[Selection of low instrumentation vertebrae with CD hybrid technique in king-types III and IV scoliosis].

OBJECTIVE: To review the King-types III and IV patients treated by the CD hybrid technique and evaluate clinical results on the shorter fusion levels. METHODS: Fifty-eight patients with idiopathic scoliosis were treated by the CD hybrid method from March 2000 to January 2003, among whom 40 were grouped as King-type I and 18 as King-type N; 41 were female and 17 were male. The Cobb angle of the thoracic curve was averaged 64 degrees (range 50-83 degrees), and the curve flexibility was 62%. The compensative lumbar curve was averaged 37 degrees (range 16-48 degrees), and the curve flexibility was 105%. With the neutral rotational vertebrae as a basis to select the low instrumentation vertebrae, the neutral rotational vertebrae or the vertebrae at 1 or 2 levels proximal to the neutral rotational vertebrae were selected as the low instrumentation vertebrae in all the patients. Standing AP and lateral radiographs were taken respectively at the discharge, during the follow-up after discharge, and at the final follow-up. RESULTS: The patients were followed up for an average of 2. 4 years (range 1.8-3.2). The corrected curves lost an average of 3.1 degrees (range--1-5 degrees)and the correction rate of the thoracic curve was 68% at the final follow-up. The plumb line from C7 was parallel to the sacral midline in 56 patients. The lumbar curves were corrected to an average of 8 degrees (2-13 degrees) automatically. The lumbosacral angle was corrected automatically by 53% and the low instrumentation vertebrae in 48 patients turned into stable vertebrae. The low instrumentation vertebrae lost 1.4 segments on average compared with the Harrington principle. No spinal imbalance was clinically observed in all the patients. CONCLUSION: The choice of the low instrumentation vertebrae as the neutral rotational vertebrae can have a good result in the clinical practice. It can be applied in the CD hybrid technique in treatment of idiopathic thoracic curves.

The ex vivo microenviroments in MLTC of poorly immunogenic tumor cells facilitate polarization of CD4+CD25+ regulatory T cells.

CD4+CD25+ regulatory T (TR) cells play an important role in maintaining a balanced peripheral immune system. Recent studies have shown that TR cells may also play a key role in suppressing anti-tumor immune response. In order to investigate the tumor immune microenvironment and its influence on TR polarization, poorly immunogenic tumor cell line D5 (C57BL/6, H-2b), immunogenic tumor cell lines FBL3 (C57BL/6, H-2b) and H22 BALB/c, H-2d) were used to establish the syngeneic/allogeneic, poorly immunogenic/immunogenic mixed lymphocytes-tumor cell culture (MLTC). Our results revealed that the proportion of CD4+CD25+ T cells in MLTC of syngeneic primed splenocytes stimulated with D5 tumor cells was higher than that with H22 cells (0.43% vs 0.044%, and the similar results appeared in allogeneic splenocytes stimulated with D5 tumor cells (0.39% vs 0.04%). The splenocytes stimulated with supernatant from syngeneic MLTC of D5 tumor cells demonstrated higher proportion of CD4+CD25+ cells than that from allogeneic MLTC of D5 tumor cells, and the splenocytes stimulated with supernatant from syngeneic or allogeneic MLTC of H22 tumor cells generated lower proportion of CD4+CD25+ T cells than that of D5 tumor cells. The TGF-beta1 and Th2-oriented cytokines (IL-4 and IL-10) were dominated in supernatants of syngeneic MLTC of poorly immunogenic tumor cells. Our results provided useful information for studying the mechanisms underlying tumor immune surveillance as well as for the tumor immunotherapy.

[The expression of TLR4 in rat acute lung injury induced by lipopolysaccharide and the influence of Eucalyptus globulus oil].

