. The sequences of the novel allele have been submitted to GenBank (AY899825). Through BLAST analysis, the novel allele was found to be different from DRB1*120101 at position 199A-->C in exon 2, that results in an amino acid change from Ile to Leu at codon 67. CONCLUSION: This allele is a novel and has been officially named as DRB1*1212 by the WHO Nomenclature Committee.

[Protective effect of epimedium flavonoids injection on experimental myocardial infarction rats].

OBJECTIVE: To investigate the protective effect of epimedium flavonoids Injection (EFI) on experimental acute myocardial infarction (AMI) rats. METHODS: Rats were randomly divided into 6 groups, the acute myocardial infarction model was established by ligating left anterior descending branch of coronary artery (LAD). After operation, the rats in the sham-operation and model group were intravenous injected with 5% glucose injection, those in the positive medicine group were intravenous injected with nitroglycerin 0.3mg/kg, while rats in the low-, middle- and large-dose EFI group were intravenous injected with TFE in a dose of 10, 20, 40 mg/kg respectively. ECG was monitored before and after coronary artery ligation, and after treatment at different time points. At the same time, the millivolt of ST and ST-T segment were measured. The changes of serum creatine phosphokinase (CPK), lactate dehydrogenase (LDH), superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were determined, and the myocardial infarcted area was detected by MTT respectively 3 h after LAD. Results After intravenous injection of EFI in a dose of 10, 20, 40 mg/kg, the myocardial infarcted area of AMI rats could be decreased in different degree, the activity of serum CPK, LDH and the content of MDA decreased (P < 0.05 or P < 0.01), while the activity of serum SOD increased significantly (P < 0.05 or P < 0.01). It could began to lower the elevated ST-T segment 5 min after medication and the action could last for 3 h (P < 0.05 or P < 0.01). CONCLUSION: EFI has a protective effect against acute myocardial ischemia caused by LAD, and the effect is quickly initiated.

[Inhibitory effect of WT1 gene isoform transfection on proliferation of leukemia cell line NB4].

BACKGROUND & OBJECTIVE: The Wilms' tumor gene, WT1, encodes a zinc finger transcription factor, and is highly expressed in leukemia cells and negatively correlated with the prognosis of leukemia. WT1 mRNA undergoes alternative splicing at 2 sites resulting in 4 isoforms. The mRNA splice isoforms are constantly expressed in all tissues expressing WT1 with fixed proportions. The functional significance of WT1 isoforms expression in leukemia is unclear. This study was designed to explore the potential effects of exogenous WT1 gene isoforms on proliferation of leukemia cell line NB4 and its possible molecular mechanisms. METHODS: The recombinant eukaryotic expression vector pCB6+/WTA containing full-length human WT1 isoform WTA (-17AA/-KTS) cDNA, and pCB6+ vector with neomycin resistance gene were transfected into NB4 cells by electroperforation. The positive cell clones (NB4/WTA) were selected with G418. The transfected cells were monocloned by limited dilution. The expressing of WTA mRNA and protein in transfected cells was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot. The proliferating ability of leukemia cells was measured by trypan blue exclusion assay, MTT assay, colony formation assay and cell cycle analysis. The expression of p53, p21, Cyclin E, Cyclin D1 and Cyclin A1 was detected by semi-quantitative RT-PCR. RESULTS: WT1 gene isoforms were transfected into NB4 cells, and the cell line stably expressing exogenous WT1 gene isoforms was established successfully. The monoclonal cells overexpressing WT1 were obtained. The growth of NB4/WTA cells was slower than that of untransfected NB4 cells and NB4 cells transfected with pCB6+ vector (NB4/CMV): the growth of NB4/WTA cells reached plate phase [(78.7+/-18.0 x 10(4)/ml] at the 3rd day, while that of control cells reached plate phase [(146.0+/-21.0) x 10(4)/ml] at the 4th day. The inhibitory rates of NB4/WTA cells at different time points were significantly higher than those of control NB4 cells (P<0.005). The colony forming efficiency of NB4/WTA cells decreased markedly and became much lower when exposed to 0.2 micromol/L As2O3. The proportion of S phase cells was increased in NB4/WTA group. The expression of p21 and Cyclin A1 was higher in NB4/WTA cells than in control cells. CONCLUSIONS: Exogenous WTA gene could inhibit the proliferation and colony forming ability of leukemia cells through up-regulating the expression of p21 and Cyclin A1 and arresting cell cycle at S phase.

