Analysis of the STR loci HUMF13A01, HUMFXIIIB, HUMLIPOL, HUMTH01, HUMTPOX and HUMVWFA31 in a Japanese population.

Population studies on six short tandem repeat loci, HUMF13A01, HUMFXIIIB, HUMLIPOL, HUMTH01, HUMTPOX and HUMVWFA31 were carried out in a sample of unrelated Japanese individuals (n = 337-545) living in Gifu Prefecture (central region of Japan). Five alleles could be identified for HUMFXIIIB, six for HUMF13A01, HUMLIPOL, HUMTH01 and HUMTPOX, and eight for HUMVWFA31. For all/six loci no deviations from the Hardy-Weinberg equilibrium hypothesis were detected. The mean exclusion chance ranged from 0.22 to 0.60, the power of discrimination from 0.63 to 0.93, and the expected heterozygosity from 0.43 to 0.80. Allele frequency distributions for the loci in the Japanese sample were not similar to those in samples from other racial or ethnic groups except for the Chinese (for HUMTPOX). The results demonstrate that HUMTH01, HUMTPOX and HUMVWFA31 are more useful for forensic investigations in the Japanese population that the other three loci.

Isoelectric focusing differences in the mobility of blood and semen PGM1 bands--the role of hemoglobin as a cause.

When using isoelectric focusing for the analysis of phosphoglucomutase isoenzymes, differences between the mobility of the first locus phosphoglucomutase (PGM1) bands have been noted in blood and semen samples from the same individual. This study was thus initiated to determine whether hemoglobin is responsible for these differences. Results revealed that the mobility of the PGM1 bands in diluted hemolysate and hemoglobin-removed hemolysate was similar to that seen in non-treated seminal plasma. Further, the mobility of PGM1 bands in hemoglobin-added seminal plasma was also similar to that seen in non-treated hemolysate. Additionally, on isoelectric focusing of the pI-markers, as well as non-treated hemolysate, hemoglobin-removed hemolysate, hemoglobin-added seminal plasma, and non-treated seminal plasma, distortions of iso-pH lines, i.e., "crank" formations were seen in the lanes of the non-treated hemolysate and hemoglobin-added seminal plasma. The above findings appear to indicate that differences in the mobility of PGM1 bands in the blood and semen result from the presence of high concentrations of hemoglobin in the analyzed samples.

Ultrastructural localization of carbonyl reductase in mouse lung.

The immunocytochemical localization of tetrameric carbonyl reductase in the mouse lung was determined by an electron-microscopical immunogold procedure using monospecific antibodies against the enzyme. The labelling of carbonyl reductase was observed within the mitochondria of the ciliated and non-ciliated cells of the bronchioles and the type II alveolar pneumocytes, and the density of labelling in the non-ciliated cells was higher than those in the other cells. No significant labelling was detected over other compartments of the epithelial cells. The labelling was undetectable in the type I alveolar cells, alveolar macrophages and connective tissue cells of the lung. These results clearly indicate the localization of carbonyl reductase to the mitochondrial matrix of these epithelial cells, of which the non-ciliated bronchiolar cells contained particularly high amounts of the enzyme.

Analysis of the VNTR locus D1S80 in a Japanese population.

A population study on the VNTR locus D1S80 was carried out in a sample of 377 unrelated Japanese individuals living in and around Gifu Prefecture (central region of Japan). A total of 29 different alleles was distinguished. Alleles 18, 24 and 30 were found to be the 3 most common alleles in this population sample and their frequencies were 0.147, 0.200 and 0.162, respectively. Out of the 435 possible phenotypes, 120 were observed. The observed heterozygosity was 0.88 and the power of discrimination was 0.98. No significant deviations from Hardy-Weinberg equilibrium could be found in this Japanese population sample.

A rare case of serous cystadenocarcinoma of the pancreas.

Serous cystadenocarcinoma of the pancreas, a rare disease, developed in a 63-year-old Japanese woman. Pathologic examinations of the pancreatic tumor at the subtotal pancreatectomy showed it to be serous cystadenoma with focal atypical lesions. Three years after the operation, however, metastatic liver nodules were found, and the histologic characteristics of these lesions were quite similar to those of the pancreatic neoplasm. Both primary and metastatic tumors were composed of multiple cysts separated by fibrous septa. The epithelium of cysts was cuboidal and had clear cytoplasm, which had positive results for periodic acid-Schiff (PAS) and negative results for PAS with diastase, Alcian blue, and mucicarmine. To the knowledge of the authors, serous cystic neoplasms of the pancreas have been uniformly benign in biologic behavior. Recently, however, serous cystadenocarcinoma of the pancreas has been reported as a new entity. The current case is the second reported case and might support the existence of serous cystadenocarcinoma of the pancreas.

