Identification of human saliva by antisera to alpha-amylase in human salivary glands.

Amylase activities were detected significantly in saliva from human, macaques and rodents and slightly in the vegetable and fruit extracts. Dried stains on filter paper prepared from human and mammalian saliva, and the vegetable and fruit extracts were subjected to starch-iodine test and blue starch polymer-agar plate test. Both tests showed strong positive reactions with the macaque and rodent saliva stains as well as human, but the vegetable and fruit stains showed clear positive reactions only in the starch-iodine test. The results suggest that these tests are not specific for human saliva and that for screening test of saliva stains, blue starch polymer-agar plate test is more suitable than starch-iodine test. Rabbit antisera against alpha-amylase isolated from the human submaxillary glands were prepared. In double immunodiffusion test with the human and mammalian saliva, human and macaque saliva produced precipitation lines. Human saliva gave patterns of partial identity with saliva from Japanese monkey and crab-eating monkey and these two macaque saliva gave total identity with each other. Anti-alpha-amylase sera absorbed with Japanese monkey saliva reacted only with human saliva. This suggested that the anti-alpha-amylase sera absorbed with macaque saliva make it possible to identify human saliva stain.

Nitrosamide-induced sarcomas in the gastric pylorus of hamsters. A histological and ultrastructural survey on the histogenesis.

A histological and ultrastructural survey on the histogenesis of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced gastric sarcomas in hamsters was attempted. Within 3 months after the beginning of oral administration of MNNG, simple spindle cell proliferation in the submucosal zone of the pyloric region of the glandular stomach was evident. By 5-6 months, the cells had acquired a sarcomatous appearance. Ultrastructurally, the spindle cells in the early stage, were divided into 3 types, i.e., angiogenic cells, fibroblastic cells, and primitive mesenchymal cells. Similarly, those in the late-appearing sarcomatous tumors were also classified into different types of angiogenic, fibroblastic, histiocytic and miscellaneous cells. Among the cell population of the 2 major groups, angiogenic cells were the most predominant cell type. Some morphological transition appeared to occur between the corresponding cell types at the early and late stages. DNA content of cell nuclei of the representative forms of the nitrosamide-induced sarcomas was higher than that of early appearing spindle cells. Thus, the evidence suggests that the early appearing spindle cells are precursors for the gastric sarcomas.

Enhancing effect of preadministration of carbon tetrachloride on methylazoxymethanol acetate-induced intestinal carcinogenesis.

This study concerns the modifying effect of carbon tetrachloride (CCl4) on methylazoxymethanol acetate (MAM)-induced intestinal carcinogenesis in ACI rats of both sexes. Forty five animals were given CCl4 (0.5 ml/kg body weight) through a stomach tube, followed by an i.p. injection with MAM (25 mg/kg body weight) 24 hours after CCl4 treatment. The paired administrations were done once a week for 4 weeks and animals were observed until sacrifice 30 weeks later. Pretreatment with CCl4 caused not only early death from chemical toxicity of MAM but also an increase in small-bowel tumors.

Some toxic properties of a carcinogenic pyrrolizidine alkaloid, petasitenine.

Some toxic properties of petasitenine, a hepatocarcinogenic pyrrolizidine alkaloid, were confirmed in 3 parts of studies. Electron microscopic observation of rat liver on the stage of acute toxicity following administration of a toxic dose of petasitenine disclosed the distinctive changes i.e., nucleolar segregation, degradation of endoplasmic reticulum such as formation of concentric whorls in the liver cells. Biochemical test for the mitochondrial function in the isolated mitochondria from rat liver cells revealed on inhibitory effects of petasitenine in the respiratory system, indicating that minor changes of mitochondria on the pyrrolizidine alkaloid may be induced by some other indirect factor. Investigation for the effect of petasitenine on the cell cycle of the cultured cell line from the human carcinoma showed some inhibitory effects in the appearance of cells in S-phase or M-phase, suggesting a possibility that toxicity of petasitenine will be developed in the tissues other than the liver or in the species apart from rodents.

Lack of promotive effect of quercetin on methylazoxymethanol acetate carcinogenesis in rats.

Promoting effect of quercetin in the intestinal tract was examined in Sprague-Dawley strain rats. The rats were injected with methylazoxymethanol acetate (MAM) in saline solution at 25 mg/kg body weight once a week for 3 weeks and fed a diet containing 1% quercetin for the following 459 days. Most tumors found in this experimental group were also found in the group injected with MAM alone and there was no significant difference in tumor incidence between these two groups. No tumors were found in the group fed the 1% quercetin diet alone. Thus, quercetin was shown to lack of promoting activity on MAM-induced intestinal carcinogenesis in Sprague-Dawley rats.

A cytochemical study of lactic dehydrogenase (LDH) isoenzymes in human lung cancer.

The cytochemical and electrophoretic LDH isoenzyme patterns in cells from lung cancer tissues were examined with 2.6 M urea treatment and the correlation between the LDH isoenzyme pattern and histopathological entities of lung cancer was also studied. The zymograms of the nonlesional lung tissues indicated the main peak at LDH3, with the M/H ratio 0.74. In tumor tissues, epidermoid carcinoma, adenocarcinoma, and large cell carcinoma showed almost similar patterns of isoenzymes with LDH4 peak. Though small cell carcinomas had a peak at LDH3, they indicated lower activity of %LDH1 and %LDH2 than those in nonlesional lung tissues. Most of the tumor tissues showed a high M/H ratio, more than 1.0. In cytochemical stain for LDH, no inhibitory effect of urea treatment was seen in the nonlesional lung tissues. On the other hand, in the cells obtained from three types of lung cancer, except for small cell carcinoma, urea inhibition was noticed. Cytochemical stainability for LDH-M subunits corresponded well to the results of LDH zymogram. These results suggest that the LDH stain with urea treatment is a useful method for detecting malignancy in cytological specimens.

Malignant mesenchymoma of the heart.

A case of a rare primary cardiac tumor in a 46-year-old woman is described. The tumor arose from the left atrium and was histologically composed of multiple mesenchymal elements of fibrosarcoma, rhabdomyosarcoma, chondrosarcoma, myosarcoma, and liposarcoma. Metastasis of this tumor occurred in the left femur, lung, and hilar lymph nodes after the second heart operation. Histogenesis of malignant mesenchymoma was considered with a survey of the literature.
