[Effects of sense vascular endothelial growth factor cDNA transfection on mononuclear chemotaxis protein-1 expression in rat pulmonary microvessel endothelial cells].

OBJECTIVE: To study the transformation characteristics of tight junction of microvessel endothelial cells in bleomycin (BLM) induced pulmonary fibrosis (PF), and the effects of vascular endothelial growth factor (VEGF) on mononuclear chemotaxis protein-1 (MCP-1) mRNA expression in pulmonary microvessel endothelial cells (EC). METHODS: Forty healthy rats were equally divided into a control and an experimental group in random. In the experimental group, PF were induced by BLM application. In each group, 10 rats were instilled by lanthanum sal at 3, 7, 14 and 28 d after BLM application, and lung samples were made and observed by transmission electron microscopy. The pulmonary microvessel EC of the other 10 rats in each group were preserved by tissue culture methods at the same time, and the cells were transfected with sense VEGF cDNA, and then MCP-1 mRNA expression was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) technique. RESULTS: Blood vessel endothelial cells of the control group were intact. The basement membrane was shown as a continuous line. Granules of lanthanum sal did not cross the tight junction of endothelial cells. The width of the junction gap in rats of the experimental group treated at different times was increased and lanthanum particles of high density were seen in the gap junction, particularly on day 3, and were distributed in the area of subendothelium. The MCP-1 mRNA expression in VEGF transfected microvessel EC was significantly higher than that of the control group (1.21 +/- 0.22 vs 0.36 +/- 0.06, P < 0.05). CONCLUSION: In BLM induced PF, the opening of the tight junction of EC, and the high expression of MCP-1 induce inflammatory cell infiltration and cytokine over-secretion, which in turn enhances proliferation of fibroblasts, one of the important factors underlying lung fibrosis.

[Isolation, culture and identification of rat retinal progenitor cells in vitro].

OBJECTIVE: To characterize embryonic rat retinal progenitor cells (RPCs) by flow cytometry (FACS), immunofluorescence and (3)H-Thymidine assay in vitro. METHODS: RPCs were prepared from the retina of embryonic day 19 Sprague Dawley rats and were cultured in DMEM: F12 medium with N2-supplement, epidermal growth factor (EGF), basic fibroblast growth factor (bFGF) and leukemia inhibitory factor (LIF) for 0 to 4 days. Cell proliferation and cluster formation were quantified by (3)H-Thymidine assay and morphometric analysis, respectively. Antibodies used to characterize the RPCs included markers for neural progenitors (Nestin and Sox-2), astrocytes (glial fibrillary acidic protein [GFAP]), horizontal cells (Calretinin), ganglion cells (CD90), rods (Rhodopsin), biopolar cells (PKC) and amacrine cells (Syntaxin) by the use of FACS. RESULTS: In primary cells, 22.23%, 16.04% and 19.66% of cells were found to express Nestin, Sox-2 and Calretinin, respectively. GFAP, CD90, Rhodopsin, PKC, Syntaxin were not detected or trail. Meanwhile, more than 90% cells were Nestin positive by immunofluorescence. After 6 days culture in vitro, 43.36% cells express Nestin by FACS. Nestin and GFAP were expressed by use of immunofluorescence. There was significant proliferation of RPCs within the first 4 days as evaluated by (3)H-Thymidine assay. CONCLUSIONS: The results demonstrate that combine FACS, immunofluorescence and (3)H-Thymidine assay together could characterize RPCs. RPCs proliferate well at the early period of cell culture.

[Transfer of endostatin gene for inhibition of retinal angiogenesis in mice].

OBJECTIVE: To evaluate the effect of liposome mediated plasmids encoding endostatin (ES) injected into the vitreous to inhibit experimental retinal neovascularization. METHODS: Cationic liposome mediated ES expression plasmid PCDNA(3)-ES was constructed. One-week-old C57Bl/6N mice were exposed to (75 +/- 2)% oxygen for 5 days, then returned to the room air to induce retinal neovascularization. Cationic liposome mediated ES complex (2 microl) was injected into the vitreous in the treatment group. PBS 2 microl or liposome with carrier DNA complex were injected in the control group. The ES protein expression in the retina was tested with immunohistological methods at 1, 3, 7 and 14 days after injection. Retinal neovascularization was evaluated by angiography with injection of fluorescein dextran and quantification of neovascular proliferative retinopathy after 5 days in room air. To examine the toxicity of the liposome and plasmid PCDNA(3)-ES complex, the histological changes in the retina were examined by light and electron microscopy. RESULTS: ES protein was expressed in the retina 24 hours after injection. Most of them presented in the retinal ganglion layer. This could last for 2 weeks at least. Retina of the PBS-injected eyes of retinal neovascular animal model showed prominent neovascular tuft and fluorescein leakage. Fewer neovascular tufts could be seen after ES injection. Retinal neovascularization in the eyes injected with ES plasmids complex was reduced as compared with the control group. No side effect on the retina was observed by light and electron microscopy. CONCLUSIONS: Liposome mediated plasmids encoding ES can be transferred to retinal cells by vitreous injection and can suppress retinal neovascularization, no side or toxic effects are presented in the retina. Further studies should be done to improve the treatment results.