OBJECTIVE: To observe the distribution of toll-like receptor 4 (TLR4) in rats' respiratory tract. To study the influence of LPS and Eucalyptus globulus oil on the distribution of TMR4. METHOD: The Sprague-Dawley rats were intratracheally instilled with lipopolysaccharide (LPS,2 mg x kg(-1) per day) for two days to induce acute lung injury. The rats were sacrificed at 72 hours after LPS instillation. Lung morphology was studied. Leukocytes in Bronchoalveolar lavage fluid (BALF) were measured and TLR4 were detected by immunohistochemistry. RESULT: The result of immunohistochemistry showed that TLR4 distributed widely in common rats' respiratory tract. In the group of acute lung injury, the number of leucocyte in BALF was increased apparently, the inflammation in bronchus and bronchioles was more apparently than that of the control group in morphology. And the expression of TLR4 was reinforced in main bronchus and bronchioles. In the group of E. globules oil (300 mg x kg(-1)), the leucocyte number was decreased apparently in BALF, the inflammation was lightened and the expression of TLR4 decreased as compared with the group of models. CONCLUSION: The expression of TLR4 distributes widely in rats' respiratory tract. The stimulation of LPS can reinforce the expression of TLR4. The E. globules oil can reduce the increase of TLR4 induced by LPS in bronchioles.

[The phagocytosis of apoptotic allogeneic lymphocytes by recipient liver antigen-presenting cells].

OBJECTIVE: To track the location of the transfused apoptotic allogeneic lymphocytes and asses the process of their accumulation and phagocytosis removal as consequences on allograft tolerance in recipient mice. METHODS: Donor spleen lymphocytes were labeled by CFSE and induced to apoptosis by dexamethasone incubation. After purification by anti-annexin V-conjugated magnetic beads isolation, apoptotic lymphocytes were transfused into recipient mice through the tail veins. Tissue samples from various organs were taken at various time points to analyze the fates of the apoptotic allogeneic lymphocytes and the phagocytosis of them by organ resident APCs. RESULTS: Using fluorescent microscopy and flow cytometry, after the apoptotic cells were recognized and uptaken, the largest amount of labeled cells were accumulated in the livers and disappeared within not more than 12 hours. Recipient liver APCs were highly efficacious in phagocytosis of apoptotic allogeneic lymphocytes; the removal was completed within 15 minutes after incubation. LSEC, KC and LDC all phagocytosized the apoptotic allogeneic lymphocytes but with significantly different rates. Considering the numbers of those cells in a normal liver, it could be calculated that LSEC and KC had greater effects in this activity. CONCLUSIONS: The liver deserves foremost attention for study of the mechanism of allografts tolerance induced by pre-transfusion of apoptotic donor spleen lymphocytes. LSEC and KC are the main functional APCs to the alloantigens.

[The relationship between arousal and nocturnal heart rate variability in patients with obstructive sleep apnea-hypopnea syndrome].

OBJECTIVE: To investigate the relationship between arousal and nocturnal heart rate variability (HRV) in patients with obstructive sleep apnea-hypopnea syndrome (OSAHS). METHODS: Twenty-seven patients with moderate to severe OSAHS who underwent overnight polysomnography (PSG) were enrolled, excluding patients with cardiopulmonary and nervous system diseases. An hour continuous PSG monitoring in non-rapid eye movement (NREM) was recorded for all patients. The mean heart rate (HR) in 10 s before each arousal and the peak HR in 10 s after arousal were compared, and the breathing disorder related arousal index (B-ArI) and pulse rate rise index (PRRI) were calculated at this time, and then a correlation analysis was performed. Furthermore, according to minimal oxygen saturation (minSaO(2)), 18 patients with moderate-to-severe OSAHS were matched into 10 events with and without EEG arousal in NREM, and then DeltaHR at apnea termination between these events were compared. RESULTS: The peak HR in 10 s after arousal onset [(81.6 +/- 9.4) beats/min] was significantly higher than the mean HR in 10 s before arousal onset [(69.6 +/- 7.3) beats/min, t = -14.87, P < 0.01], and B-ArI was positively correlated to PRRI (r = 0.97, P < 0.01). The DeltaHR of events with EEG arousal at apnea termination [(11.1 +/- 2.8) beats/min] was higher than those without EEG arousal [(7.0 +/- 2.4) beats/min, t = 4.702, P < 0.01]. CONCLUSIONS: The results indicate that arousal can influence the function of cardiovascular regulation. Frequent arousal is one of the main causes of the cardiovascular consequences in OSAHS patients.