Purification and characterization of pig lung carbonyl reductase.

A pyrazole-sensitive carbonyl reductase from pig lung was purified to homogeneity by electrophoretic criteria. Chemical cross-linking study suggested that the native enzyme is a tetramer with a Mr of 103,000, consisting of apparent identical subunits of Mr 24,000. The enzyme reduced aliphatic and aromatic carbonyl compounds with NADPH as a preferable cofactor to NADH and catalyzed the oxidation of secondary alcohols and the aldehyde dismutation in the presence of NAD(P)+. Immunohistochemical study with the antibodies against the enzyme revealed that the enzyme was localized in the ciliated cells, nonciliated bronchiolar cells, Type II alveolar pneumocytes, and the epithelial cells of the ducts of the bronchial glands in the pig lung. In addition to the properties and distribution, the pig lung enzyme was immunochemically similar to the pulmonary enzymes in the guinea pig and mouse. However, the pig enzyme showed the following unusual features. (1) The enzyme exhibited an equatorial specificity in the reduction of 3-ketosteroids; the 4-pro-S hydrogen of NADPH was transferred to the carbonyl carbon atom of 5 alpha- and 5 beta-androstanes, and the respective reduced products were identified as 3 beta- and 3 alpha-hydroxysteroids. (2) Although the NADPH-linked reduction of carbonyl compounds apparently obeyed the Michaelis-Menten kinetics at pH 6.0, the double-reciprocal plots of the velocity vs concentrations of the carbonyl substrates were convex at pH higher than 6.5. The Hill coefficients and [S]0.5 values for the substrates decreased as the pH for reaction increased. The results suggest that the pig enzyme exhibits negative cooperativity with respect to the carbonyl substrates and that the hydrogen ion acts as an allosteric effector abolishing the negative interaction.

Vomit identification by a pepsin assay using a fibrin blue-agarose gel plate.

Reported is a simple and reliable method for identifying the presence of gastric fluid in forensic samples by an assay that reveals the pepsin activity. These samples are usually vomit found at the scene of a crime, either in fresh form or as a dried stain on clothing. The pepsin within the sample is assayed for its proteolytic activity which is revealed in a fibrin blue-agarose gel plate, as a result of an enzymatic reactivity that takes the form of a concentric, blue, translucent ring around the tested sample. Apart from being able to determine the pepsin content of fresh or recent forensic samples, this method has also achieved positive reactions in aged gastric fluid stains that were kept at room temperature. No body fluids other than the gastric fluid and no proteolytic enzymes other than pepsin show a positive reaction with the use of this method. This method has an additional advantage, in that the enzymatic activity seen on the gel plate can be photographed and the gel plate, on drying, can also be preserved as evidence.

Extra-adrenal pheochromocytoma-ganglioneuroma. A case report.

A case of rare extra-adrenal tumor composed of pheochromocytoma-ganglioneuroma which developed in a 48-year-old Japanese male is reported. Histologically, the tumor contained equal proportion of two distinct patterns, pheochromocytoma and ganglioneuroma. Immunohistochemical examination revealed that pheochromocytoma cells were positive for Leu-7 and ganglion cells in ganglioneuroma were positive for vasoactive intestinal peptide (VIP), respectively. Neuron specific enolase (NSE) was positive in the neoplastic cells of both components, and S-100 protein was also positive in fibers around ganglion cells. Ultrastructural examination revealed that neurosecretory granules were present in the neoplastic cells.

Pathological approaches to SIDS. I. Special emphasis on the histopathological differences between SIDS and asphyxia.

Histopathological observation was performed to clarify pathological differences of the lung between SIDS and asphyxia. In regard to pulmonary edema, there was no significant difference between SIDS and asphyxia. Occurrences of microemboli in the pulmonary vessels in cases of SIDS showing aspiration of gastric content into the lung, asphyxia and drowning were significantly high, compared with that of SIDS cases without aspiration.

Effect of hair dyes and bleach on the hair protein patterns as revealed by isoelectric focusing.

The effect of hair dyes, i.e., temporary, semi-permanent, or permanent hair dyes, or hair bleach on the isoelectric focusing (IEF) hair protein patterns was studied. A permanent hair dye (metallic, alkaline oxidative, or acidic oxidative) and hair bleach induced changes in the IEF hair protein patterns and in the intensity of hair protein bands. The changes in the IEF patterns, caused by the alkaline oxidative dye or the bleach, are considered to result from the combined effect of an alkaline agent and an oxidative agent in the alkaline oxidative dye and in the hair bleach.

Distribution of A and B antigens in organs of blood group AB individuals: observations disclosed by a double immunoenzymatic labeling method.

