Multi-omics identification of ZFPM2 and CD44 as key candidates for testicular size in sheep.

Testicular size is a key determinant of ram fertility, yet its genetic architecture in sheep remains poorly understood. Crossbred sheep exhibit substantial testicular developmental variation and serve as ideal models for screening fertility-related genes. Here, we performed whole-genome sequencing (WGS)-based genome-wide association study (GWAS) on 115 rams, including seven crossbred populations (each derived from a distinct sire breed crossed with Hu sheep) and a purebred Hu sheep population. We identified two variants within ZFPM2, including an intronic single nucleotide polymorphism (SNP) rs421073404 and a missense SNP rs1086332841, both significantly correlated with testis weight, length, and width. Importantly, these variants exerted testis-specific effects, consistent with their weak correlation with body weight (r&#x202f;<&#x202f;0.2). Genomic selection scans (FST and &#x3c0;-ratio) between rams with large (>150&#x202f;g/side) and small (<75&#x202f;g/side) testes revealed multiple divergent genomic regions, with prominent haplotype differentiation at the CD44 locus on chromosome 15. Notably, two linked missense variants (rs160734053 and rs414318703) in CD44 exerted opposing effects on testicular size, suggesting allelic heterogeneity at this locus. Integrative RNA-seq and ATAC-seq across 0-12 months further uncovered prepubertal stage-specific expression and chromatin accessibility patterns of ZFPM2 and CD44, providing mechanistic insights into their regulatory roles. Collectively, this study provides candidate SNPs for sheep marker-assisted selection and multi-omic evidence for dissecting the genetic basis of testicular development in ovine breeding.

Diesel exhaust exposure favors TH2 cell recruitment in nonatopic subjects by differentially regulating chemokine production.

BACKGROUND: The prevalence of allergic diseases has increased in the last 20 years, and a number of studies have shown that diesel exhaust particle-associated polyaromatic hydrocarbons can exacerbate the allergic reaction. Much less is known about their potential capacity to generate a T(H)2-type allergic reaction in nonatopic subjects. OBJECTIVE: The aim of this study was to test the hypothesis that diesel exhaust exposure might favor, in nonatopic donors, T(H)2-type cell recruitment, either through increased production of T(H)2-attracting chemokines or decreased production of T(H)1-attracting chemokines. METHODS: PBMCs from nonatopic donors were incubated with diesel exhaust particle-polyaromatic hydrocarbons, and the supernatants were evaluated for the presence of pro-T(H)1 chemokines (IFN-gamma-induced protein 10 and monokine Induced by IFN-gamma) and pro-T(H)2 chemokines (macrophage-derived chemokine, I-309, and pulmonary and activation-regulated chemokine) by means of ELISA. The functional effect was evaluated by using chemotaxis assays with polarized T(H)1 and T(H)2 cells. RESULTS: Diesel exhaust exposure of PBMCs from nonatopic donors induced a late increase after 48 hours in pulmonary and activation-regulated chemokine mediated by IL-13 and a decrease in IFN-gamma-induced protein 10 levels selectively at both the protein and mRNA levels. The functional effect of these chemokine variations resulted in an enhanced chemotaxis of T(H)2, but not T(H)1, cells. CONCLUSION: These findings show that diesel exhaust exposure might be involved in the genesis of allergic diseases by differentially regulating chemokines favoring the recruitment of T(H)2 cells in nonatopic subjects. CLINICAL IMPLICATIONS: Environmental factors, especially air pollution, might favor the genesis of allergic diseases.

CCR5 usage by CCL5 induces a selective leukocyte recruitment in human skin xenografts in vivo.

CCR5 is one of the major inflammatory chemokine receptors with potential therapeutical applications in humans. However, the redundancy of chemokines and their receptors, and the species specificity of chemokine receptor antagonists pose challenges to understanding of the role they play in pharmacological situations. To address this question, we used a humanized severe combined immunodeficient mouse model grafted with human skin and autologous leukocytes, and evaluated the effect of a blocking antibody against human CCR5, on CCL5-induced cutaneous leukocyte recruitment in vivo. At baseline, CCL5 induced a significant recruitment of T cells mainly of the memory phenotype, of monocytes/macrophages, eosinophils, and IFN-gamma(+) but not IL-4(+) and IL-5(+) cells. In vivo, anti-CCR5 antibody was able to almost completely inhibit the recruitment of monocytes/macrophages and T-helper (Th)1-type cells to inhibit partially the attraction of memory T cells, but had no effect on eosinophil infiltration, although all these cell types express other CCL5 binding chemokine receptors than CCR5. These results indicate that the in vivo environment regulates target cell specificity of CCL5 leading to differential cell recruitment, suggesting that antagonizing CCR5 receptor may be of therapeutic value in diseases such as acquired immuno deficiency syndrome, where CCL5/CCR5, monocytes, and Th1-type cells play a predominant role.