[In vivo tracing of transferred apoptotic cell labeled using CFSE: a flow cytometry-based assay method].

OBJECTIVE: To establish an assay method for detecting the migration of transferred apoptotic cells into the recipient using flow cytometry. METHODS: Spleen lymphocytes were isolated and labeled with an intracellular amine dye, carboxyfluorescein diacetate succinimidyl ester (CFSE), to allow discrimination. The labeled cells were induced with dexamethasone to undergo apoptosis and transferred into recipient mice via tail venous transfusion. Flow cytometry and histological examination of different tissues were performed at different time points. The stability of CFSE labeling for apoptotic cells was also tested. RESULTS: The CFSE-labeled apoptotic cells were highly fluorescent with a positive labeling rate of (98.0+/-1.9)%. The stability of CFSE-labeling was testified, and the CFSE-labeled apoptotic cells entering different tissues at different time points were detected by flow cytometry and verified by histological examination. CONCLUSION: Flow cytometry using CFSE labeling is reliable, sensitive, precise and convenient for apoptotic cell tracing in vivo and in vitro.

[Expression of cell adhesion molecule and nitric oxide synthase in nasal mucosa in allergic rhinitis].

OBJECTIVE: To explore the role of cell adhesion molecules (CAMs) and nitric oxide synthase (NOS) in the pathological mechanism of allergic rhinitis. METHOD: Three kinds of CAMs, which are intercellular adhesion molecule-1 (ICAM-1), vascular cellular adhesion molecule-1 (VCAM-1) and lymphocyte function antigen-1 (LFA-1), as well as three kinds of NOS, which are neuronal nitric oxide synthase (nNOS), inducible isoform of nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (eNOS) were orthotopic detected by strept avidin-biotin complex (SABC) immunohistochemistry method in 40 cases of inferior nasal concha of allergic rhinitis. RESULT: There are only small number of ICAM-1, VCAM-1 and LFA-1 positive cells in the normal concha mucosa epithelial but more positive cells in allergic rhinitis. The number of positive cell of ICAM-1, LFA-1, VCAM-1 in the concha mucosa was [(14.4+/-2.2), (17.2+/-3.3), (11.5+/-2.7) per scope (x400) +/-s] respectively; in the control group the number of positive cell of ICAM-1, LFA-1, VCAM-1 in the concha mucosa was (8.7+/-1.8), (10.3+/-2.1), (6.9+/-1.8) respectively. The statistic differences were significant (t = 11.57, 10.02, 8.07 respectively; P<0.01). A certain number of nNOS positive cells were found in the concha mucosa in allergic rhinitis patients, it was mainly expressed in mucosa epithelial and submucosa glandular; a large number of iNOS positive cells were found in the concha mucosa in allergic rhinitis patients, iNOS was mainly expressed in mucosa epithelial, submucosa glandular, endothelial and submucosa inflammatory cell; the number of eNOS positive cell was few. In the normal control, few positive cells of nNOS, iNOS or eNOS could be found. Between the allergic rhinitis patients and the normal control, the number of nNOS positive cell was [(9.4+/-1.7), (4.7+/-1.3) per scope (x400)] respectively, t = 12.62 (P<0.01); the number of iNOS positive cell was (27.5+/-3.2), (4.3+/-1.7) respectively, t = 36.03 (P<0.01) the number of eNOS positive cell was (6.5+/-2.1), (6.2+/-1.9) respectively, t = 0.62 (P>0.05). CONCLUSION: These adhesion molecules might be involved in edema, exudation and infiltration of inflammatory cells of nasal mucosa in allergic rhinitis patients. The number of nNOS, iNOS positive cell in inferior concha mucosa in allergic rhinitis patients was significant higher than in normal control. It suggests that nNOS and iNOS might be involved in the pathogenesis of allergic rhinitis.

