[A cross-sectional study on prevalance and risk factors of disabilities in aged 0-6 years children in Beijing, China].

OBJECTIVE: To explore the epidemiological status of disabilities on vision impairment (VI), hearing loss (HL), mental retardation (MR), autism spectrum disorder (ASD) and motor disorder (MD) in aged 0-6 years old children in Beijing. METHODS: A total of 28 738 children under 7 years old were recruited from permanent residents of Beijing City by 2-phase cluster sampling. The screening procedure was 2-phase, and the diagnosis criteria were developed by the experts group. RESULTS: The overall disability rate was 11.45 per thousand (95 % CI:10.22-12.68). The false negative rates in HL and ASD were 0.14 per thousand, 0.80 per thousand, respectively, with a adjusted overall rate of 12.19per thousand. The prevalence rates of different kind disabilities from high to low were MR 9.31per thousand, MD 2.12per thousand, ASD 1.53per thousand (0.73per thousand before adjusted), HL 1.04per thousand (0.91per thousand before adjusted) VI 0.73per thousand. The results of logistic regression analysis showed that the possible non-biological risk factors for those disabilities were being male, living in city area, advancing age, mother with low education, mother engaged in labor work, and family with low income. It was primarily (49.62%) those prenatal factors other them the known suspected factors that causing disabilities. CONCLUSIONS: Compared with data from a countrywide study in 1987, the overall disability rate had a mild decrease (16.36%) with the most (56.85%) appeared in HL. It is indispensable to establish a disabilities surveillance program for the early recognition and intervention of children with disabilities. It is also crucial to clarify a disability definition for children combined with their characteristics of growth and development. We strongly recommended in developing a new definition on children' disabilities and establishing new criteria according to the contents of developmental disabilities of Center for Disease Control, USA.

[Electrical charges and coagulation efficiency of Al13 species in polyaluminum chloride (PAC)].

The polynuclear Keggin species, Al12AlO4(OH)24(H2O)12(7+) (Al13), was separated and purified from polyaluminum chloride (PAC) by gel column chromatographic method. The purified Al13 was characterized by Al-Ferron timed complexation spectrophotometry method and ZATESIZER 3000HSA tester compared with PAC and ALCl3. Jar tests were performed to test the coagulation efficiency of the three coagulants in treating synthetic or actual water samples. The results showed that Al13 could be obtained by the column chromatographic method and the content of it could reach about 99%. Compared with PAC and AlCl3, Al13 gives the best results for turbidity, humic acid and color removal, and achieves the highest charge-neutralizing ability. Al13 species is a higher positive-charged and wide pH range application, it is much more steady in hydrolyze process and most effective polymeric Al species in water and wastewater treatment.

On-line optical determination of floc size of Fe(III) coagulants.

This study investigated the floc aggregation, average floc size, floc size variance and floc growth velocity when ferric chloride (FeCl3) and polyferric chloride (PFC) were used to treat the simulated water samples. The factors including coagulant dose, ionic strength and solution pH, which affect the floc aggregation, were studied. Experiments were carried out in a bench-scale reactor using photometric dispersion analyzer (PDA). Results showed that there were great differences between the floc aggregation of PFC and FeCl3. The average floc size and floc growth velocity of PFC were much larger than those of FeCl3. Compared with FeCl3, PFC gave a better coagulation performance in wider range of pH, dosage and ionic strength. It was also found that the coagulation efficiency of PFC did not depend on average floc size but on floc growth velocity.

[Poorly-immunogenic tumor is capable of inducing proliferation of CD4 (+) CD25 (+) regulatory T-lymphocytes in vitro].

OBJECTIVE: To investigate the distribution of regulatory T-lymphocytes in the splenocytes cocultured with syngeneic low-immunogenic tumor cells, as compared with that of highly-immunogenic tumor cells, to investigate the mechanism underlining tumor evasion. METHODS: Three different immunogenic tumor cells were cocultured with syngeneic splenocytes individually to mimic cancer immunity in vitro. The proliferation response of splenocytes was measured by thymidine incorporation. The distribution of TR cells, CD4(+) IFN-gamma (+) T cells and CD4(+) IL-10(+) T cells were analyzed by flow cytometry. The secretion of IFN-gamma and IL-10 in supernatants was measured by ELISA assay. RESULTS: The stimulation Index of splenocytes cocultured with syngeneic highly-immunogenic H22 or FBL3 was much higher than that of poorly immunogenic melanoma D5. In each group, stimulation Index of splenocytes cocultured with allogeneic tumor cells was higher than that of the corresponding tumor immunity model. In addition, compared with those of highly-immunogenic tumors, there were more TR, CD4(+)IL-10(+) and less CD4 (+)IFN-gamma(+) T cells in the splenocytes, and higher IL-10 and lower IFN-gamma levels in the supernatant of the splenocytes stimulated with low-immunogenic D5 cells. CONCLUSION: Poorly-immunogenic tumor cells can induce the proliferation of TR cells, which may play an important role in tumor evasion.