Involvement of CCL18 in allergic asthma.

Allergic asthma is associated with a pulmonary recruitment of Th type 2 cells, basophils, and eosinophils, mainly linked to chemokine production. CCL18 is a chemokine preferentially expressed in the lung, secreted by APCs, induced by Th2-type cytokines, and only present in humans. Therefore, CCL18 may be involved in allergic asthma. PBMC from asthmatics allergic to house dust mite cultured in the presence of Dermatophagoides pteronyssinus 1 (Der p 1) allergen secreted CCL18, 48 and 72 h after stimulation, whereas those from healthy donors did not. Part of CCL18 was directly derived from Der p 1-stimulated plasmacytoid dendritic cells, whereas the other part was linked to monocyte activation by IL-4 and IL-13 produced by Der p 1-stimulated T cells. In bronchoalveolar lavages from untreated asthmatic allergic patients, CCL18 was highly increased compared with controls. Functionally, CCL18 preferentially attracted in vitro-polarized Th2 cells and basophils, but not eosinophils and Th1 cells, and induced basophil histamine and intracellular calcium release. These data show a new function for CCL18, i.e., the recruitment of Th2 cells and basophils, and suggest that CCL18 may play a predominant role in allergic asthma.

Dichotomains A and B: two new highly oxygenated phenolic derivatives from Dicranopteris dichotoma.

[structure: see text] Dichotomains A (1) and B (2), two new highly oxygenated phenolic derivatives that feature a spirodilactone moiety in their structures, were isolated from the fronds of Dicranopteris dichotoma. Their structures were elucidated on the basis of NMR and MS spectroscopic data, and the stereochemistry of 1 was finally determined by single-crystal X-ray diffraction. Compound 2 showed weak anti-HIV-1 activity.

Sphenadilactones A and B, two novel nortriterpenoids from Schisandra sphenanthera.

[structure: see text] Two novel nortriterpenoid compounds, sphenadilactones A (1) and B (2), have been isolated from the leaves and stems of Schisandrasphenanthera. The structural elucidation of 1 and 2 was accomplished by extensive NMR analysis. The relative stereochemistry of 1 was established by single-crystal X-ray crystallography. Both compounds were tested for their cytotoxicities against K562, A549, and HT-29, and compound 1 was further tested for its anti-HIV-1 activity.

Difference of gene expression profiles between Barrett's esophagus and cardia intestinal metaplasia by gene chip.

The difference of gene expression profile changes in Barrett's esophagus (BE) and cardia intestinal metaplasia (CIM) epithelium was studied and the novel associated genes were screened in the early stage by cDNA microarray. The cDNA retro-transcribed from equal quantity mRNA from BE and CIM epithelial tissues were labeled with Cy3 and Cy5 fluorescence as probes. The mixed probe was hybridized with three pieces BiostarH-40s double dot human whole gene chip. The chips were scanned with a ScanArray 4000. The acquired images were analyzed using GenePix Pro 3.0 software. It was found a total of 141 genes were screened out that exhibited differentially expression more than 2 times in all three chips. It was identified that in gene expression profiles of BE, 74 genes were up-regulated and 67 down-regulated as compared with CIM. The comparison between the difference of gene expression profile changes in BE and CIM epithelia revealed that there existed the difference between BE and CIM at gene level. 141 genes with the expression more than two time were probably related to the occurrence and development of BE and the promotion or progress in adenocarcinoma.

Daphnioldhanins A-C, alkaloids from Daphniphyllum oldhami.

Three new Daphniphyllum alkaloids, daphnioldhanins A-C (1-3), along with four known ones, were isolated from the aerial parts of saplings of Daphniphyllum oldhami. The structures of these alkaloids were established by spectroscopic methods. The relative configuration of 1 was further confirmed by a single-crystal X-ray diffraction analysis. In addition, the (1)H and (13)C NMR data of the free base of daphmanidin A (4) were compared with its hydrochloric salt form.

[Expression of intestine-specific transcription factor CDX2 in different subtypes of intestinal metaplasia and gastric carcinoma].