[Adverse effects of intravenous arsenic trioxide and their prevention].

OBJECTIVE: To study the main side effects of As(2)O(3) and the way of prevention. METHODS: The changes of body weight and various systems of body were observed after treatment with As(2)O(3) injection. The arsenic content in blood, urine and hair was detected with atom absorbed-spectrum analysis. RESULTS: Mild reactions were observed in 57.43% (147/256) of the patients and they could subside after cessation of arsenic treatment or allotherapy. Chronic mild poisoning manifestations including arsenic furuncle, liver dysfunction and peripheral nerve injury were found in 2.73% (7/256) of the patients and they subsided gradually after treatment. Chronic severe poisoning was found in 1.17% of the patients and all of them died of liver failure. As(2)O(3) might cause decrease of peripheral blood WBC in catabatic patients. There was no infection after allopathy without ceasing arsenic medication. The results showed that As(2)O(3) could distribute over and discharge from the plasma without accumulation in blood. The results also demonstrated the main route of excretion for As(2)O(3) is through urine and there is definite accumulation in the hair 50 days after treatment. CONCLUSION: Most of the side effects of As(2)O(3) are mild and recoverable. Allotherapy could be effective to relieve the complications without stopping arsenic medication. A few patients with complicating hepatitis may suffer from chronic poisoning. As(2)O(3) may cause increase of peripheral blood WBC in induced catabatic APL patients. The side effects must be prevented early.

[Genotype of wild-type measles viruses circulating in Beijing in 2003].

OBJECTIVE: To investigate genotype of wild-type measles viruses circulated in Beijing in 2003. METHODS: Throat swabs specimens were collected from patients seen during an outbreak of measles and from clinically suspected sporadic measles patients in 2003. Vero/SLAM cell lines recommended by WHO were used to isolate measles virus. Four hundreds and fifty nucleotides of COOH-terminal of nucleoprotein (N) genes were amplified by using PR-PCR. The amplified products were sequenced and the sequences were compared with references viruses from GeneBank. RESULTS: Eight strains of measles viruses were isolated from throat swabs of patients who came from seven districts and counties of Beijing. Sequence analysis of the 450 nucleotides of COOH-terminal of nucleoprotein (N) genes indicated that these 8 strains belonged to H1a genotype. The average genetic distances of these 8 strains to H1a genotype, Chin9322, H1b genotype, Chin9475 and H1c genotype, Chin9427, were 0.004 - 0.011, 0.026 - 0.031 and 0.015 - 0.022, respectively. The average genetic distances of these 8 strains to H1a genotype, Anhui 01 - 1/Anhui 02 - 2, were 0.000 - 0.009 (0 - 5 nucleotide variation). CONCLUSIONS: Major genotypes of wild-type measles viruses circulated in Beijing in 2003 were H1a genotype. The genotypes H1c, H1b and H2 may have disappeared in Beijing.

[Regularity of sugar-uptake in human red blood cells].