[Targeted inhibition of vascular endothelial growth factor (VEGF) expression in human osteosarcoma cell line by antisense VEGF165 cDNA promoted by hypoxia reaction element].

OBJECTIVE: Utilizing the hypoxia inducible factor 1/hypoxia reaction element (HIF-1/ HRE) gene regulation system to construct antisense vascular endothelial growth factor (VEGF165) cDNA eukaryotic expression vector promoted by HRE, and investigate its targeted inhibiting VEGF expression of osteosarcoma cells in hypoxia environment. METHODS: Eukaryotic expression plasmid with HRE promoter was constructed containing luciferase reporter gene and antisense VEGF165 cDNA by using PCR and recombinant DNA techniques. The recombinant vectors were transfected into osteosarcoma cells with lipofectin method. Hypoxia-inducible reporter gene expression was determined by liquid scintillation analyzer and the expression of VEGF protein was detected by ELISA method. RESULTS: The eukaryotic expression plasmid containing antisense VEGF165 and luciferase promoted by HRE was constructed successfully. After being transferred into MG63 cells, luciferase expression was increased 3.5 x 10(2) times and VEGF protein expression decreased 45% under hypoxia condition. CONCLUSION: Antisense VEGF165 cDNA expression, efficiently realized by HRE promoter under hypoxia condition, provides an experimental basis for targeted antiangiogenesis of tumors.

[Change in endothelial progenitor cells and endothelial function in patients with unstable angina pectoris].

OBJECTIVE: To investigate the relationship between endothelial progenitors cells (EPCs) and endothelium-dependent vasodilation in patients with unstable angina pectoris. METHODS: Thirty patients with unstable angina pectoris (UAP) and thirty control subjects were recruited. Flow-mediated dilation (FMD) in the brachial artery was evaluated by using ultrasound Doppler flow method. The number of circulating EPCs was analyzed by flow cytometry analysis. Total mononuclear cells were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin-coated culture dishes. CD34 antigen of adherent cells was identified by immunohistochemical assay. EPCs were characterized as adherent cells double positive for FITC-UEA-I binding and DiI-acLDL uptake by direct fluorescent staining under inverted fluorescent microscope. RESULTS: FMD was significantly impaired in the UAP group compared with the control group (5.93% +/- 2.67% vs 11.1% +/- 4.36%, P < 0.05). There was no significant difference in NMD between two groups (13.60% +/- 5.03% vs 14.18% +/- 4.50%, P > 0.05). The number of CD34(+) cells significantly increased in the UAP group compared with the control group (0.13% +/- 0.05% vs 0.09% +/- 0.04%, P < 0.05). There was a negative association between impaired FMD and increased CD34(+) cell (r = -0.385, P < 0.05). A positive antigen of CD34 of adhesion cells and double positive adhesion cells were found. CONCLUSION: This study shows that the levels of peripheral CD34(+) cells in patients with UAP increase with an impaired endothelial function. The increase in EPCs may be an important compensatory response to acute coronary ischemia and impaired endothelium in patients with UAP.

[Arachidonic acid epoxygenases and their metabolites promote angiogenesis].