BACKGROUND & OBJECTIVE: Intestinal metaplasia (IM) is thought as the precancerous lesion of gastric carcinoma, and CDX2 gene plays important roles in development and differentiation of intestinal epithelium, and maintenance of intestinal phenotype. Recent studies found that CDX2 were expressed aberrantly in IM of chronic atrophic gastritis (CAG) and some gastric carcinomas, which implied that CDX2 may play an important role in IM formation and gastric carcinogenesis. This study was to investigate the roles of CDX2 in the development and progression of IM and gastric carcinogenesis, and determine the correlation of IM to gastric carcinogenesis. METHODS: A tissue microarray containing 46 cases of CAG with IM, 40 cases of gastric carcinoma, and 32 cases of IM foci in paracancerous tissues was constructed. High iron diamine/alcian blue (HID/AB) and HE staining were used to classify IM and gastric carcinoma, and the expression of CDX2 protein and mRNA in different gastric lesions was assessed with immunohistochemistry and in situ hybridization, respectively. RESULTS: The proportion of type III IM was significantly higher in IM foci in paracancerous tissues than in CAG with IM (56.25% vs. 21.74%, P<0.01). The positive rates of CDX2 protein were 69.56% in IM foci in CAG, 53.13% in IM foci in paracancerous tissues, and 42.50% in gastric carcinomas, and the positive rates of CDX2 mRNA were 63.04%, 46.87%, and 35.00%, respectively. The positive rates were significantly lower in gastric cancer than in IM in CAG (P<0.01), but there was no significant difference between gastric cancer and IM foci in paracancerous tissues (P>0.05). The expression of CDX2 protein and mRNA was significantly higher in intestinal-type gastric cancer than in diffuse-type gastric cancer (54.55% vs. 27.78%, 45.45% vs. 22.22%, P<0.05). The expression of CDX2 protein was significantly lower in type III IM than in type I IM (46.42% vs. 79.31%, P<0.05). CONCLUSIONS: CDX2 may play important roles in the development and progression of IM and gastric carcinogenesis.

[A comparative study on seeding methods of human mesenchymal stem cells in bone tissue engineering].

Seeding is the crucial step in bone tissue engineering. In current study, static and dynamic seeding methods for human bone marrow stem cells (hBMSCs) were compared. The methods for assay of DNA content in the constructs after seeding were adopted. The optic microscopy for histological apearance and the scanning electron microscopy (SEM) and fluorescent RT-PCR for osteogenic markers were performed. The maximal initial seeding concentration in static seeding is lower than that in dynamic seeding. Histology and SEM revealed the even distribution and spreading of cells in the dynamically seeded constructs, but showed cell aggregation in the statically seeded counterparts. Fluorescent RT-PCR again revealed stronger osteogenic potential of dynamically seeded constructs. Therefore, this initial study demonstrated that dynamic seeding of human bone marrow stem cells is a promising technique in bone tissue engineering.

[Clinical observation on auricular point sticking for treatment of 25 children of semiluxation of circo-axis vertebrae].

OBJECTIVE: To search for the best method for child semiluxation of circo-axis vertebrae. METHODS: Fifty cases were randomly divided into a treatment group of 25 cases treated with auricular point sticking, and a control group of 25 cases treated with normal acupuncture. RESULTS: The total effective rate was 92.0% in the treatment group and 64.0% in the control group with a significant difference between the two groups (P < 0.05). CONCLUSION: Auricular point sticking has a good effect on child semiluxation of circo-axis vertebrae.

Distinction between short-segment Barrett's esophageal and cardiac intestinal metaplasia.

AIM: To investigate the roles of mucin histochemistry, cytokeratin 7/20 (CK7/20) immunoreactivity, clinical characteristics and endoscopy to distinguish short-segment Barrett's esophageal (SSBE) from cardiac intestinal metaplasia (CIM). METHODS: High iron diamine/Alcian blue (HID/AB) mucin-histochemical staining and immunohistochemical staining were used to classify intestinal metaplasia (IM) and to determine CK7/20 immunoreactivity pattern in SSBE and CIM, respectively, and these results were compared with endoscopical diagnosis and the positive rate of gastroesophageal reflux disease (GERD) symptoms and H pylori infection. Long-segment Barrett's esophageal and IM of gastric antrum were designed as control. RESULTS: The prevalence of type III IM was significantly higher in SSBE than in CIM (63.33% vs 23.08%, P< 0.005). The CK7/20 immunoreactivity in SSBE showed mainly Barrett's pattern (76.66%), and the GERD symptoms in most cases which showed Barrett's pattern were positive, whereas H pylori infection was negative. However, the CK7/20 immunoreactivity in CIM was gastric pattern preponderantly (61.54%), but there were 23.08% cases that showed Barrett's pattern. H pylori infection in all cases which showed gastric pattern was significantly higher than those which showed Barrett's pattern (63.83% vs 19.30%, P< 0.005), whereas the GERD symptoms in gastric pattern were significantly lower than that in Barrett's pattern (21.28% vs 85.96%, P< 0.005). CONCLUSION: Distinction of SSBE from CIM should not be based on a single method; however, the combination of clinical characteristics, histology, mucin histochemistry, CK7/20 immunoreactivity, and endoscopic biopsy should be applied. Type III IM, presence of GERD symptoms, and Barrett's CK7/20 immunoreactivity pattern may support the diagnosis of SSBE, whereas non-type III IM, positive H pylori infection, and gastric CK7/20 immunoreactivity pattern may imply CIM.

