Molecular nature of three liver alkaline phosphatases detected by drug administration in vivo: differences between soluble and membranous enzymes.

1. Activities of alkaline phosphatase, liver-membranous, liver-soluble and serum-soluble, were dramatically induced in dogs by treatment with both phenobarbital and brovanexine. The treatment induced a 17-fold increase in membranous, a 155-fold increase in soluble, and a 105-fold increase in serum alkaline phosphatases. 2. There was no difference in the enzymatic behavior of the three forms of alkaline phosphatase, on heat stability, amino acid inhibition and optimum pH. 3. When the three alkaline phosphatases were treated initially with n-butanol, their apparent molecular size was identical. After treatment with phosphatidylinositol-specific phospholipase C, the liver-soluble and serum-soluble alkaline phosphatase were of the same molecular size. Liver-membranous alkaline phosphatase, however, was larger in molecular size than the other two forms, suggesting a difference between soluble and membranous alkaline phosphatase forms. 4. In terms of the sugar moiety of the three alkaline phosphatase forms, the membranous enzyme showed more of the higher affinity fraction and less of the lower affinity fraction of concanavalin A, compared with the soluble enzymes. 5. Consequently, it is possible that the membranous enzyme may be solubilized by an enzyme such as phosphatidylinositol-specific phospholipase C and modify further the sugar moiety of alkaline phosphatase molecules, resulting in serum alkaline phosphatase transfer from the soluble enzyme in liver.

[Comparative studies on multiple forms of serum cholinesterase in various species].

Multiple forms of serum cholinesterase (ChE) were compared in 8 species by electrophoretic technique and the following characteristics were noted. The first moving fraction markedly hydrolyzed butyrylthiocholine and the activity was not inhibited by 10(-5)M eserine in the serum of some rabbits tested. Electrophoretic patterns of the ChE were obtained by use of two thiocholines as substrate, and the number of fractions against acetylthiocholine were more than against butyrylthiocholine in dogs, miniature pigs, rabbits, and hamsters. The activities of ChE fractions of dogs (C3), miniature pigs (C1, C2), rabbits (C1), and hamsters (C3) were inhibited by 6.1 X 10(-2)M caffein but not by 10(-4)M ethopropazine, which suggests that the fractions are all true-ChE.

Purification and some properties of bovine liver cytosol thioltransferase.

A cytosol thioltransferase was purified 37,000-fold from bovine liver by essentially the same procedure as reported for rat liver enzyme by Axelsson et al. [1978) Biochemistry 17, 2978-2984). The purified enzyme appears to be homogeneous on sodium dodecyl sulfate (SDS)-gel electrophoresis and has a molecular weight (Mr) of 11,000, an isoelectric point (pI) of 8.1, and an optimum pH with S-sulfocysteine and GSH as substrates of 8.5. It is specific for disulfides including L-cystine, S-sulfocysteine, ribonuclease A, trypsin, soybean kunitz trypsin inhibitor, soybean Bowman Birk trypsin inhibitor and insulin, and converts Bowman Birk trypsin inhibitor to an inactive form. The enzyme does not act as a protein : disulfide isomerase, as measured by reactivation of "scramble" ribonuclease and Kunitz soybean trypsin inhibitor. Thioltransferase activity was found in cytosol of various bovine tissues.

Coronary spasm as a cause of perioperative myocardial infarction.

The case of a patient with severe left main trunk disease is presented, in which spasm of the right coronary artery resulted in intraoperative myocardial ischemia and perioperative myocardial infarction. It is suggested that coronary spasm may be a cause of perioperative myocardial infarction, and has to be considered in case of unknown etiology.

Two cases of red cell aldolase deficiency associated with hereditary hemolytic anemia in a Japanese family.