The localization of A and B antigens in the organs of blood group AB individuals has been studied using a double immunoenzymatic labeling method. Both A and B antigens were found in the various epithelial cells of these organs, but the epithelial cells could be classified into the following four types depending on the reaction pattern with anti-A and anti-B sera: type 1: cells that stained positive with both anti-A and anti-B sera; type 2: cells that stained positive with anti-A serum only; type 3: cells that stained positive with anti-B serum only; type 4: cells that were negative with both sera. The distribution of each of these epithelial cell types varied considerably, even in the same tissue and individual. Our results seem to suggest that a dissociation in the conversion to the A and B antigens occurs in the tissue of individuals belonging to blood group AB and that the degree of this dissociation varies from tissue to tissue and from cell to cell.

Localization of pulmonary carbonyl reductase in guinea pig and mouse: enzyme histochemical and immunohistochemical studies.

We studied the localization of carbonyl reductase (E.C. 1.1.1.184) in guinea pig and mouse lung by enzyme histochemistry and immunohistochemistry, using antibodies against the guinea pig lung enzyme which crossreacted with the lung enzymes of both animals. Carbonyl reductase activity was detectable in the bronchiolar epithelial cells of small airways and in alveolar cells. In the immunohistochemical staining for carbonyl reductase, the reaction was strongest in the non-ciliated bronchiolar cells (Clara cells) and was weak in the ciliated cells and type II alveolar pneumocytes. Injection of a single dose of naphthalene led to significant impairment of carbonyl reductase activity and of microsomal mixed-function oxidase activities in mouse lung, with a marked decrease in both activity and immunoreactive staining in the bronchiolar epithelial cells. The results indicate that carbonyl reductase is localized primarily in the Clara cells, which are known to be sites of pulmonary drug metabolism.

Dimeric dihydrodiol dehydrogenase in monkey kidney. Substrate specificity, stereospecificity of hydrogen transfer, and distribution.

Chemical cross-linking and NADPH binding studies suggested that the native dihydrodiol dehydrogenase from monkey kidney is a basic dimer having a molecular weight of 78,000 and one active site per the subunit. The enzyme oxidized specifically trans-dihydrodiols of benzene and naphthalene, whereas it catalyzed the reduction of dihydroxyacetone and dihydroxyacetone phosphate at a physiological pH, 7.4. The Km and kcat values for dihydroxyacetone phosphate were 5.0 mM and 4.3 s-1, respectively. The enzyme transferred the 4-pro-R hydrogen atom of NADPH to the carbonyl substrate. Immunochemical experiments using an antibody against the dimeric enzyme revealed the specific distribution of the enzyme in the kidney of this animal. By immunohistochemical staining with the specific antibody, the immunoreactivity was found in proximal and distal tubules of the cortex, and in the loop of Henle of the medulla.

An application of a quantitative analytical system for the grading of pulmonary fat embolisms.

The severity of pulmonary fat embolism in 5 autopsied cases has been measured using a quantitative image analytical system. With reference to the mean size of the fat emboli, the cases were divided into 2 groupings regardless of the number of the emboli. The mean sizes of the emboli in 3 cases of the first group were significantly larger (about 490-600 microns 2) than those found in the 2 cases of the other group (about 220 and 235 microns 2). An investigation into the localization of fat emboli revealed that more were lodging in the small arteries and arterioles in the first group than in the second. Our results have indicated that a reliable grading of pulmonary fat embolism can not be established without a quantitative image analysis of the size and localization of the fat emboli, and that this quantitative analytical method is useful in achieving this reliable grading.

Medicolegal aspects of death of patients with cerebral palsy in Japan.

A total of 24 cases of patients who had had cerebral palsy, recorded between 1980 and 1986, in the Annuals of the Medico-Legal Autopsy Cases in Japan, were investigated to determine their ages and causes of death. Fourteen died of illness, four in accidents, and six by murder. Most of those dead of illness had been under the age of 16, and the disease most frequently cited as a cause of death was pneumonia. The four persons who died by accident were over 16 years old. All of the six murder victims were killed by their mother.

Immunohistochemical localization of human lung adenocarcinoma-associated antigen with a monoclonal antibody.

The monoclonal antibody, 5C7, reacted immunohistochemically with 62 out of bronchial cells of normal lung tissue, adjacent to neoplastic lesions, were negative for lung adenocarcinoma-associated antigen. Reactions with the antibody were observed in half the cases of squamous cell lung cancer, but were only sporadic. The antibody appears to react with an antigen which is either restricted to malignant cells or is at least greatly amplified in expression by malignant cells compared to normal human tissues. The major value of this monoclonal antibody at present is in classifying lung cancers as either adenocarcinoma or non-adenocarcinoma.
