[Value of dynamic enhanced magnetic resonance in diagnosis of pancreatic carcinoma and endocrine tumors].

OBJECTIVE: To explore the value of dynamic enhanced magnetic resonance (MR) in diagnosis of pancreatic carcinoma and endocrine tumors. METHODS: Twenty-two patients with pancreatic carcinoma and 9 patients with pancreatic endocrine tumors underwent routine MR scanning and dynamic contrast enhanced MR examination before they underwent operation. RESULTS: No tumor image was seen in 4 of the 22 patients with pancreatic carcinoma by routine MR scanning. Fat-saturated T(1)-weighted imaging (FS-T(1) WI) showed that 11 of them were hyposignal, 6 slightly hyposignal, and 1 isosignal; and fat-saturated T(2)-weighted imaging (FS-T(2) WI) showed that 5 were hypersignal, 7 slightly hypersignal, 5 isosignal, 1 slightly hyposignal, and 13 with heterogeneous signals. After dynamic MR imaging no tumor was enhanced in the arterial phase (AP); and all the tumors were obviously enhanced during the pancreatic phase (PP) and portal venous phase (PVP), 5 of which showed isosignal intensity, 10 slightly hyposignal intensity, and 3 hyposignal intensity. Four of the 5 cases with small pancreatic carcinoma were demonstrated as isosignal and one was slightly hyposignal. During the delayed enhancement phase (DEP) the signal intensity of the tumors became slightly weaker. No tumor image was seen in 14 of the 9 patients with pancreatic endocrine tumors by routine MR scanning. FS-T(1) WI showed that 5 of them were hyposignal, and 3 slightly hyposignal; and FS-T(2) WI showed that 2 were hypersignal, 4 slightly hyposignal, and 2 isosignal. After dynamic MR imaging 2 cases were moderately enhanced and 6 cases mildly enhanced during AP, and all pancreatic endocrine tumors were obviously enhanced during PP and PVP, with isosignal intensity in 2 cases, slightly hyposignal intensity in 1 case, and slightly hypersignal intensity in 5 cases. Peritumor thin-walled enhancement was seen in 6 cases. CONCLUSION: Dynamic enhanced MR has a certain value in the diagnosis of pancreatic carcinoma and endocrine tumors. Pancreatic carcinoma shows various enhanced forms by dynamic enhanced magnetic resonance, the degree of the greatest enhanced form not surpassing that of normal pancreatic tissue. Pancreatic endocrine carcinoma shows obvious enhanced forms by dynamic enhanced magnetic resonance, the degree of the greatest enhanced form surpassing that of normal pancreatic tissue. Peritumor thin-walled enhancement is a characteristic feature of pancreatic endocrine carcinoma.

[The surgical management of unilateral Craniofacial Atrophy and Hypoplasia].

OBJECTIVE: To study the surgical reconstruction of unilateral Craniofacial Atrophy and Hypoplasia. METHODS: According to the etiological factors and severity of the facial deformities, different methods are employed, including bone framework reconstruction, soft tissue transplantation, orthognathic surgery. RESULTS: From September 1998 to August 2004, 42 cases were treated, Include: Hemifacial Microsomia 22 cases, Hemifacial Atrophy 16 cases, unilateral facial hypoplasia due to radiation 4 cases. Miniplate and transplants extrusion occurred on 2 post radiation patients due to poor soft tissue coverage, infection occurred on 1 patient after mandibular ramus reconstruction using autogenous rib and contralateral mandibular outer cortex. The leaving patients recovered well and the facial asymmetry were improved greatly. CONCLUSIONS: Facial asymmetry due to unilateral Craniofacial Atrophy or Hypoplasia is a common and complex condition for surgical management, The surgical plan should be delicated made individually according to the severity of the soft tissue and the underlying bone framework.

[Study on cloning of hTERT promoter and its targeting transcriptional activities in telomerase-positive lung cancer cells].