Lyophilization of human red blood cells has important significance in clinical application. Some sugars, especially trehalose, can be more tolerant of some organism or cells to dry environments, But, how to bring sugars into cells is a challenge. This study was aimed to investigate the regularity of sugar-uptake in human red blood cells. The absorption rate of trehalose and glucose in red blood cells, free hemoglobin level and erythrocyte deformation index were determined at different incubation temperature (4, 25 and 37 degrees C), different sugar concentration (0, 0.2, 0.4, 0.6, 0.8 and 1 mol/L) and different incubation time (1, 3, 5, 7 and 9 hours). The results showed that with increase of temperature and extracellular sugar concentration, the uptake of sugar in red blood cells also increased, the intracellular trehalose and glucose concentrations were over 30 mmol/L and 40 mmol/L respectively. The effects of incubation time on uptake of trehalose and glucose were different. With prolonging of incubation time, the uptake of trehalose showed firstly increase and then decrease, however, the uptake of glucose showed a constant increase. But the loading process had side-effect on free hemoglobin and maximum deformation index (MAXDI) of red blood cells, especially for trehalose, which mainly come from high osmotic pressure. It is concluded that the uptake of sugars in red blood cells is closely dependent on incubation temperature, extracellular sugar concentration and incubation time. In certain condition, the efficiency of sugar uptake is very high, but this process also damages red blood cells so as to affect the application of sugars in lyophilization of red blood cells. The research in the future should focus on how to deal with the relation between cell injury and uptake efficiency of sugar in red blood cells.

[Study on dynamic spectrum and its frequency domain extracting method].

For non-invasive blood components detection using near-infrared spectrum, eliminating the influence of the individual difference is very important. In the non-invasive component measurement, the individual discrepancy refers to the difference of personal feature in measured tissues except the pulsatile component of artery blood, including hairs, horny layer, subcutaneous tissue, muscle, skeleton etc. It is declared that the individual discrepancy is different from the personal feature; it is an important factor blocking the practice of the technology. In the present article, a new method of detecting blood components is presented, which is based on the principle of photoplethysmography and Fourier transform. With this method, the influence of individual difference caused by skin and other tissues can be mostly eliminated, and the absorbance of artery blood could be obtained. The concept of the frequency domain extracting method was presented. The experiment was done and the dynamic spectrum which only evaluates the pulsatile part of the entire optical signal was obtained. The advantage of dynamic spectrum with frequency domain extracting method is analyzed. The result shows that the method can be realized and has many advantages.

[Analysis of volatile constituents in leaves of three cypress species by gas chromatography/mass spectrometry].

Volatile oils in leaves of three cypress species were extracted by hydrodistillation method and analyzed by gas chromatography/mass spectrometry. Sixty-four constituents were separated and identified. The majority of the components were found to be monoterpenes and sesquiterpenes. There were 33, 37, 37 compounds in the oils of leaves of Sabina chinensis (L.) Ant, Cupressus lusitanica 'zhongshan' Mill and Sabina chinensis (L.) Ant. Cv. Kaizuca, respectively. Fourteen compounds were common to them, which were thujene, alpha-pinene, camphene, sabinene, beta-myrcene, alpha-terpinene, gamma-terpinene, alpha-terpinolene, bornyl acetate, beta-elemene, alpha-amorphene, germacrene D, delta-cadinene and elemol. In addition, each species had particular compounds. The main components were found to be: sabinene (20.99%), limonene (19.78%) and bornyl acetate (11.68%) for Sabina chinensis (L.) Ant; alpha-pinene (10.39%), sabinene (11.19%) and delta-3-carene (8.88%) for Cupressus lusitanica 'zhongshan' Mill; limonene (24.56%), beta-myrcene (8.04%) for Sabina chinensis (L.) Ant. Cv. Kaizuca.

[Construction and study of replication-defective adenovirus targeting hepatocarcinoma].