[Effect of acupuncture at Sanyinjiao (SP 6) on glucose metabolism in the patient of dysmenorrhea].

OBJECTIVE: To investigate the central modulating mechanism of acupuncture at Sanyinjiao (SP 6) in treatment of primary dysmenorrhea. METHODS: 18F-FDG positron emission tomograph imaging of whole brain was performed in six patients with primary dysmenorrhea during two stimulation: pseudo acupuncture and real acupuncture at Sanyinjiao (SP 6). The areas of cerebral glycometabolism change were obtained by using statistical parametric mapping (SPM). Meanwhile the pain intensity before and after pseudo-acupuncture and real acupuncture was assessed with 0-10 numerical pain intensity scale. RESULTS: No differences in the values of pain before and after pseudo-acupuncture (P > 0.05). The value of pain after acupuncture was significantly lower than that before acupuncture (P < 0.01). Most of the activated brain areas were shared with the areas activated by pain as described in the literature. The brain areas of increasing glycometabolism were ipsilateral lentiformn nucleus (globus pallidus, putamen), cerebellum and insula, bilateral thalamus, ipsilateral paracentral lobule, bilateral amygdala, contralateral midbrain, bilateral second somatosensory cortex, ipsilateral hippocampus and anterior cingulate, contralateral mammillary body. The brain areas of decreasing glycometabolism were limited in small cerebral cortex. CONCLUSION: Acupuncture at Sanyinjiao (SP 6) can relieve significantly the pain of the patient. Primary dysmenorrhea is cured mainly by activating the area involved in pain. It is indicated that acupuncture can relieve pain and balancing the pain-related central networks. Also neuroendocrine may play a role in the therapy.

[Alternative splicing of insulin receptor mRNA in cancer and type 2 diabetes mellitus: a review].

The insulin receptor exists in two isoforms that result from alternative splicing of exon 11 in the primary transcript. The two isoforms have different affinities for insulin and insulin-like growth factors, and mediate different biological actions by different intracellular pathways. Differential expression of the two isoforms not only might contribute to insulin resistance in type 2 diabetes mellitus, but also play an important role in the pathogenesis of cancer. Alternative splicing can be regulated by metabolic and hormonal factors, such as hyperinsulinemia and hyperglycemia, but the precise mechanism is unknown. It has also been demonstrated that there are certain important nucleotide sequences in insulin receptor gene that can influence alternative splicing.

[Primary culture of human omental preadipocytes].

OBJECTIVE: To establish a primary culture method of human omental preadipocytes. METHODS: Using enzyme-digesting method, fibroblast-like cells from the human omental adipose tissues were cultured, and then differentiated by conditional medium, and identified by oil red O staining. RESULTS: The cultured cells were highly homogeneous, and highly proliferative in 4-5th generation. During the process of induction by conditional medium, the cells became round-like and larger, and more adipose droplets were aggregated. By oil red O staining, we confirmed the differentiated cells were mature adipocytes. CONCLUSION: In human omental adipose tissues, there are some preadipocytes, which can differentiate into mature adipocytes with appropriate stimulus.

[The effect of glutamine on the growth, metabolism and endostatin production of microencapsulated rCHO cells].

Cell transplantation is a promising technology in cancer therapy, however, immunological rejection is the major problem of cell transplantation. Based on the permselective property of microcapsule membrane, encapsulated cells can be immuno-protected. The normal physiological state and function expression of cells can be maintained so as to realize allo- or xenotransplantation. The microencapsulated cells grow in three dimensions, giving a more biologically representative in vivo model, which hints difference in characters of growth and metabolism compared to the monolayer cells. Therefore, characterization of growth and metabolism of microencapsulated recombinant CHO cells is essential for further large-scale culture. In present study, the effect of concentration of glutamine on the growth, metabolism and endostatin production of microencapsulated cells was investigated. In the experimental range of initial glutamine concentrations from 2.69mmol/L to 9.05mmol/L in the culture of microencapsulated recombinant CHO cells, the maximum density of active cells and multiplication ratios almost kept constant. The specific consumption rate of glucose increased with lower initial glutamine concentration (2.69mmol/L). When initial glutamine concentration was much higher (7.91mmol/L to approximately 9.05mmol/L), the specific consumption rates of both glucose and glutamine increased while the efficiencies of glucose and glutamine decreased. The highest efficiencies of glucose and glutamine utilization were observed with initial glutamine concentration of 4.97mmol/L. It was also demonstrated that glutamine had significant effect on the accumulation of endostatin. The accumulative concentration of endostain reached its peak of 546.36 ng/mL with the initial glutamine concentration of 4.97mmol/L.