[Experimental study on repairing damage of corneal surface by mesenchymal stem cells transplantation].

OBJECTIVE: To investigate the survival, migration and differentiation of the cultured human mesenchymal stem cells derived from bone marrow (hMSCs) after transplanted onto the alkaline burn rabbit cornea. METHODS: Alkaline burn on rabbit corneas was induced with NaOH solution. One month later, hMSCs cultured with a feeding of amniotic membrane were transplanted onto the surface of alkaline burn rabbit corneas. Amniotic membrane alone transplantation was used at the same time as control group. One month after transplantation, the changes of corneal morphology were analyzed by clinical observations and HE staining. Immunohistochemistry was carried out with the antibodies against human nuclei and cytokeratin 12 to investigate the distribution and differentiation of hMSCs. RESULTS: The rabbit cornea became totally opaque one month after alkaline burn. Blood vessels could be seen within the superficial layer and stroma. The surface of cornea was rough and dry. There were many goblet cells found in the corneal epithelial layer. One month after transplantation of hMSCs, the surface of alkaline burn cornea became smoother. The amount of the new blood vessels of the cornea reduced and the goblet cells disappeared. Anti-human nuclei antibody staining showed positive cells on the surface layer and superficial stroma while cytokeratin 12 positive cells were only present in the epithelium layer. In the amniotic membrane transplantation control group, no clinical improvement of the cornea was found. Goblet cells were still seen on the corneal surface. Both anti-human nuclei antibody and cytokeratin 12 staining were negative. CONCLUSIONS: hMSCs survive and migrate to stroma after transplanted onto the surface of alkaline burn rabbit cornea. No rejection is detected. hMSCs on the corneal surface differentiate to corneal epithelium, where those migrated to the stroma differentiate to cells other than epithelium. hMSCs transplantation decrease corneal conjunctivization after alkaline burn.

[Cell penetrating peptides enhance intracellular translocation and function of siRNA encapsulated in Pegylated liposomes].

AIM: To prepare the PEGylated liposomes modified with cell penetrating peptides, which protect siRNA from nuclease degradation and deliver efficiently siRNA into cells to facilitate silencing of target gene. METHODS: The purity of R8-PEG-PE and pNP-PEG-PE was detected by HPLC; the quantity of R8, PEG-DPPE modified R8, and R8 attached to the out membrane surface of the liposomal siRNA by transfer from R8-PEG-DPPE micelles to the liposomes was tested by fluorescence; Size and size distribution of siRNA loaded liposomes with and without attached R8 were determined by Zetasizer 5000; A comparison of mediated siRNA transfection efficiency between R8-liposomes and lipofectamine 2000 was examined by individual inside cell fluorescence intensity; The growth inhibition of small cell lung carcinoma NCI-H446 cells treated with R8-liposomal hdm2-siRNA or lipofectamine 2000-hdm2-siRNA complex was tested by MTT assay. RESULTS: The retention times of PEG-DPPE and R8-PEG-DPPE were 9.0 min and 7.8 min, respectively. Fluorescence scanning indicated that lipids composed of liposomes and siRNAs didn't interfere to the determination of R8 when it was attached to the liposomal siRNA. The cells treated with R8-liposomal hdm2-siRNA significantly enhanced the cellular uptake of hdm2-siRNA and facilitated the functions of hdm2-siRNA through silencing of target gene which, in turn, inhibited tumor cell growth, compared with lipofectamine 2000. CONCLUSION: The R8 attached liposomes are shown to be powerful carriers for delivery siRNAs into cell to silence targeted gene.

[Analysis of the volatile components of YL2000 decoction by GC-MS].