OBJECTIVE: To investigate the angiogenetic effects of endogenous and exogenous epoxyeicosatrienoic acids (EET) and the relevant signaling mechanisms involved. METHODS: Bovine aortic endothelial cells (BAEC) were incubated with synthetic EETs or infected with recombinant adeno-associated viruses (rAAV) containing CYP2C11-CYPOR, CYP2J2 or CYP102 F87V mutant to increase endogenous expression levels of EETs. BAEC proliferation measured by cell counting and chromatometry, migration assessed by transwell analysis, and capillary formation determined by chicken embryo chorioallantoic membrane assays (CAM) and tube formation tests on matrigel and angiogenesis were analysed in vivo. The potential involvement of various signaling pathways were explored using selective inhibitors. RESULTS: Transfection with rAAV-2C11OR, rAAV-2J2 or rAAV-F87V promoted BAEC proliferation, migration, and capillary tubule formation. However, the effects of EETs on proliferation, migration and capillary tubule formation were attenuated by inhibitors of mitogen-activated protein kinase (MAPK) and phosphatidylinositol 3-kinase (PI3 kinase)/Akt pathways, and partially attenuated by endothelial nitric oxide synthase (eNOS) inhibitor, but not by a protein kinase C inhibitor. In a rat ischemic hind limb model, rAAV-mediated epoxygenase transfection induced angiogenesis. CONCLUSIONS: Arachidonic acid epoxygenase and its metabolites can promote angiogenesis through activating MAPK and PI3 kinase/Akt signaling pathways, and to some extent, the eNOS pathway, and the angiogenic effects may provide protection to ischemic tissues.

[Tumor necrosis factor-alpha induces release of endothelial microparticles from human endothelial cells].

OBJECTIVE: The present study was designed to investigate whether Tumor necrosis factor (TNF)-alpha stimulates release of endothelial microparticles (EMPs) by human endothelial cells, and whether EMPs may serve as a promising marker for endothelial injury and dysfunction. METHODS: Human umbilical venous endothelial cells (HUVEC) were incubated with or without TNF-alpha for 24 hours at 37 degrees C. EMPs generated on the surface of HUVEC were observed with a scanning electron microscopy. The CD31 and CD51 positive EMPs in culture supernatants were measured by flow cytometer. RESULTS: Fewer vesicles were observed on cell surface of control group, in TNF-alpha-stimulated one, however, cells manifested a blebby surface (eruption phenomenon) and more vesicles on surface were observed. The levels of EMPs were significantly increased in TNF-alpha stimulated cells compared with controls [CD31 + EMP, (164 +/- 63)/1000 cells vs. (42 +/- 10)/1000 cells, P < 0.05; CD51 + EMP, (260 +/- 108)/1000 cells vs. (19 +/- 4)/1000 cells, P < 0.05]. CONCLUSION: TNF-alpha can stimulate HUVEC to release EMPs which may serve as a surrogate marker for endothelial injury and dysfunction.

Impaired up-regulation of CD25 on CD4+ T cells in IFN-gamma knockout mice is associated with progression of myocarditis to heart failure.

Inflammation has been recognized increasingly as a critical pathologic component of a number of heart diseases. A mouse model of autoimmune myocarditis was developed to study the role of immune mediators in the development of cardiac dysfunction. We have found previously that IFN-gamma deficiency promotes inflammation in murine myocarditis. It has been unclear, however, how IFN-gamma deficiency in myocarditis affects cardiac function and what underlying immune mechanisms are responsible for these effects. In this work, we show that IFN-gamma knockout (KO) mice have more pronounced systolic and diastolic dysfunction and greater frequency of progression to dilated cardiomyopathy and heart failure compared with WT mice. Cardiac dysfunction in the KO mice is associated with the expansion of activated (CD44(high)) CD3+ T cells due to reduced apoptosis of CD4+, but not CD8+, T cells. CD4+ T cells in the KO mice show impaired up-regulation of CD25 upon activation, resulting in the expansion of CD4+CD44+CD25- T cells and their infiltration into the heart. CD4+CD25- T cells are less apoptosis-prone compared with the CD25+ population, and their infiltration into the heart is associated with greater severity of myocarditis. We conclude that IFN-gamma deficiency in autoimmune myocarditis is associated with preferential expansion of CD4+CD44+CD25- T cells resulting in increased cardiac inflammation. An exaggerated inflammatory response in IFN-gamma KO mice causes cardiac dysfunction, leading to dilated cardiomyopathy and heart failure.

Discovery of 4-aryl-4H-chromenes as a new series of apoptosis inducers using a cell- and caspase-based high-throughput screening assay. 1. Structure-activity relationships of the 4-aryl group.