BACKGROUND & OBJECTIVE: Antioncogene p16 is one of the most important genes used in tumor gene therapy. Apoptosis induced by adenovirus mediated overexpression of p16 in cancer cells has been confirmed in various p16 gene inactive cancers. Studies have indicated that p16 gene is frequently inactive in human primary hepatocarcinoma. Therefore this study was to investigate the effect of exogenous p16 gene driven by alpha fetoprotein (AFP) core promoter on human hepatocellular carcinoma cells and further explore the potentials of p16 in hepatocellular carcinoma gene therapy. METHODS: The recombinant replication-defective adenovirus Ad-AFP-p16 containing p16 gene downstream the AFP core promoter-AF0.3 was constructed and infected hepatocellular carcinoma cells. The expression of p16 was detected by Western blot. The effects of exogenous p16 gene on cell growth and apoptosis were measured by MTT, flow cytometry and DNA ladder in vitro. Subcutaneous injection of mouse hepatocarcinoma cell line H22 infected with Ad-AFP-p16 was applied to observe the effect of Ad-AFP-p16 on tumorigenesis in vivo. RESULTS: Over-expression of exogenous p16 gene was confirmed in hepatocarcinoma cells infected with Ad-AFP-p16. Cell growth was inhibited by (94.42+/-11.70)% and (94.99+/-6.74)% in Be1-7402 and HepG2 cells on the 6th day after virus infection; 39.57% and 39.75% apoptotic cells were also induced respectively in these two cell lines on the 2nd day. Moreover, the infection of Ad-AFP-p16 significantly inhibited the tumorigenesis of mouse hepatocarcinoma cell line H22 in vivo. The tumor volumes of control, Ad-GFP, Ad-AFP-p16 and Ad-CMV-p16 groups were (1.54+/-0.65)cm(3), (1.71+/-1.01)cm(3), (0.25+/-0.39)cm(3) and (0.25+/-0.45)cm(3), respectively. CONCLUSION: The expression of p16 gene driven by AFP promoter can induce apoptosis in hepatocarcinoma cells.

[Bezafibrate up-regulate endothelial nitric oxide gene expressions via peroxisome proliferator-activated receptors alpha-dependent and independent pathways in cultured Bovine endothelial cells].

OBJECTIVE: To investigate the effects and related mechanisms of bezafibrate, a ligand of peroxisome proliferator-activated receptors alpha (PPARalpha), on endothelial nitric oxide synthase (eNOS). METHODS: Firstly, in cultured bovine aorta endothelial cells (BAEC), the effects of bezafibrate on eNOS mRNA and protein levels were investigated by Northern blot and Western blot. The half-life of eNOS mRNA and NO production determined from BAECs treated with bezafibrate were performed by real-time quantitative PCR and NO assay. Next, the effects of bezafibrate on signal pathways in BAEC, through inhibitors of PPARalpha, PI3 kinase and MAPK, were investigated by Western blot. Then luciferase reporter gene driven by human eNOS promoter were cloned and transfected endothelial cells to see the effects of bezafibrate on eNOS promoter-driven luciferase activity. RESULTS: In cultured BAEC, bezafibrate significantly upregulated eNOS expressions at protein and mRNA levels in a concentration-dependent fashion (50 - 200 micromol/L) (P < 0.05), and increased nitric oxide (NO) production, respectively[control (14.97 +/- 1.29) micromol/L, different concentration groups (25.12 +/- 1.25) micromol/L, (30.12 +/- 1.85) micromol/L, (33.47 +/- 1.22) micromol/L], and enhanced phosphorylation of eNOS-ser-1179 site (P < 0.05), but not eNOS-thr-497 site. Through real-time quantitative PCR, bezafibrate prolonged eNOS mRNA half-life from 3.1 hour to 6.1 hour were demonstrated. Further studies showed that bezafibrate treatments significantly enhanced dose-dependently luciferase activity (relative luciferase activity in 100 micromol/L and 200 micromol/L groups 4429.43 +/- 391.41 and 6187.5 +/- 307.53), compared with untreated luciferase reporter gene group (3361.81 +/- 316.85) (P < 0.05 and P < 0.01, respectively), and induced MAPK phosphorylation expression (P < 0.05 and P < 0.01, respectively). Then these effects of bezafibrate upregulated eNOS expressions could be blocked by PPARalpha antagonist, MAPK and PI3K inhibitor while not affected by PKC and MEK inhibitor (P < 0.01). CONCLUSIONS: Bezafibrate can upregulate eNOS expression, enhance eNOS-ser-1179 site phosphorylation, increase NO production and stabilize eNOS mRNA through PPARalpha dependent pathway and PPARalpha independent MAPK and PI3K pathways. This mechanism provides additional anti-atherosclerotic and anti-hypertension benefits of bezafibrate in addition of lipid-lowering effects.