OBJECTIVE: To analyze the volatile components of YL2000 decoction, which contain four herbs: rhizome and root of the Notopterygium incisum, the root of the Angelica pubescens f. biserrata, Scutellaria baicalensis and Coptis chinensis and investigate the changes of volatile constituents from those four herbs before and after compatibility of the herbal medicines. METHOD: The volatile components of YL2000 decoction were extracted by water-steam distillation, separated and identified by GC-MS. The relative percent content of each volatile component was quantified by area normalization method. The volatile components of YL2000 decoction were compared with the composition of the volatile oil from individual herb respectively. RESULT: 39 of the 146 separated constituents in volatile oil of YL2000 decoction, accounting for 85.66%, were identified and quantified. After compatibility of the herbal medicines, most volatile oil components reported by high proportion in individual herb were not detected, in the mean time, some components in volatile oil of YL2000 decoction have never been reported before in those of all four herbs. CONCLUSION: The changes of volatile oil from those four herbs before and after compatibility of the herbal medicines indicate that solubilization, chemical reactions and evaporation of some volatile components during decocting may induce changes of several components.

[RbAp46 gene activates the expression of IGFBP-rP1 gene in K562 leukemic cells].

OBJECTIVE: To explore the mechanism of action of RbAp46 gene on leukemic cells. METHODS: K562 leukemic cells and SHG44 glioma cells were transfected with eukaryotic expression vector carrying full-length cDNA of RbAp46 driven by the cytomegalovirus promoter mediated by lipofectamine transfection reagent. Empty vector were transfected at the same time as control. G418-resistant colonies were selected after 3 weeks culturing. Series genes were amplified using RT-PCR. Growth curve and colony formation assays were performed. RESULTS: The number of K562/RbAp46 and K562/CMV cells were (90.00 +/- 8.40) x 10(4) and (119.58 +/- 9.87) x 10(4), respectively after 4 days growth, and SHG44/RbAp46 and SHG44/CMV cells were (89.13 +/- 4.88) x 10(4) and (149.42 +/- 10.83) x 10(4), respectively after 5 days growth. Seven-day yields of K562/RbAp46 and K562/CMV cell colonies were 131.67 +/- 15.57 and 250.33 +/- 26.31, respectively (P < 0.01), while those of SHG44/RbAp46 and SHG44/CMV cells were 50.78 +/- 6.77 and 206.67 +/- 37.18, respectively (P < 0.01). The fraction of K562/RbAp46 and K562/CMV cells in S phase was (48.88 +/- 4.35)% and (62.78 +/- 4.78)% (P < 0.01), and in G(0)/G(1) phase was (29.10 +/- 4.14)% and (22.40 +/- 2.43)%, respectively (P < 0.05), and that of SHG44/RbAp46 and SHG44/CMV cells in G(0)/G(1) phase was 65.6% and 48.8%, and in S phase was 22.6% and 38.4%, respectively. Insulin-like growth factor binding protein-related protein-1 (IGFBP-rP1) gene was induced to express only in the RbAp46-over expressing K562 cells and was not in SHG44 cells. CONCLUSION: A regulatory pathway between RbAp46 and IGFBP-rP1 genes might exit in K562 leukemic cells.

[Individual 3-D image prototyping models for orbital reconstruction].

OBJECTIVE: To fabricate a rapid prototyping (RP) 3-D image models for individual reconstruction of orbital bony loss. METHODS: The skull was placed on a helical CT scanner table and the Frankfort plane was perpendicular to the table. The CT data was obtained by a Aquilion (TOSHIBA, Japan) with 1 mm thickness section in spiral mode. By adjusting of CT threshold value and pixels in order to stack the segmental defects, we obtained an approximate 3-dimension visual model of the scanned skull using MedGraphics software. An orbital RP model based on the dataset of the 3-dimension visual model was fabricated by RP machine. Both 3-dimension visual model and RP model were measured against the skull with several anatomic landmarks to examine the accuracy of the models, and the errors were analysed. RESULTS: Integrity precision rapid RP models of the orbital region were reconstructed. The anterior orbital rim, middle orbital section and posterior orbital section were all fabricated. Optic foramen, superior orbital fissure, infraorbital foramen, inferior orbital fissure, lacrimal sac socket and naso-lacrimal duct were shown clearly. But some fine hole and slot, such as the anterior ethmoidal foramen, posterior ethmoidal foramen and zygomaticofrontal suture were not obviously seen. The mean difference between the 3-dimension visual model and the skull was 0.10 +/- 1.02mm. For the RP and dry skull, the mean difference was 0.22 +/- 1.04mm. There were no statistical differences between them. CONCLUSIONS: Integrity precision orbital RP models were fabricated which fulfilled the requirements of the individual reconstruction with bony orbital pathologic changes. The keys to fabricate the precision orbital RP models included a closer cooperation between the surgeon and engineer, thin CT slice in 1mm thick and an appropriated threshold value. Better results for the orbital deformities should be achieved for the contour of orbital region or eye function.