[The changes of the muscular fiber phynotype at different condition of nerve injury].

OBJECTIVE: To observe the change of the muscular fiber phynotype at different condition of nerve injury. METHODS: Rabbits were used as an animal model in this study. The trigeminus and facial nerves of the animal were simultaneously severed (group 1) or only the latter was severed (group 2). The morphologic change of the muscular fibers was observed with histochemical methods; the tension of contraction and the threshold value of electrical stimulation were observed with electrical physiological methods. RESULTS: Trigeminus nerve play an important role in delaying muscular atrophy after facial nerve was severed. The atrophy degree of type II a and II b muscular fibers was less in group 2 than that in group 1. The change of type 1 muscular fibers was not affected by trigeminus nerve. New muscular fibers emerged in group 2. The tension of contraction and the threshold value of electrical stimulation were better in group 2 than that of in group 1. CONCLUSIONS: Sensory nerve could delay muscle atrophy after denervation. Sensory nerve should be repaired simultaneously when the motor nerve was sutured. The results of this study would facilitate clinical treatment for facial palsy.

[Seminal parameter analysis in noninflammatory chronic prostatitis/chronic pelvic pain syndrome].

OBJECTIVE: To investigate seminal parameters in noninflammatory chronic prostatitis/chronic pelvic pain syndrome (CAP III B). METHODS: A total of 74 consecutive cases of patients who had been diagnosed as CAP III B and 46 cases of controls were included in the study. Severity of symptoms in men with CAP III B was defined according to the NIH Chronic Prostatitis Symptom Index (NIH-CPSI). All of them underwent a 'four glass-test' including leukocyte determination in expressed prostatic secretions (EPS), voided urine after prostatic massage (VB3) and ejaculate semen followed by analysis according to WHO. The analysis included seminal volume, pH, duration of liquefaction, sperm density, vitality, motility(a + b) and morphology. Correlations between the duration or the severity of symptoms and spermiogram results in patients with CAP III B were assessed respectively. RESULTS: The CAP III B group and the control group differed significantly in ejaculate volume, duration of liquefaction and motility, while the remaining parameters did not differ significantly. The duration of chronic pelvic pain showed apparently positive correlationship with liquefaction time, while the symptom duration negatively correlated with sperm motility. The NIH-CPSI score had no significant relationship with seminal volume, duration of liquefaction and sperm motility. CONCLUSION: Our results indicate that CAP III B can have a significant negative impact on sperm volume, liquefaction and motility. Our data also supports the results that the longer the duration of symptoms, the more influences on semen liquefaction and motility might be.

[Changes of tight junction and Cx43 expression in microvessel endothelial cells of mouse lung induced by bleomycin].

OBJECTIVE: To investigate the changes of expression of connexin-43 (Cx43) and the tight junction of microvessel endothelial cells (EC) to approach the effects in bleomycin (BLM) induced pulmonary fibrosis (PF). METHODS: Forty healthy rats were equally and randomizedly divided into the control group and the experiment group. In both group, vWf in blood serum was measured with ELISA method on 3rd, 7th, 14th, 28th day after BLM treatment. Rats in each group were infused with lanthanum nitrate on 3rd, 7th, 14th, 28th day after BLM treatment. The lung samples were made and the tight junction and the distribution of the granules of lanthanum in the microvessel EC were observed with transmission electron microscopy in the control and experiment groups. The lung microvessel EC of the rats in each group were preserved by tissue culture methods at the same time, and the expression of Cx43 were observed by laser scanning confocal microscopy. RESULTS: The serum vWf in the peripheral blood of the experiment group was significantly higher than that of the control group, and was the highest on 3rd day after BLM treatment (P < 0.01). The blood vessel EC of the control group were intact. The basement membrane was uninterrupted. Granules of lanthanum nitrate did not penetrate the tight junction of EC. The width of junction gap in the experimental group was increased and the lanthanum granules of high density were found deposited in the linear form in the gap junction. Low expression of Cx43 was observed in experiment group. The expression rate of Cx43 was 25%, 38%, 45% and 71% respectively on 3rd, 7th, 14th, 28th day, significantly less than those in the control group (P < 0.05). CONCLUSION: It may be the important pathological basis for the BLM induced abnormality of the interstitial tissues in the lung that the tight junction of EC is continuously in the open state, which causes the effusions of inflammatory cells and the corresponding cytokine secretion, and thus initiates the overproliferation of fibroblasts.