By applying a novel cell- and caspase-based HTS assay, 2-amino-3-cyano-7-(dimethylamino)-4-(3-methoxy-4,5-methylenedioxyphenyl)-4H-chromene (1a) has been identified as a potent apoptosis inducer. Compound 1a was found to induce nuclear fragmentation and PARP cleavage, as well as to arrest cells at the G(2)/M stage and to induce apoptosis as determined by the flow cytometry analysis assay in multiple human cell lines (e.g. Jurkat, T47D). Through structure-activity relationship (SAR) studies of the 4-aryl group, a 4- and 7-fold increase in potency was obtained from the screening hit 1a to the lead compounds 2-amino-4-(3-bromo-4,5-dimethoxyphenyl)-3-cyano-7-(dimethylamino)-4H-chromene (1c) and 2-amino-3-cyano-7-(dimethylamino)-4-(5-methyl-3-pyridyl)-4H-chromene (4e), with an EC(50) of 19 and 11 nM in the caspase activation assay in T47D breast cancer cells, respectively. The 2-amino-4-aryl-3-cyano-7-(dimethylamino)-4H-chromenes also were found to be highly active in the growth inhibition MTT assay, with GI(50) values in the low nanomolar range for compound 1c. Significantly, compound 1c was found to have a GI(50) value of 2 nM in the paclitaxel resistant, p-glycoprotein overexpressed, MES-SA/DX5 tumor cells. Functionally, compound 1c was found to be a potent inhibitor of tubulin polymerization and to effectively inhibit the binding of colchicine to tubulin. These results confirm that the cell-based caspase activation assay is a powerful tool for the discovery of potent apoptosis inducers and suggest that the 4-aryl-4H-chromenes have the potential to be developed into future anticancer agents.

Lung injury and mortality with hyperoxia are increased in peroxiredoxin 6 gene-targeted mice.

Overexpression of peroxiredoxin 6 (Prdx6) has been shown to protect lungs of mice against hyperoxia-mediated injury. In this study, we evaluated whether genetic inactivation of Prdx6 in mice increases sensitivity to oxygen toxicity. We evaluated mouse survival, lung histopathology, total protein and nucleated cells in bronchoalveolar lavage fluid (BALF), and oxidation of lung protein and lipids by measurement of protein carbonyls and thiobarbituric reactive substances (TBARS), respectively. The duration of survival for Prdx6 -/- mice was significantly shorter than that observed in wild-type mice on exposure to 85 or 100% O(2); survival of Prdx6 +/- mice was intermediate. After 72-h exposure to 100% O(2), lungs of Prdx6-/- mice showed more severe injury than wild-type with increased wet/dry weight, epithelial cell necrosis and alveolar edema on microscopic examination, increased protein and nucleated cells in BALF, and higher content of TBARS and protein carbonyls in lung homogenate. These findings show that Prdx6 -/- mice have increased sensitivity to hyperoxia and provide in vivo evidence that Prdx6 is an important lung antioxidant enzyme.

[Clinical report of cervical arthroplasty in management of spondylotic myelopathy].

OBJECTIVES: To investigate clinical effects and manual operational point of Bryan cervical disc prosthesis in Asian, to observe the stability and range of movement (ROM) postoperatively. METHODS: Bryan disc prosthesis replacement applied in 8 cases (10 levels) of cervical spondylotic myelopathy (CSM). Clinical (JOA grade and Odom's scale) and radiological (X-ray of bending, extending; left and right bending position) follow-up was performed 3 months postoperatively, (mean follow-up 3.6 month). Systemic radiographic study about stability and ROM of replaced level post operationally were measured. CT or MRI scan were applied in all cases 3 months postoperatively to find out the excursion of the prosthesis and heterotopic-ossification in the replaced levels. RESULTS: At least 3 months follow-up were done in all this patients. There was no complication. Improvement in all of 8 patients according to the Odsm's scale. JOA score increased from average 8.8 to 15.8. There was no prosthesis subsidence or excursion. Replaced segment achieved stability and restored partial of normal ROM, 4.75 degrees (3.8 degrees approximately 5.8 degrees ) in flex and extension position and 3.38 degrees (2.3 degrees approximately 4.4 degrees ), 3.13 degrees (2.5 degrees approximately 4.1 degrees ) in left and right bending position. No obvious loss of lordosis was found. CT or MRI follow-up show excursion (<2 mm) in 1/10 levels; (<2 mm excursion <4 mm) in 1/10 levels, and no heterotopic ossification in the replaced levels. CONCLUSIONS: Byran cervical disc prosthesis restored motion to the level of the intact segment in flexion-extension and lateral bending in post operational images. At the same time, it can achieve good anterior decompression treatment effect and immediate stability in replaced 1 or 2 levels, and which is a new choice for the treatment of CSM in Asian.
