Extraction and protein component analysis of venom from the dissected venom glands of Latrodectus tredecimguttatus.

Black widow spiders (genus Latrodectus) have attracted increasing attention due to frequently reported human injuries caused by them and the potential applications of biologically active components in their venoms. Although a number of studies have described the biological properties and structures of several venomous proteins such as latrotoxins, a comprehensive analysis of protein component of the venom from the spider is not available. We used combinative proteomic strategies to assess the protein components of the crude venom collected from Latrodectus tredecimguttatus by extracting the dissected venom glands. The experiments demonstrated that the crude venom of L. tredecimguttatus has a high abundance of acidic proteins with molecular masses greater than 15 kDa, and the content of proteins and peptides of below 15 kDa is low. 86 unique proteins were identified, part of which were contaminations of cellular components during the extraction, determined in comparison with venom obtained by electrostimulation. Except for members of latrotoxin family that were commonly considered as the primary toxic components of the venom, several other special enzymes and proteins were detected such as protease, phosphatase, lysozyme, inhibitory protein, and so on. These protein components, particularly the proteases, were speculated to play important roles in the action of L. tredecimguttatus venom.

A novel mitochondrial DNA missense mutation at G3421A in a family with maternally inherited diabetes and deafness.

OBJECTIVE: Mutations in mtDNA are thought to be responsible for the pathogenesis of maternally inherited diabetes. Here, we report a family with maternally inherited diabetes and deafness whose members did not harbour the mtDNA A3243G mutation, the most frequent point mutation in mitochondrial diabetic patients. This study aimed to investigate a possible other mtDNA mutation and its prevalence in type 2 diabetic patients. METHODS: Height, body weight, waistline, and hip circumference were measured and serum biochemical marks determined in all members of the family. In addition, a 75 g oral glucose tolerance test and electric listening test were conducted in these members. Genomic DNA was prepared from peripheral leukocytes. Direct sequencing of PCR products was used to detect the mtDNA mutation in this family. The prevalence of mtDNA G3421A nucleotide substitutions was investigated by restriction fragment length polymorphism analysis in 1350 unrelated type 2 diabetic patients recruited by random cluster sampling from the central city area of Shanghai, China. RESULTS: (1) A new missense homoplasmic mutation of mtDNA G3421A was found in a maternally inherited diabetic family and existed neither in 1350 unrelated type 2 diabetic patients nor in 50 non-diabetic individuals. (2) The mode of mutation and diabetes transmission was typical maternal inheritance in this family. (3) All diabetic family members were found to have an onset at 35-42 years of age, accompanied by deafness of varying degrees. CONCLUSION: mtDNA G3421A (Val39Ile) found in a family with maternally inherited diabetes and deafness is a novel missense mutation. Whether this is a diabetogenic mutation and its effect on mitochondrial function needs to be further studied.

High prevalence of albuminuria in population-based patients diagnosed with type 2 diabetes in the Shanghai downtown.

OBJECTIVE: The prevalence of albuminuria and the risk factors associated with albuminuria were evaluated among the Chinese patients diagnosed with type 2 diabetes aged over 30 in the Shanghai downtown. We also evaluated the variability of urinary albumin-to-creatinine ratio (ACR) among the three measurements and the relationship between diabetic retinopathy (DR) and albuminuria. METHODS: The 1039 Chinese patients diagnosed with type 2 diabetes aged over 30 were investigated by randomized cluster sampling in the Shanghai downtown and 1018 patients were analyzed in this study. Body mass measurements including height, weight, waist circumference and hip circumference, resting blood pressure, fasting blood measures, urinary ACR and the digitally stored fundus images were investigated. The prevalence of albuminuria was calculated and the risk factors associated with albuminuria were evaluated by stepwise logistic regression. The concordance of urinary ACR was evaluated by observed agreement. The relationship between albuminuria and DR was also evaluated. RESULTS: (1) The mean age of all patients was 66.10+/-11.54 years and the duration of diabetes was 7.89+/-7.16 years. (2) The prevalence of albuminuria was 49.6% among the Chinese patients diagnosed with type 2 diabetes aged over 30 in the Shanghai downtown, 41.4% with microalbuminuria and 8.2% with macroalbuminuria. (3) Microalbuminuria was significantly associated with systolic blood pressure, gender and waist circumference. Macroalbuminuria was significantly associated with systolic blood pressure and duration of diabetes. (4) Observed agreement among the three urinary ACR measurement for albuminuria staging was 73.3% (first versus second), 64.5% (first versus third) and 77.5% (second versus third). Observed agreement in the albuminuria staging between the single urinary ACR measurement and all three urinary ACR measurements was 85.8% (first versus all three), 87.6% (second versus all three) and 81.9% (third versus all three). (5) The percentage of DR in the macroalbuminuric group (59.2%) was significantly higher than that in the normalbuminuria group (16.1%) and microalbuminuria group (24.6%). (6) The macroalbuminuric patients with DR had significantly increased fasting blood glucose and HbA1c compared with the macroalbuminuric patients without DR. CONCLUSION: The prevalence of microalbuminuria observed in the Chinese patients diagnosed with type 2 diabetes aged over 30 in the Shanghai downtown reached up to 41.4% though the observations in our study might be representative of the diabetic patients of the Shanghai downtown. We agreed that at least two of the three urinary collections were done in a 3- to 6-month period because of the day-to-day variability in albumin excretion. The percentage of DR among the patients with macroalbuminuria was 59.2%, and the macroalbuminuric patients with the significantly high plasma glucose and DR were prone to diagnose DN.