Two cases of red cell aldolase deficiency associated with congenital nonspherocytic hemolytic anemia are reported. The proband is a fourteen-month-old Japanese boy. Consanguineous marriage was not proven but probable in this family, as the parents were born in the same small island. The proband had moderate to mild anemia aggravated by upper respiratory infections, 1 cm hepatomegaly and 2.5 cm splenomegaly, but was unremarkable in other respects and has thus far not shown mental or growth retardation. He did not have dysmorphic features. The red cell aldolase activity was 6% of the normal mean. The enzyme was unstable with respect to heat, and Km for fructose 1,6-diphosphate (F-1,6-DP) was high. The parents and other heterozygotes showed intermediate activity between that of the proband and that of normal subjects. Red cell F-1,6-DP concentration in this case was remarkably increased. Red cell glucose consumption, and lactate formation, as well as hexose monophosphate shunt activity, were decreased as compared with a comparable reticulocyte-rich hereditary spherocytosis patient. Hexose monophosphate dehydrogenase by a high concentration of F-1,6-DP in his red cells. As a result of family study, another homozygous aldolase deficiency case associated with hemolytic anemia was found. He is 13 years old and a nephew of the proband's paternal grandmother. His hemolytic anemia also is moderate to mild and aggravated by upper respiratory infections. He does not seem to have mental or growth retardation, nor does he possess dysmorphic features.

[Exercise echocardiography: interventricular septal thickness and motion in patients with effort angina during ergometer exercise (author's transl)].

To detect abnormal interventricular septal (IVS) motion during exercise-induced ischemia, ergometer exercise echocardiography was performed using a specially devised transducer in 12 patients (pts) with effort angina (left anterior descending artery disease) and 10 normal subjects (N) at rest, and during exercise and recovery. During exercise, percent systolic IVS thickening (% delta T) and IVS excursion (Ex) increased from 52 +/- 13% at rest to 73 +/- 19% and from 7.0 /- 1.3 mm at rest to 10.6 +/- 1.9 mm, respectively, in N, and also from 52 +/- 23% to 67 +/- 36% and from 7.3 +/- 1.9 mm to 9.7 +/- 2.1 mm in all of 3 pts with distal left anterior descending artery disease. On the other hand, in 9 pts with proximal left anterior descending artery disease, % delta T and Ex during exercise decreased from 41 +/- 17+ at rest to 26 +/- 25% and from 7.7 +/- 1.2 mm to 5.1 +/- 4.6 mm. The late systolic wall thickening of IVS was observed during peak exercise in 2 of the 9 pts, one of whom exhibited systolic IVS thinning and a decrease in diastolic thickness (from 6 mm to 4.5 mm). In 5 pts with IVS asynergy during exercise diastolic IVS thickness increased maximally from 10.2 +/- 3.3 mm at rest to 11.4 +/- 3.5 mm during recovery (reactive hyperemia). Exercise echocardiography is useful to predict the location of left anterior descending artery disease and to evaluate IVS performance during exercise-induced ischemia.

[Hematochemical characteristics of miniature pig Göttingen (miniature pig G) (author's transl)].

The biochemical components, enzymes, and other items from the sera and plasma of normal male and female Göttingen miniature pigs from 3 to 20 months of age were investigated. The data were compared with the normal biochemical values of rhesus monkeys, beagle dogs, and JCL: SD rats. The following characteristics were noted. 1. Activities of serum enzymes were higher and their normal ranges wider than in the other experimental animal sera. 2. Of lipid components measured, total cholesterol and phospholipid showed strikingly lower values than in the monkey and the dog. 3. The serum urea nitrogen levels were lower than in the other experimental animal sera. 4. Serum creatinine and potassium were present at relatively high levels, with wide normal ranges. 5. There was a tendency with advancing age toward lower alkaline phosphatase and higher total protein and creatinine. 6. Leucine aminopeptidase was higher while total cholesterol and phospholipid were lower, in the male than in the female. 7. The electrophoretic patterns of serum protein and lipoprotein differed markedly among the species.

[Fundamental and clinical studies of cefoperazone in pediatric patients (author's transl)].

1. Susceptibility, concentrations in serum and spinal fluid, and clinical effect of cefoperazone (CPZ), a newly developed cephalosporin in Japan, were examined in 28 infants from 2 days to 12 years of ages with various infections. 2. The overall clinical evaluation in 22 cases received CPZ alone was found to be markedly effective in 11 cases and effective in 6 cases; the efficacy rate was as good as 85.0%. 3. The mean half-life following a single intravenous injection of 25 mg/kg to 3 infants was 80.7 minutes. 4. The side effects observed during the therapy were 2 cases of rash and 1 case of vascular pain and these symptoms improved after interruption of therapy. The side effects of CPZ seemed to be not more noticeable than other cephalosporins.