Correlations between edema and the immediate and prolonged painful consequences of inflammation: therapeutic implications?

The precise relationship between the degree of pain and the degree of inflammation in the individual remains debated. A quantitative analysis simultaneously applied to the immediate and prolonged painful consequences of inflammation has not yet been done. Thus, the correlations between edema, nociception and hypersensitivity following an inflammatory insult were assessed in rodents. To better understand the therapeutic value of modifying specific aspects of inflammation, the effects of an anti-inflammatory drug were compared to the results. Inbred strains of mice and outbred rats received an intraplantar injection of honeybee venom and the between-group and within-group correlations were calculated for spontaneous nociceptive measures, thermal and mechanical hypersensitivity, and edema and temperature. The effect of indomethacin on the pain and inflammation measures was examined. Edema correlated with spontaneous flinching, licking and lifting of the injected paw (P< or =0.003), and not with thermal or mechanical hypersensitivity. Indomethacin affected edema and spontaneous nociception dose-dependently, and affected hypersensitivity only at the highest dose tested (P< 0.05). These results suggest that edema may contribute only to immediate spontaneous nociceptive responses to an inflammatory insult, and not to the more clinically relevant prolonged hypersensitivity. This analysis represents a method for determining which inflammatory processes are the most promising therapeutic targets against the multiple painful consequences of inflammation.

Effects of interferon-alpha on expression of hepatic stellate cell and transforming growth factor-beta1 and alpha-smooth muscle actin in rats with hepatic fibrosis.

AIM: To investigate the effect of interferon-alpha (IFN-alpha) on preventing or reversing hepatic fibrosis in rat experimental model induced by CCl(4). METHODS: One hundred and ten Sprague-Dawley rats were divided into five groups: group A (normal controls, n=18), group B (fibrotic model controls, n = 22), group C (IFN-alpha prevention, n=22) initially treated with intra-muscular injection of IFN-alpha in saline daily at the doses of 1X105 U for 6 wk, group D (IFN-alpha treatment, n = 24) treated with intra-muscular injection of IFN-alpha in saline daily at the doses of 1X105 U for 6 wk after the first 6 wk, group E (0.9% sodium chloride treatment control, n = 24) treated with intra-muscular injection of 0.01 mL/kg daily for 6 wk after the first 6 wk. At the end of the experiment, all rats of each group were killed. Samples of the liver obtained by biopsy were subjected to histological, immunohistochemical and electron microscopic studies for the expressions of transforming growth factor-beta1 (TGF-beta1) and alpha-smooth muscle actin (alpha-SMA). RESULTS: The expressions of TGF-beta1, the number of activated hepatic stellate cells and alpha-SMA in hepatic tissue of group C were significantly less than those of group B (P<0.01). The degree of fibrosis score in group B was also significantly less than that of group C under light microscope (P<0.01). CONCLUSION: IFN-alpha can inhibit the production of TGF-beta1, decrease HSC activation and stimulate its apoptosis.

A systematic heuristic approach for feature selection for melanoma discrimination using clinical images.

BACKGROUND: Numerous features are derived from the asymmetry, border irregularity, color variegation, and diameter of the skin lesion in dermatology for diagnosing malignant melanoma. Feature selection for the development of automated skin lesion discrimination systems is an important consideration. METHODS: In this research, a systematic heuristic approach is investigated for feature selection and lesion classification. The approach integrates statistical-, correlation-, histogram-, and expert system-based components. Using statistical and correlation measures, interrelationships among features are determined. Expert system analysis is performed to identify redundant features. The feature selection process is applied to 19 shape and color features for a clinical image data set containing 355 malignant melanomas, 125 basal cell carcinomas, 177 dysplastic nevi, 199 nevocellular nevi, 139 seborrheic keratoses, and 45 vascular lesions. RESULTS: Experimental results show reduced lesion classification error rates based on condensing the shape and color feature set from 19 features to 13 features using the feature selection process. Specifically, average test lesion classification error rates for discriminating malignant melanoma from non-melanoma lesions were reduced from 26.6% for 19 features to 23.2% for 13 features over five randomly generated training and test sets. CONCLUSIONS: The experimental results show that the systematic heuristic approach for feature reduction can be successfully applied to achieve improved lesion discrimination. The feature reduction technique facilitates the elimination of redundant information that may inhibit lesion classification performance. The clinical application of this result is that automated skin lesion classification algorithm development can be fostered with systematic feature selection techniques.