A dispersed-ozone flotation (DOF) separator for tertiary wastewater treatment.

A dispersed-ozone flotation (DOF) separator was devised for a pilot study of tertiary wastewater treatment for re-use purposes. As a compact device combining coagulation, ozonation and flotation in an integrated unit, the DOF separator achieved a very high removal of SS, TOC, UV254 and colour, as well as effective inactivation of coliform and total bacteria within a short hydraulic retention time of 30 min. The finished water quality is comparable to or better than that by a conventional tertiary treatment process using coagulation, sedimentation, filtration and chlorine disinfection, and meets the quality standards for non-drinkable domestic reuse.

Safety of treated water for re-use purposes--comparison of filtration and disinfection processes.

A study was conducted on the distribution of pollutants in treated wastewater and the its safety for re-use purposes. Based on the results of a series of tertiary treatment experiments, the effects of three filtration processes, i.e. coagulation-filtration, ozonation-biological activated carbon filtration (O3-BAC) and ultrafiltration (UF), and two chemical disinfection processes, i.e. chlorination and ozonation, on the safety of water re-use were evaluated. It was found that the concentrations of the main pollutants in the secondary effluent and further filtered water follow a log-normal distribution and, therefore, a log-normal probabilistic function can be used to evaluate the suitability of the treated water for re-use purposes. Among the three filtration processes evaluated, UF is the most effective in turbidity removal but less effective in colour and COD removal, while coagulation-filtration and O3-BAC can ensure a good removal of all these pollutants. Regarding chemical disinfection, although chlorine is very effective in inactivation of coliform bacteria, it can not achieve a substantial decrease in viruses. As ozone is applied, effective virus removal can be achieved.

Water shortage and needs for wastewater re-use in the north China.

This paper analyses the present condition of the water shortage in north China where annual rainfall is low and per capita water resource is below the line of regular water stress, or even the line of absolute water scarcity. Of the available water resources, the percentge of water withdrawal in all the north basins is high--the Yellow River and Huai River basins being greater than 80% and the Hai River basin mainly depending on interbasin water transfer. Over-withdrawal of water also results in serious water environmental problems including "flow cut-off" of the Yellow River main channel and water pollution of many rivers. The paper also analyses the potential of wastewater as a resource and the demand for treated wastewater re-use. In north China, due to low rainfall and high potential evaporation environmental re-use, gardening, afforestation, etc. is considered as the main usage of the treated wastewater. Considering the economic restrictions in the less developed area, a decentralised system can be taken as an important option in formulating water re-use strategies.

Loss of heterozygosity at 6p21.3 underlying HLA class I downregulation in gastric cancer.