[Cloning and sequence analysis of a novel member of the rab gene family from Euplotes octocarinatus].

Rab proteins belong to a subfamily of small GTP-binding proteins of the Ras superfamily, which play an important role in intracellular vesicular traffic. In this study, a rab gene was obtained from Euplotes octocarinatus by polymerase chain reaction (PCR) and RT-PCR. The rab gene from macronucleic DNA was 884 bp in length, including non-coding regions and telomeric sequences at both ends. The rab gene from micronuclear DNA (723 bp), lacking of internal eliminated sequences, was identical to rab gene from macronuclear DNA. RT-PCR showed that the opening reading frame of the rab gene was 663 bp long. The rab gene from macronuclear DNA contained an intron of 60 bp at the position from 153 bp to 212 bp of macronuclear DNA. The rab gene had two in-frame TGAs encoding for cysteine in Euplotes octocarinatus. The rab gene used TAG as stop codon, which was the first report in Euplotes octocarinatus. The result of BLAST in NCBI demonstrates that the Rab shares a homology of 49-52% at the amino acid level with Rab1 proteins from a number of other eukaryote, which suggesting that the Rab is a Rab1 homolog. The rab gene was therefore designated Eo-rab-1N (GenBank accession number: DQ105562). The evolution of Eo-rab-1N was analyzed using phylogenetic tree of amino acids sequences of Rab1 obtained from GenBank.

[Effect of prepared radix Polygoni multiflori on the elevation of extracellular hydroxyl radical in striatum of rats induced by intracerebral perfusion of 6-hydroxy dopamine].

OBJECTIVE: To investigate the effect and mechanism of prepared Radix Polygoni Multiflori (RPM) on the elevation of extracellular hydroxyl radical in striatum of rats induced by intracerebral perfusion of 6-hydroxy dopamine (6-OHDA). METHODS: Cerebral microdialysis was used to establish the model. Hydroxyl radical was captured by salicylic acid, and 2,3-dihydroxy benzyl acid (2,3-DHBA) and 2,5-dihydroxy benzyl acid (2,5-DHBA) formed by hydroxyl radical in vital brain were measured by high performance liquid chromatography-electrochemical detector (HPLC-ED). RESULTS: After perfusion of 6-OHDA in brain of rats, the levels of 2,3-DHBA and 2,5-DHBA in the model group increased rapidly. The former was higher during the whole course of observation (P<0.01), while the latter was higher at most time points than that in the control group (P<0.05 or P<0.01). The level of 2,3-DHBA in the RPM group was lower than that in the model group at 5 time points (P < 0.05, P < 0. 01). CONCLUSION: RPM could inhibit the elevating of extracellular hydroxyl radical in striatum of rats induced by intracerebral perfusion of 6-OHDA, indicating one of the brain protective mechanisms of RPM may be related to its anti-oxidation effect.

[The association of cardiovascular risk factors and prevalence of carotid atherosclerosis at 10 years].