[A case-control study of risk factors in mild cognitive impairment].

OBJECTIVE: To explore the risk factors of mild cognitive impairment (MCI). METHODS: A case-control study including 97 MCI patients and 143 cognitive normal controls (NC) in Chinese PLA General Hospital was conducted. The cases and controls were matched for age, gender and occupation. The relationship between MCI and various factors was analysed by univariate and multivariate analysis. RESULTS: Results data from univariate analysis showed that the history of coronary heart diseases, stroke, anaemia, and the education level had significant differences between NC and MCI patients. Multivariate analysis confirmed that the history of coronary heart diseases, stroke, anaemia and the education level were significantly related to MCI and their ORs (95% confidence interval) were 2.21 (1.18 - 4.14), 2.18 (1.20 - 3.98), 4.63 (1.79 - 11.97), 0.75 (0.58 - 0.97) respectively. CONCLUSION: The history of coronary heart diseases, stroke and anaemia were independent risk factors of MCI and high education level seemed to be a protective factor of MCI.

[Study of sexual function in women after surgery for stress urinary incontinence].

OBJECTIVE: To investigate the change of the patients' sexual function after the surgeries for stress urinary incontinence (SUI). METHODS: Pelvic organ prolapse-urinary incontinence sexual function questionnaire (PISQ) was sent to 62 sexually active patients who underwent laparoscopic Burch colposuspension or had a tension free vaginal tape (TVT) inserted at least 6 months after the operation, to a maximum of 3 years. The patient was divided into two groups according to different operations, different follow-up intervals and whether there was concomitant coital incontinence before the operations respectively. The changes of PISQ scores before and after the operation were compared between two groups. For the groups not having significant difference in PISQ score, we compared the rate of PISQ score improvement and the rate of satisfaction postoperatively. Multivariate logistic regression analyses were performed to determine whether age, parity, body mass index, menstrual status, surgical history, medical illness, severity of SUI, concomitant hysterectomy and intraoperative or postoperative complications affected the change of sexual function. RESULTS: Fifty-six patients answered the questionnaire giving a follow-up rate of 90%. The average score of PISQ was 85.2 +/- 10.7 and 82.4 +/- 17.7 before and after the surgery, and there was no significant difference (P > 0.05); however behavioral emotional domain scores worsened (30.5 +/- 6.6 vs 26.8 +/- 10.7, P < 0.05). The PISQ scores of the patients who underwent the operation within a year declined significantly (86.0 +/- 10.7 vs 77.7 +/- 18.6, P < 0.05), especially in behavioral emotional domain (31.0 +/- 7.2 vs 23.7 +/- 11.3, P < 0.01); however after a year, there was no significant change (84.4 +/- 10.8 vs 86.5 +/- 16.1, P > 0.05) and even an improvement in the physical domain (34.5 +/- 5.6 vs 36.9 +/- 4.3, P < 0.05). Regarding the sexual function improvement ratio, the patients who had the TVT operations (54% vs 12%) or had concomitant coital incontinence before the operations (62% vs 23%) were better (P < 0.01). The change of PISQ scores was independent of clinical features. CONCLUSIONS: The operations of SUI have an adverse effect on the patients' sexual life within a year, which may recover after that period. TVT operation has a better effect on the patients sexual function compared with laparoscopic Burch colposuspension; the patients who have concomitant coital incontinence before the operations have a better sexual function improvement.

[Sequence analysis of a novel HLA-A*3308 allele].