Human major histocompatibility complex (HLA), located on 6p21.3, encodes for genes involved in antigen processing and presentation. Loss of heterozygosity (LOH) at 6p21.3 may cause downregulated expression of HLA, thus provide tumor cells with an immune-escape tumor phenotype. In the present study, we detected HLA class I expression in gastric cancer and correlated it with LOH at 6p21.3. The expression of HLA class I antigen was analyzed by immunohistochemical procedure in 50 fresh surgically removed gastric cancers and corresponding normal tissues using 5 monoclonal antibodies (mAbs). LOH studies were detected by using 6 microsatellite markers located at 6p21.3, 1 marker located at 6q21 and 2 microsatellite markers flanking the beta2m gene. HLA class I complex was obviously downregulated in gastric cancer compared with corresponding normal tissues (t=4.00, p<0.01). The expression of beta2m gene was also downregulated in tumor, but not concordant with HLA class I surface downregulation. Out of the 50 tumors, 25 (50%) showed LOH of at least one STR marker at 6p21.3 while only 11 (22%) showed LOH of the two markers flanking the beta2m gene. The LOH frequency of D6S105, which is close to HLA-A gene, was the highest in all STR markers studied (34%). Downregulation of HLA class I expression was correlated with loss of chromosomal regions at 6p21.3 (chi-squared = 5.13, p<0.05). Our results suggested that LOH of 6p21.3 contributed to HLA class I downregulated expression in gastric cancer especially at HLA-A locus. LOH of HLA-A gene might be one of the mechanisms underlying the abnormal expression of HLA class I complex.

Application of a virological tracer method for the assessment of pathogen removal by physicochemical treatment and chemical disinfection.

Coxsackie B3 (CoxB3) virus was used as a virological tracer for an assessment of the efficiency of pathogen removal by several typical physicochemical treatment and chemical disinfection processes, such as coagulation-filtration, ultra-filtration, and disinfection using chlorine and ozone, with regard to the pathogenic quality of the treated domestic wastewater for reuse purposes. The CoxB3 virus was seeded to sterilized secondary effluent to make a raw water of known pathogenic level. After applying the raw water to each treatment or disinfection process, the residual virus in the finished water was concentrated, and virus assay was carried out by the Tissue Culture Infectious Dose technique. TCID50 was used as an indicative parameter of CoxB3 virus in the raw and treated water. Parallel experiments were also conducted to evaluate the effectiveness of each process for the removal of coliform bacteria. It was noticed from the experiment that both coagulation-filtration and ultrafiltration could achieve substantial removal of TCID50 at about the same level (2-log removal). However, the effect of the two processes on the removal of coliform bacteria was much different: 2-log removal by coagulation-filtration and 4 to 5-log removal by ultrafiltration. The TCID50 removal correlates more closely with the removal of turbidity than that of coliform bacteria. Chlorine was found to be effective in coliform removal but almost had no effect on TCID50. As ozone was applied, a high removal of both coliform bacteria and TCID50 could be obtained.

Pilot study of a fluidized-pellet-bed technique for simultaneous solid/liquid separation and sludge thickening in a sewage treatment plant.

A fluidized-pellet-bed separator with movable sludge hoppers was applied in pilot scale for the separation and thickening of activated sludge mixture liquid. Under the condition of suspension SS around 4,000 mg/L, polymer (CJX103, cationic, MW 5 x 10(6)) dose at a dry solid ratio of 0.003 and upward flow rate at 5.4 m/hr, the fluidized pellet bed performed solid/liquid separation and sludge thickening well. The SS concentration of the treated water was about 5 mg/L on average and the moisture content of the sludge after screening for 5 min was less than 94%, which is much lower than that after conventional settling and thickening and easy to be finally disposed. At a higher upward flow rate of 7.2 m/hr, similar results could also be obtained but higher polymer dose (solid ratio of 0.004) was required. The morphological characteristics and density-size relationship of the granular particles formed in the fluidized pellet bed were also investigated by image analysis and settling velocity measurement of individual particles. The two-dimensional fractal dimension was evaluated to be 1.6-1.8, showing a good quasi-spherical morphology of the granular particles with their density much higher than the conventional flocs. The results of the pilot study indicate a possible way to innovate the conventional secondary settling and gravitational thickening processes for solid/liquid separation and sludge handling, especially for small scale wastewater treatment plants to reach the goal of space saving and higher treatment efficiency.

Organo-chlorine pesticide (DDT and HCH) residues in the Taihu Lake Region and its movement in soil-water system I. Field survey of DDT and HCH residues in ecosystem of the region.

