Designing polymer matrix for microchip-based double-stranded DNA capillary electrophoresis.

Polyacrylamide (PAM) was used as a model polymer to build up an empirical model that relates polymer molecular weight, polymer concentration and solution viscosity. The desired random copolymers of acrylamide (AM) and N,N-dimethylacrylamide (DMA) used as DNA separation media for different specifications were synthesized under the guidance of the empirical model. The separation performances of rationally designed copolymers were tested in a 1.2 cm long separation channel, simulating microchip-based capillary electrophoresis. pBR322/HaeIII digest was successfully separated with good separation resolution and fast speed. Validation of the sieving ability of our polymers was performed in the Agilent 2,100 Bioanalyzer. The results of the 10 bp (base pair) DNA ladder separation demonstrate the potential of our approach for the sieving matrix in microchip-based electrophoresis.

Enhanced photo-catalytic activity of TiO2 films with doped La prepared by micro-plasma oxidation method.

Mesoporous thin titanium dioxide films have been prepared on titanium plates through micro-plasma oxidation. To increase the films' photo-catalytic activity, La ions with different concentrations (0, 0.025, 0.005, 0.075, 0.1g/L) were added into the H(2)SO(4) electrolyte solution. X-ray diffraction, X-ray photoelectron spectroscopy, atomic force microscopy, and scanning electron microscopy techniques were applied to characterize the modified films. A kind of typical textile industry pollutant (Rhodamine B) was used to evaluate the photo-catalytic activity of the films. The results showed that this activity of the films had been improved by adding La ions into the electrolyte solution. The enhanced photo-catalytic activities might be resulted from the increase of mesopores' number, producing more reactive sites to absorb and oxidize pollutants. Also, the improvement was related to the forming of titanium dioxide lattice distortion, which could accept more photoexcitated holes and produce more strong surface free radicals to oxidate adsorptive molecules.

Reversible thermo-responsive sieving matrix for oligonucleotide separation.

A reversible thermo-responsive gel system, consisting of Pluronic copolymer mixture of F87 and F127, has been used to successfully carry out the separation of oligonucleotides, for the first time, by microchip-based capillary electrophoresis. Pluronic triblock copolymers F87 (E(61)P(40)E(61)) and F127 (E(99)P(69)E(99)), with E, P, and subscript denoting oxyethylene, oxypropylene, and segment length respectively, have a unique temperature dependent viscosity-adjustable property and a dynamic coating ability in aqueous solution, including 1 x TBE buffer. The mixture solution has a reversible thermo-responsive property and its sol-gel transition temperature can be adjusted ranging from about 17 degrees C to 38 degrees C by varying the relative weight ratio of F87 and F127 at an optimized concentration of approximately 30% (w/v) for oligonucleotide separations. Oligonucleotide sizing markers ranging from 8 to 32 base could be successfully separated in a 1.5 cm long separation channel by the mixture solution in its gel-like state. A 30% (w/v) with a F87/F127 weight ratio of 1 ratio 2 which has a "sol-gel" transition point of about 26 degrees C shows the best sieving ability. The sieving ability of the mixture solution was further confirmed in an Agilent Bioanalyzer 2100 system. Fast separation of oligonucleotides has been achieved within 40 s with one base resolution.

Scale-up development of high-performance polymer matrix for DNA sequencing analysis.

Linear polyacrylamide (LPA) has been widely used as a replaceable separation matrix in CE. An increase in the molecular weight of the separation medium favors the separation of larger DNA fragments. In order to obtain ultrahigh-molecular-weight (UHMW) LPA, a "frozen" method was developed to synthesize the LPA homopolymer. This approach has three major advantages when compared with other existing routes of LPA synthesis: (i) long LPA chains could be obtained easily, with their average molecular weight (MW) being in the high 10 MDa range; (ii) the desired MW could be adjusted over a broad range by controlling the temperature and the concentration of initiators during synthesis; (iii) the product solution contains only a tiny amount of impurity besides the solvent and LPA. Both static and dynamic laser light scattering measurements were carried out to characterize the synthesized LPA in the buffer solution. The DNA sequencing matrix prepared from LPA using this method was studied and the results were compared with the newly developed commercial product POP7 from Applied Biosystems. It should be noted that this approach can be applied to synthesize other water-soluble polymers, resulting in UHMW products because the chain transfer constant is smaller at lower temperatures.

Natural and synthetic cage compounds incorporating the 2,9,10-trioxatricyclo[4.3.1.03,8]decane type moiety.

The 2,9,10-trioxatricyclo[4.3.1.0(3,8)]decane moiety is a tetracyclic cage-like orthoester incorporated in the structure of a series of daphnane derivatives such as resiniferatoxin (RTX), kirkinine, synaptolepis factors, huratoxin etc., exhibiting various biological activities. The approaches to the preparation of cage-like orthoesters starting from partially acylated or unmodified 1,2,4-trihydroxycyclohexane moieties built onto natural as well as synthetic compounds are discussed. Orthoester derivatives of RTX analogs, Ceverathrum alkaloids, myoinositol and pyranoses are included. Stereochemical requirements to the formation of the orthoester unit are discussed. The biological activity of different compounds containing the cage-like orthoester structural fragment is given. The literature is reviewed till 2004.

