[Cloning of A73 gene and its coding sequence analysis].

OBJECTIVE: To clone A73 gene of Epstein-Barr virus (EBV) and examine the variation of its coding sequence. METHODS: A73 coding sequence (CDS) amplified from 7 patients in nasophryngeal carcinoma (NPC) biopsies by RT-PCR was cloned into pGEM-T-Easy vector to construct the recombinant plasmid, which was subjected to sequence analysis in Gen Bank database using Blast software. RESULTS: A73 gene from nasophryngeal carcinoma was successfully cloned into pGEM-T Easy vector. A locus with conversion of A-->C in A73 was found at CDS 45 nt, located in the exonVB157154 nt, which did not result in an amino acid replacement, and the variation frequency was 7/7. CONCLUSION: There is a point mutation in A73 CDS of EBV isolated from NPC tissue, which might produce NPC-associated polymorphism with possible involvement in the composition of some subtype of EB virus.

2-Methoxyestradiol induces cell cycle arrest and apoptosis of nasopharyngeal carcinoma cells.

AIM: To investigate 2-methoxyestradiol induced apoptosis and its mechanism of action in CNE2 cell lines. METHODS: CNE2 cells were cultured in RPMI-1640 medium and treated with 2-methoxyestradiol in different concentrations. MTT assay was used to detect growth inhibition. Flow cytometry and DNA ladders were used to detect apoptosis. Western blotting was used to observe the expression of p53, p21(WAF1), Bax, and Bcl-2 protein. RESULTS: 2-methoxyestradiol inhibited proliferation of nasopharyngeal carcinoma CNE2 cells with IC(50) value of 2.82 micromol/L. The results of flow cytometry showed an accumulation of CNE2 cells in G2/M phase in response to 2-methoxyestradiol. Treatment of CNE2 cells with 2-methoxyestradiol resulted in DNA fragmentation. The expression levels of protein p53 and Bcl-2 decreased following 2-methoxyestradiol treatment in CNE2 cells, whereas Bax and p21(WAF1) protein expression were unaffected after treatment with 2-methoxyestradiol. CONCLUSION: These results suggest that 2-methoxyestradiol induced cell cycle arrest at G2/M phase and apoptosis of CNE2 cells which was associated to Bcl-2 down-regulation.

[Gene expression profiling in nasopharyngeal carcinoma determined by high density cDNA microarray].

BACKGROUND & OBJECTIVE: Multiple steps and gene expression changes were involved in the development and progression of nasopharyngeal carcinoma (NPC), but the molecular mechanism is not understanding clearly. This study was designed to investigate the profiling of differently expressed genes in nasopharyngeal carcinoma compared with noncancerous nasopharynx tissues to explore the feature of gene expression in NPC and identify new candidate genes related to the development and progression of NPC. METHODS: Differently expressed genes of NPC in 21 cases, which were divided into three pools, were investigated by using high density cDNA microarray representing 18,000 cDNA clones. The alterations in gene expression levels were confirmed by reverse-transcription PCR in 2 randomly selected genes. RESULTS: A total of 317 genes were identified to be differently expressed in NPC of three pools as compared with noncancerous controls. Among the 317 genes, 23 genes were found to be consistent in at least two NPC sample pools, including 16 up-regulated genes and 7 down-regulated genes. Six genes were found to be consistent in all the three NPC sample pools. The 6 genes include 2 up-regulated unknown genes, 3 up-regulated unknown genes and 1 down-regulated unknown gene. CONCLUSION: The present study represents a global view of gene expression of NPC and provides important clues for further study of NPC related genes.

[Inhibiting target gene expression and controlling growth of Epstein-Barr virus transformed cells by antisense RNA transcripts].

BACKGROUND & OBJECTIVES: The latent membrane protein gene (LMP) of Epstein-Barr virus (EBV) was thought to play an important role in the carcinogenesis of nasopharyngeal carcinoma (NPC). In this study, the authors investigated the effects of antisense RNA (AsRNA) on LMP for down regulating at the target gene over expression in EBV transformed lymphoid cells, and set up an antisense system to inhibit LMP expression. METHODS: Constructing the single strand antisense transcription system in vitro, the authors have got large amount of AsRNA. Designing and setting up an antisense tracing system in situ (ATSIS), the authors could observe the living particles of AsRNA/sense RNA duplex dimer. With time lapse phase-contrast microscopy, the agglutination phenotype on living cells was easily detected by MTT test, the inhibition rate on EBV transformed cells was calculated. RESULTS: LMP 1.9 fragment ligated into pGEM vector in reverse orientation have been constructed and produced a plentiful amount of AsLMPmRNA which could incorporated into both B95-8 and C1936 cell lines by endophagocytosis and formed the duplex dimer of As/Sense RNA. This particles have been visualized in situ when labelling 35S isotope by ATSIS. When AsLMPmRNA acted as agents for specific inhibition to LMP over expression, the transform phenotype of cell agglutination have been suppressed and MTT particle formatin was apparently reduced both two EBV tansformed cell lines. CONCLUSION: AsLMPmRNA targets at sense strand have a high effectiveness of down-regulation on EBV-LMP overexpression. This down regulating function of LMP and growth inhibition on transformed cell is demonstrated by the antisenes tracing system in situ (ATSIS). The results provide a clue to overcome the latent EBV infection in human bodies all living long time and to prevent it inducing NPC in high incidence area by antisense strategies.