The use of organo-chlorine (DDT and HCH) has been banned in China for 20 years. A field survey was carried out during 1999-2000 in the Taihu Lake Region. Organo-chlorine pesticide (OCP) residues in soil, water, fish and sediment samples were investigated. DDT was detected in 5 out of 10 samples with concentration ranging from 0.3 to 5.3 microg/kg in the surface (0-15 cm) layer, 6 out of 10 with 0.5 to 4.0 microg/kg in the subsoil layer (16-30 cm), and 4 of 10 with 0 to 2.7 microg/ kg in the deep soil layer (31-50 cm). Results for HCH residues in soil samples were similar to those of DDT. These results indicate that OCP residues in 0-50 cm profile had been leached out or degraded to safe level. In river water DDT was detected in 10 out of 13 samples ranging from 0.2 to 9.3 microg/l, with an average of 1.0 microg/l. While HCH was detected in 12 out of 13 samples ranging from 0.02 to 36.1 microg/l, with an average 5.6 microg/l. DDT residues in sediment ranged from 0.1 to 8.8 microg/kg, while HCH ranged from 0.3 to 66.5 microg/kg. DDT residues in fish body ranged from 3.7 to 23.5 microg/kg and HCH ranged from 3.7 to 132 microg/kg. These results demonstrate an accumulation through food chain (from soil-water-sediment-microbes-crop-fish-... etc.), also that HCH residues are generally more persistent than DDT residues. However, all these data are well below than the state warning standard limit.

Status of trace elements in paddy soil and sediment in Taihu Lake region.

Thirteen paddy soil profiles and river sediments which are sources of irrigation water were collected around the Taihu Lake, and the trace elements were estimated. The content of La and Ce in paddy soil and sediment were 39.3 and 68.6 mg/kg soil and 36.9 and 65.1 mg/kg soil, being within the range of background values. The values for Pb, Cu, Ni, Cr, Co, Mn, Zn, Se in paddy soil were 23.3, 27.8, 25.5, 63.5, 10.2, 386, 68.7 and 0.25 mg/kg soil respectively, all below the national permission level. There was a decline of Zn in paddy soil. Some of the river sediments were seriously polluted. The river in Yangjin site was most contaminated with 5.47 g Cu/kg and 7.4 g Zn/kg. The high concentration of Pb and Ni also was observed in this sediment. River in Weitang, Huashi, Xinzhuang and Meiyan were contaminated with Pb, Cu and Ni to some extent. Zn, Cu and Pb were the main pollutants in present experiment sites. The fast development of village/township industries have caused severe environmental pollution in the Taihu Lake region, especially irrigation river sediments. Se content in plant and seed was 0.04 and 0.03 mg/kg respectively, showed Se-deficiency in paddy soil in the Taihu Lake region.

Inhibition of protein phosphatase 2A- and protein phosphatase 1-induced tau hyperphosphorylation and impairment of spatial memory retention in rats.

Tau hyperphosphorylation leads to formation of paired helical filament/neurofibrillary tangles, the hallmark lesion seen in Alzheimer's disease (AD) brain. An imbalanced regulation in protein kinases and protein phosphatases in the affected neurons is proposed to be a reasonable causative factor to the disease process. To verify the hypothesis, we have injected in the present study calyculin A, a potent and specific inhibitor of protein phosphatase (PP) 2A and PP1, into rat hippocampus bilaterally, thus reproduced an Alzheimer's-like deficiency in dephosphorylation system. It was found that calyculin A-injected rats developed lesions in spatial memory retention in Morris water maze test. At mean time, tau was hyperphosphorylated at Ser396/Ser404 (PHF-1) and Ser-262/Ser-356 (12E8) sites determined both by immunohistochemistry and Western blot. It is implicated that (1) PP2A and PP1 participate in the in vivo regulation of tau phosphorylation, and down-regulation of the two phosphatases will result in tau hyperphosphorylation; (2) hyperphosphorylation of tau at PHF-1 and 12E8 sites might be crucial to affect spatial memory in AD.

Subunit interaction slows the unfolding of the N-terminal domain of creatine kinase in urea.

Fluorescence emission intensity changes with two different excitation wavelengths were used to measure the unfolding rate constants of different domains of muscle type creatine kinase (CK-MM) according to the heterogeneity of aromatic amino acid distributions in the crystal structure of CK-MM. The results were compared with those of brain type creatine kinase (CK-BB) and dithio-bis(succinimidyl propionate) cross-linked CK-MM. CK-BB differed greatly in its distribution of aromatic amino acids in each domain and the unfolding process of cross-linked CK-MM was not accompanied by the dissociation of the dimer. The N-terminal domain of CK-MM was shown to be well protected by subunit interaction during the unfolding of CK-MM in 4 M urea. Dissociating the CK dimer in high urea concentration (> or = 6 M) eliminated the subunit protection. Subunit interactions are also important in preserving secondary structure and forming contracted conformation at low urea concentration.