Flocculation control study based on fractal theory.

A study on flocculation control based on fractal theory was carried out. Optimization test of chemical coagulant dosage confirmed that the fractal dimension could reflect the flocculation degree and settling characteristics of aggregates and the good correlation with the turbidity of settled effluent. So that the fractal dimension can be used as the major parameter for flocculation system control and achieve self-acting adjustment of chemical coagulant dosage. The fractal dimension flocculation control system was used for further study carried out on the effects of various flocculation parameters, among which are the dependency relationship among aggregates fractal dimension, chemical coagulant dosage, and turbidity of settled effluent under the conditions of variable water quality and quantity. And basic experimental data were obtained for establishing the chemical coagulant dosage control model mainly based on aggregates fractal dimension.

[Study on multipotential differentiation of human bone marrow mesenchymal stem cells in vitro].

OBJECTIVE: To study the capability of human bone marrow mesenchymal stem cells (MSCs) to differentiate into adipocyte, osteoblasts, and neurocytes under different experimental conditions in vitro. METHODS: The induced human bone marrow MSCs were examined by cytochemistry staining and immunohistochemistry staining. RESULTS: Induced human bone marrow MSCs formed alkaline phosphatase-positive aggregates and Von Kossa stain-positive nodules under the condition of osteogenic differentiation. Under the condition of adipogenic differentiation, the isolated human bone marrow MSCs formed oil red-O-positive cells. Immunohistochemistry results showed that differentiated human bone marrow MSCs expressed neuron-specific enolase (NSE) and neurofilament (NF-M). CONCLUSION: Human bone marrow MSCs have the capability of multipotential differentiation in vitro.

[Study on association between gestational diabetes mellitus and sulfonylurea receptor-1 gene polymorphism].

OBJECTIVE: To investigate allelic frequency of sulfonylurea receptor-1 (SUR1) in gestational diabetes mellitus (GDM) women of Han nationality in north China in order to find out susceptible gene associated with GDM, and to assess the association between SUR1 allele and body mass index (BMI) and secretion of insulin. METHODS: Seventy cases of pregnant women were selected. It included 35 cases of pregnant women with GDM (GDM group), 35 cases of normal pregnant women (normal control group). Another 35 women patients with type 2 diabetes in the same period as T2DM group. By using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), we detected the distribution of SUR1 alleles frequency in all individuals of three groups. We also examined BMI, fasting insulin (INS(0)) and 2 h insulin levels (INS(120)). RESULTS: There was significantly increased c alleles frequency of SUR1 in GDM and T2DM women compared with normal pregnancy (70.0%, 71.4% and 52.9%, P < 0.05). Also significantly increased A alleles frequency of SUR1 in GDM and T2DM women was found compared with normal pregnancy (41.4%, 44.3% and 24.3%, P < 0.05). In GDM group, women carrying cc genotype had a higher BMI [(29.2 +/- 5.6) kg/m(2)], INS(0) [(14.9 +/- 8.7) mU/L] and INS(120) [(40.2 +/- 12.1) mU/L] as compared with those in carriers of other genotypes (ct and tt) (all P < 0.05). Also they had a lower insulin sensitivity (HOMA-IR) (3.9 +/- 2.5) as compared with carriers of ct genotype. Women carrying AA genotype had a higher INS(0) as compared with carriers of other genotypes (AG and GG) [(15.4 +/- 3.2), (12.1 +/- 4.5) and (11.5 +/- 4.8) mU/L, both P < 0.05]. Logistic regression analysis showed there was relationship between SUR1 cc genotype and severity of GDM (P = 0.005, RR = 25.128). CONCLUSIONS: There are -3t-->c and A/G polymorphism of SUR1 gene in the Han nationality. Alleles c of SUR1 are significantly implicated in the susceptibility to GDM. It suggests that the defect in SUR1 gene (cc and AA) may contribute to insulin hypersecretion, which might be the cause of increased body weight and decreased insulin sensitivity and genotype cc of SUR1 is connected with severe type of GDM. It might be the predictable index of GDM conversion to type 2 diabetes.