[Preliminary study on self-concept psychological feature of cleft lip and palate adolescents].

OBJECTIVE: To obtain the self-concept psychological feature of cleft lip and palate (CLP) adolescents further and accurately, and offer reasons for timely and pointed psychological intervention. METHODS: Based on the procedure and criteria of development of psychological questionnaire, a psychological questionnaire for CLP adolescents was designed. 93 CLP and 88 healthy adolescents were studied by using it and their half-quantified score raging 0-9 were analyzed statistically. RESULTS: (1) There was no statistical difference between CLP and normal adolescents, scored 5.91 and 5.66 respectively, which demonstrated that the self-concept of CLP adolescents was at a normal level without obvious passive tendency. (2) There was no significant difference between genders, but boys were more extroversion while girls did better on being unyielding and releasing strain. (3) CLP adolescents had a higher ability of psychological regulation and could face failure more bravely. CLP patients were in a tendency of protective behavior inhibition, companied less, and maximized delight mood unconsciously to decrease harm from enviroment. (4) CLP adolescents had a good parentage. (5) A majority of patients had low expectation of life-quality. (6) Much of CLP patients considered that speech was more important than appearance. CONCLUSION: Self-concept of CLP adolescents have normal self-concept and do better in psychological regulation and dependence than normal ones.

[Ways to improve measurement accuracy of blood glucose sensing by mid-infrared spectroscopy].

Mid-infrared (MIR) spectroscopy is applicable to blood glucose sensing without using any reagent, however, due to a result of inadequate accuracy, till now this method has not been used in clinical detection. The principle and key technologies of blood glucose sensing by MIR spectroscopy are presented in this paper. Along with our experimental results, the paper analyzes ways to enhance measurement accuracy and prediction accuracy by the following four methods: selection of optimized spectral region; application of spectra data processing method; elimination of the interference with other components in the blood, and promotion in system hardware. According to these four improving methods, we designed four experiments, i.e., strict determination of the region where glucose concentration changes most sensitively in MIR, application of genetic algorithm for wavelength selection, normalization of spectra for the purpose of enhancing measuring reproduction, and utilization of CO2 laser as light source. The results show that the measurement accuracy of blood glucose concentration is enhanced almost to a clinical detection level.

[Expression and significance of X-linked inhibitor of apoptosis protein and its antagonized proteins in acute leukemia].

To investigate the expression and significance of X-linked inhibitor of apoptosis protein (XIAP) and XIAP-associated factor 1 (XAF1) in acute leukemia, the expression of XIAP, XAF1, Smac, and HtrA2 mRNA in the bone marrow aspirates from 87 newly diagnosed AL patients, 23 patients in remission, 6 patients in relapse, and 17 normal controls were detected by means of reverse transcriptase polymerase chain reaction (RT-PCR), and their relationship with clinical therapeutic efficiency was analyzed. The results showed that the expression level of XIAP mRNA in newly diagnosed AL patients (0.990 +/- 0.337) was significantly higher than that in normal controls (0.395 +/- 0.148) (P < 0.01); the positive rate and expression level of XAF1 mRNA in newly diagnosed AL patients (56.32%, 0.246 +/- 0.267) were significantly lower than that in normal controls (100%, 0.964 +/- 0.387) (P < 0.01). In 69 out of 87 newly diagnosed AL patients, efficacy remained evaluable. AL patients with high level of XIAP achieved a lower complete remission (CR) rate than patients with low level of XIAP (54.55% and 86.11%, respectively) (P < 0.01). XAF1 positive patients achieved a higher CR rate than XAF1 negative patients (86.84% and 51.61%, respectively) (P < 0.01). It is concluded that the overexpression of XIAP and negativity of XAF1 may be two adverse prognostic factors in AL patients.