Fast separation of single-stranded oligonucleotides by capillary electrophoresis using OliGreen as fluorescence inducing agent.

The fast separation of oligonucleotide (oligos) sizing marker by CE using OliGreen and including effects due to the concentration of separation medium and urea denaturant is presented. OliGreen dye is found to be more sensitive than ethidium bromide (by a factor of about 6 based on S/N considerations) for the oligos' separations. Higher concentration of F127 in 1xTris-boricacid-EDTA (TBE) up to 30% w/v leads to better resolution of oligos separations. The addition of urea into the separation medium decreases the sensitivity. With an optimized running condition, the oligos sizing marker could be successfully separated with 1-base resolution within 1.3 min by using 30% w/v F127/1xTBE solution as the separation medium at an applied electric field of 800 V/cm in a 3 cm long capillary, the fastest capillary gel electrophoresis separation with high resolution reported to date for oligos in the similar size range.

Beta-adenosine, a bioactive compound in grass chaff stimulating mushroom production.

Fructification and yield of the edible mushrooms Pleurotus pulmonarius and Stropharia rugosoannulata are clearly enhanced when wheat straw is supplemented with 30% Lolium perenne grass chaff. The bioactive compound in the methanol extract of grass chaff was identified as beta-adenosine. In vitro biological activity tests showed that 0.012 mg of beta-adenosine per ml of medium stimulated earlier fructification of Pleurotus pulmonarius. Mushroom fruiting trials showed that when 12 mg beta-adenosine was added to 1 kg wet wheat straw, primordia of Pleurotus pulmonarius appeared two days earlier and primordia of Stropharia rugosoannulata appeared 18 days earlier when compared to pure wheat straw substrate. This concentration of beta-adenosine had no impact on the mushroom yield of Pleurotus, but resulted in a 2.2 fold increase in yield for Stropharia. beta-Adenosine at 25 mg per kg wet wheat straw increased the yield of Pleurotus with 52% and the yield of Stropharia with 258%, but this concentration delayed primordial formation in Pleurotus.

Antitrichomonas in vitro activity of Cussonia holstii Engl.

Cussonia holstii is a traditional medicinal plant used in Kenya, which has not been studied chemically up-to-date. Upon biological screening, a substantial antitrichomonas activity was found in the dichloromethane extract of the bark of Cussonia holstii. A detailed chromatographic study using centrifugal partition chromatography revealed that a pentacyclic triterpenoid, i.e., hederagenin, is the antitrichomonas constituent of the plant with a IC50 of 2.8 microM.

Formation of monodisperse polyacrylamide particles by radiation-induced dispersion polymerization: particle size and size distribution.

Polyacrylamide microparticles were directly produced by radiation-induced dispersion polymerization in aqueous alcohol media using poly(N-vinylpyrrolidone) as a steric stabilizer at room temperature. The hydrodynamic diameter of a polymer particle and its distribution were measured on a dynamic laser light-scattering spectrometer. This method takes advantages of the specialties of radiation induction, and highly uniform polymer microspheres were obtained with high conversion. The number of the particle produced in the early stage of the polymerization was found to be constant during the remainder of the polymerization. The effects of various polymerization parameters, such as absorbed dose rate, monomer concentration, stabilizer content, medium polarity, and polymerization temperature on the particle size and size distribution were systematically investigated.

Chemical constituents and biological activities of Zanthoxylum usambarense.

Bioassay-guided fractionation of the dichloromethane extracts of the roots and the bark of Zanthoxylum usambarense led to the isolation of two physiologically active compounds, i.e. canthin-6-one 1 (fungicide) and pellitorine 4 (insecticide). Together with oxychelerythrine 2, norchelerythrine 3, (+)-sesamin 5 and (+)-piperitol-3,3-dimethylallyl ether 6, they were isolated for the first time from this plant.

Neurotrophic and antileukemic daphnane diterpenoids from Synaptolepis kirkii.

Biological assay guided fractionation of a dichloromethane extract of Synaptolepis kirkii led to the isolation of four new and five known daphnane-type diterpene orthoesters, whose structure was established by spectroscopic data. Full spectroscopic data of the new and known natural products are reported here for the first time. Pronounced neurotrophic and substantial antileukaemia activities of these compounds were found in in vitro assays.

Daphnane-type diterpene orthoesters and their biological activities.

Daphnane orthoesters are the active ingredients of plant remedies from the Western, Chinese and African traditional medicine, and have provided important tools to investigate medicinally relevant processes like tumour promotion, apoptosis, neurotrophism, and VR1 activation. The occurrence, biological activity, and molecular pharmacology of these compounds will be reviewed.