OBJECTIVE: To explore the association of cardiovascular disease (CVD) risk factors and its impact on the 10-year prevalence of carotid atherosclerosis. METHODS: Two surveys for CVD risk factors were carried out in a population consisting of 1323 subjects in Beijing in both 1992 and 2002 and a direct measurement of carotid arteries with B-mode ultrasound imaging were performed in 2002. Intima-media thickness (IMT) and plaque of carotid arteries were regarded as the indicators of carotid atherosclerosis and main CVD risk factors were treated as the impact factors. RESULTS: (1) 10-year prevalence of carotid atherosclerosis continually increased with elevated baseline risk factors levels. Age, smoking, hypertension and hypercholesterolemia were found to be independently associated with carotid atherosclerosis. (2) 10-year prevalence of carotid atherosclerosis increased with the elevated number of risk factors clustering in individuals. (3) The prevalence of IMT and plaque were 67.1% and 42.6% in subjects with CVD risk factors in both 1992 and 2002 surveys with Odds Ratios being 1.7 and 3.1 times as those without risk factors. CONCLUSION: Traditional CVD risk factors exert impact on CVD and carotid atherosclerosis with different degrees and carotid atherosclerosis can be regarded as an early evaluation indicator for risk of atherosclerotic diseases.

[Hepatocyte growth factor did not enhance the effects of bone marrow-derived mesenchymal stem cells transplantation on cardiac repair in a porcine acute myocardial infarction model].

OBJECTIVE: To evaluate the impact of combined therapy with transplanting bone marrow-derived mesenchymal stem cells (BM-MSCs) via noninfarct-relative artery and hepatocyte growth factor (HGF) in a porcine myocardial infarction (MI) model. METHODS: BM-MSCs were obtained from pig bone marrow, expanded in vitro with a purity of > 50%. MI was induced by ligating the distal left anterior descending artery in pigs. Eighteen animals received BM-MSCs cells (5 x 10(6)/ml, n = 6), BM-MSCs cells (5 x 10(6)/ml) plus HGF (4 x 10(9) pfu, n = 6) or equal volume culture medium (IMDM) via non-infarct-related artery at four weeks after MI. Gated myocardial perfusion imaging and coronary angiography were performed before and four weeks after transplantations. Histological examination was also performed 4 weeks after transplantation. RESULTS: LVEF measured by gated myocardial perfusion imaging was similar among groups before transplantation and significantly increased in BM-MSCs (45 +/- 3 vs. 34 +/- 2%, P < 0.05) or BM-MSCs + HGF (46 +/- 6 vs. 34 +/- 3%, P < 0.05) treated animals while remained unchanged in IMDM (30 +/- 3 vs. 32 +/- 2%) treated animals 4 weeks post transplantation. Similarly, capillary density was also significantly higher and myocardial perfusion defect scores significantly decreased in BM-MSCs or BM-MSCs + HGF treated hearts than that in IMDM treated hearts. However, all these changes were similar between BM-MSCs and BM-MSCs + HGF groups. Rentrop score was similar before and 4 weeks after transplantation among various groups. CONCLUSION: HGF in combination with BM-MSCs transplantation did not enhance the cardiac repair effects of BM-MSCs transplantation alone and BM-MSCs transplantation did not improve collateral circulation in this model.

[Serum total cholesterol and 10-year cardiovascular risk in a Chinese cohort aged 35-64 years].

OBJECTIVE: To evaluate the association between serum total cholesterol (TC) and the risk of cardiovascular diseases (CVD) in subjects aged 35-64 years. METHODS: This prospective study was performed from 1992 to 2002 in 11 provinces of China and the association between baseline TC level and the development of cardiovascular diseases was analyzed in 30 384 subjects aged 35-64 years using Cox multivariate proportional hazards regression. RESULTS: (1) Compared with the group of TC < 3.64 mmol/L (140 mg/dl), multivariate-adjusted relative risk of ischemic cardiovascular disease (ICVD, including CHD and ischemic stroke) increased continuously with increasing TC level. (2) TC level was positively related to the occurrence of ischemic stroke, but not to hemorrhagic stroke. The risk of CHD increased by 74% (RR = 1.743) and the risk of ischemic stroke increased by 12% (RR = 1.119) in the group with TC > or = 5.72 mmol/L compared with the group with TC < 5.72 mmol/L (220 mg/dl). (3) High serum cholesterol was responsible to 5.9% of ICVD, 11.7% of the acute CHD and 2.9% of the acute stroke. (4) At each TC level, the 10-year risk of ICVD increased in the presence of other risk factors. CONCLUSION: Starting from TC of 3.64 mmol/L, the risk of ICVD increases continuously with increasing TC level. Multiple risk factor intervention should be strengthened to reduce the overall risk of CVD.