Effect of FGF-1 and FGF-2 on VEGF binding to human umbilical vein endothelial cells.

When FGF-1 or FGF-2 and VEGF were added together, the mitogenic effect of FGF-1 or FGF-2 and VEGF on HUVEC was additive. However, when HUVECs were preincubated for 2 days with 10 ng/ml FGF-1 in the absence of VEGF, the Scatchard plot of [125I]VEGF binding sites was shifted to the right: both affinity classes of VEGF binding sites were equally affected, such that the total number of sites increased twofold. It is suggested that this type of interaction may be related to tumor angiogenesis and wound repair.

Bifunctional effects of heparin-binding protein HBp17 on DNA synthesis in cells.

A 17 kD heparin-binding protein (HBp17) has a biphasic dose-dependent effect on DNA synthesis in 3T3 cells. Maximal stimulation of DNA synthesis occurs at 8 ng/ml HBp17, but a half-maximal inhibition occurs at approximately 500 ng/ml. This inhibition can easily be reversed by addition of 400 pg/ml aFGF or 100 pg/ml bFGF, whereas EGF had no effect. This biphasic action of HBp17 was also seen in human umbilical vein endothelial cells (HUVEC), whereas it was not found in the malignant cell line, A431-AJC. The functional relationship between HBp17 and FGF is discussed.

Microtubule dynamics are involved in stomatal movement of Vicia faba L.

To obtain a full picture of microtubule (MT) behavior during the opening and closure of guard cells we have microinjected living guard cells of Vicia faba with fluorescent tubulin, examined fine detail by freeze shattering fixed cells, and used drug treatments to confirm aspects of MT dynamics. Cortical MTs in fully opened guard cells are transversely oriented from the ventral wall to the dorsal wall. When the stomatal aperture was decreased by darkness, these MTs became twisted and patched and broken down into diffuse fragments when stomata were closed. When the closed stomata were opened in response to light, the MTs in guard cells changed from the diffused, transitional pattern back to one in which MTs are transversely oriented from stomatal pore to dorsal wall. This observation indicates a linkage between these MT changes and stomatal movement. To confirm this, we used the MT-stabilizing agent taxol and the MT-depolymerizing herbicide oryzalin and observed their effects on the stomatal aperture and MT dynamics. Both drugs suppressed light-induced stomatal opening and dark-induced closure. MTs are known to be necessary for maintaining the static kidney shape of guard cells; the present data now show that the dynamic properties of polymeric tubulin accompany changes in shape with stomatal movement and may be functionally involved in stomatal movement.

Distribution and partitioning of polycyclic aromatic hydrocarbons (PAHs) in different size fractions in sediments from Boston Harbor, United States.

The concentrations of 16 US EPA priority pollutant polycyclic aromatic hydrocarbons (PAHs) were analyzed in four size fractions (< 62, 62-125, 125-250, and > 250 microm) in three contaminated Boston Harbor sediments. Total PAH concentrations ranged from 7.3 to 358 microg/g dry wt. and varied largely among the different size fractions in these sediments. For all three sites, the highest PAH concentrations were associated with the large size (> 250 microm) fractions while the fine silt and clay fractions (< 62 microm) contained relatively low PAHs. Despite the great concentration differences, the composition of PAHs in the four size fractions of these sediments showed similar patterns dominated by PAHs with three or more rings. By examining the distribution patterns of selected alkyl homologs to parent compounds, the results indicate that the major PAHs contributing to the high contamination in the inner harbor sediments were from pyrogenic sources. A positive correlation between PAHs and sedimentary organic carbon exists for all size fractions in the sediments. Calculated organic carbon normalized partition coefficients (log K(oc)) for selected major PAHs indicate near-equilibrium partitioning of PAHs among the different size fractions despite their large concentration variations. Sedimentary organic matter associated with different size fractions was the controlling factor for the observed distribution differences of PAHs among the size fractions. Our results also suggest that sedimentary organic matter with different origins and maturities may have somewhat different PAH sorption characteristics. Particulate organic matter of charcoal, plant detritus and Capitella fecal pellets in the sediments appear to sorb PAHs more strongly than organic matter associated with clay minerals. The strong association of PAHs with these organic particles in sediments will have a great influence not only on their distribution but also on long-term environmental impact.