OBJECTIVE: To clone sequence of hTERT promoter and study its transcriptional activity and its relationship with hTERT mRNA expression and telomerase activity in various kinds of human lung cancer cells and normal cells, and to investigate the targeting transcriptional activity of hTERT promoter in tumor cells. METHODS: About 1.1 kb promoter of the 5'flanking sequence of the hTERT was amplified from genomic DNA isolated from 293 cells by polymerase chain reaction (PCR). After being confirmed by DNA sequencing, the hTERT promoter was inserted into luciferase reporter vectors (pGL3-basic) to reconstruct a recombinant named pGL3-hTERTp. Then pGL3-hTERTp was transiently transfected into lung cancer cell A549, SPC-A-1, LTEPa-2, NCI-H446, YTMLC-9, GLC-82, 95D, A2, and normal cell of MRC-5. The transcriptional activities of hTERT promoter in various cells were determined by measuring the luciferase activities. hTERT mRNA expression and telomerase activity were determined by RT-PCR and TRAP ELISA. RESULTS: Eelectrophoresis demonstrated that the hTERT promoter amplified by PCR was about 1.1 kb long, and DNA sequencing showed a sequence the same as the hTERT promoter registered in GenBank being 1084 bp in length. The recombinant of plasmid pGL3-hTERTp was confirmed by double digestion and PCR methods with correct results. hTERT mRNA and telomerase activity were expressed in all of eight lung cancer cell lines at varied levels, but not expressed in normal cell. Transient transfection assay and Luciferase assay also revealed that hTERT promoter had different transcriptional activities in various lung cancer cells, but no transcriptional activity was shown in normal cells. CONCLUSION: 1084 bp hTERT promoter cloned has specific transcriptional activities in various telomerase-positive lung cancer cells, and it may act as control element in tumor-targeting gene therapy.

[Scanning electron microscopic study of titanium-hydroxyapatite, the functionally graded-material implanted in rabbits].

OBJECTIVE: The purpose of this study was to observe the tissue tolerance of Ti-HA functionally graded-material (FGM) and the form of the material-bone interface. METHODS: The sintered Ti -HA FGM, pure HA and pure Ti were respectively implanted into the parietal bone of rabbits. The specimens were observed by SEM at 2, 4, 8 postoperative weeks. RESULTS: In the early stage, the new bone surrounding the Ti -HA FGM formed earlier with larger amount and better maturity than the pure Ti. The condition was similar to the pure HA. Two months after the operation, direct bonding of material-bone interface was formed between the Ti -HA FGM and the new bone as an integral body. However, there was a little space left between the new bone and the pure Ti. CONCLUSIONS: The Ti -HA FGM has good tissue tolerance. Its early integration with bone is similar to pure HA and better than pure Ti.

[Comparative study of dissolved matter of glycyrrhizic acid before and after supermicro-pulverization of Radix Glycyrrhizae].

OBJECTIVE: To investigate various activities of dissolved matter of glycyrrhizic acid of Radix Glycyrrhizae and the powder by supermicro-pulverization. METHOD: The contents of glycyrrhizic acid in different samples were tested. RESULT: The dissolved matter of glycyrrhizic acid was greatly increased by supermicro-pulverization. The more time used for grinding, the smaller the size of the powder, and the easier the glycyrrhizic acid would be dissolved. CONCLUSION: Supermicro-pulverization is helpful to the dissolved matter of glycyrrhizic acid of Radix Glycyrrhizae, and the size of powder exerts great influence on dissolved matter of glycyrrhizic acid.

[Determination of flavone for Scutellaria baicalensis from different areas by HPLC].

OBJECTIVE: To describe the difference between native and nonative herbs by determining contents of seven kinds of flavone for twenty-five samples from seventeen areas. METHODS: HPLC. Fluid phase: MEOH-H2O-CH3COOH(ICE) (41:59:0.2) and (50:50:0.2). Detection wavelength: 275. RESULTS: The contents of baicalin are 6%-9%, wogenin are 2%-8%, baicalein are 0.1%-1.6%, neobaicalein are 0.01%-0.2%, wogonin are 0.01%-0.3%, visidulin and oroxylin are trace amounts or undetected. CONCLUSION: The native and nonative herbs have no distinct differce in absolute component ratio. The ratio of baicalin and wogenin is under three. The ratio of baicalin and baicalein, baicalin and wogonin is between twenty and fifty.