OBJECTIVE: To investigate the molecular genetics basis for HLA novel allele HLA-A*3308 in Chinese population. METHODS: DNA was extracted from whole blood by PEL-FREEZ DNA extraction kit. The amplification of HLA-A exons 1-8 of the proband was preformed by PCR and the amplified product was cloned with TOPO cloning sequencing kit to split the two alleles apart. Both strands of exons 2, 3 and 4 of chosen clones were sequenced. The PCR-SSP was performed to confirm the mutations detected by sequencing. RESULTS: The sequencing results showed HLA-A alleles of the proband as A*0201 and the novel allele. The sequences of the novel allele have been submitted to GenBank (DQ089631, DQ089632, DQ089633). BLAST analysis showed that the novel allele got the difference from A*3303 by five nucleotides at positions 240 A>T, 256C>G, 259A>G, 261C>G and 270T>A in exon 2. This resulted in three amino acids changes from Arg to Gly at codon 62, Asn to Glu at codon 63 and Asn to Lys at codon 66. CONCLUSION: This allele is a novel allele and has been officially named A*3308 by the WHO Nomenclature Committee.

[Expression and diagnostic application of C4.4A protein in squamous cell carcinoma and adenocarcinoma].

OBJECTIVE: To investigate the diagnostic utility of C4.4A gene expression in discriminating a squamous cell carcinoma (SCC) from an adenocarcinoma by immunohistochemistry. METHODS: Immunohistochemical staining was performed to detect the expression of C4.4A protein in 157 cases of SCC and 177 cases of adenocarcinoma of various organs. RESULTS: Overall, 141 of 157 cases of SCC strongly expressed C4.4A protein. In contrast, only 8 of 177 adenocarcinomas showed partial or scattered cell expression of C4.4A protein. The statistic difference between the two groups was highly significant (chi(2) = 244.93, P = 0.000), and also when the tumors were stratified according to the degree of differentiation (P = 0.000). CONCLUSION: C4.4A protein expression may serve as a valuable tumor marker in discriminating a squamous cell carcinoma from an adenocarcinoma, and therefore, may greatly facilitate the differential diagnosis of an epithelial malignancy.

[The analysis of risk factors correlated to pulmonary hypertension in obstructive sleep apnea syndrome patients during awake state].

OBJECTIVE: To investigate the development of pulmonary hypertension in obstructive sleep apnea syndrome (OSAS) patients and to analyze the correlated factors. METHODS: Pulmonary arterial pressure was monitored by right cardiac catheterization in 15 OSAS patients, and simultaneously polysomnography was performed. Blood gas analysis and lung function were also measured. RESULTS: Pulmonary arterial pressure at awake state was correlated positively to mean maximal pulmonary pressure during sleep, body mass index (BMI) and hemoglobin (Hb), but negatively to PaO2, the percent predicted forced vital capacity (FVC% pred). Compared with OSAS patients without pulmonary hypertension, the BMI, PaCO2, and Hb of OSAS patients with pulmonary hypertension increased significantly, while FVC% pred and PaO2 decreased. Stepwise linear regression indicated that pulmonary arterial pressure at awake state was closely correlated with mean maximal pressure during sleep (beta = 0.35, standard error 0.10, R(2) = 0.89, P = 0.006) and PaCO2 (beta = 0.72, standard error 0.27, R(2) = 0.94, P = 0.022), and mean maximal pulmonary arterial pressure during sleep was closely correlated to PaCO2, BMI, PaO2 and the ratio of arterial pressure and oxygen concentration during rapid eye movement sleep (RDeltaPAP/DeltaSpO2). The regression equation was y' = -152.70 + 1.92 PaCO2 + 1.37 BMI + 0.67 PaO2 + 16.29 RDeltaPAP/DeltaSpO2. CONCLUSION: Pulmonary arterial pressure increasing in OSAS patients is induced mainly by hypercapnia and hypoxia at day time, and related to forced ventilation capacity, BMI and the ratio of pulmonary arterial pressure and oxygen concentration variation during rapid eye movement sleep. There was no obvious relation between pulmonary arterial pressure and apnea index.

[Complete sequence and gene organization of the Tibetan chicken mitochondrial genome].

Using PCR amplification, sequencing and assembling, we obtained the complete mitochondrial genome of Tibetan chicken. The complete mitochondrial genome was 16 783 bp in length. It contained 37 genes (13 protein coding genes, 2 rRNA, 22 tRNA) and a control region. The deduced restriction map revealed a unique pattern of Dra I restriction in Tibetan chicken. Phylogenetic trees based on the D-loop locus and the 13 protein coding genes by Neighbor-joining and Maximum Parsimony analysis indicated that the red junglefowl was the direct ancestor of Tibetan chicken and Tibetan chicken was closest to white leghorn and white plymouth rock, although the evolution of Tibetan chicken appeared to be relatively independent from them. A possible explanation is that the ancestor of Tibetan chicken lived in a relatively isolated environment after entering into the high altitude area and developed unique genetic characters.
