[The effect of dietary calcium on the expression of uncoupling protein 3 gene in skeletal muscle of rat fed with high fat diet].

In order to study the effects of dietary calcium on the expression of uncoupling protein 3(UCP3) in skeletal muscle of Wistar rat fed with high fat diet, 54 male Wistar rats were randomly divided into 6 groups: Group A was fed with basic diet(0.5% calcium), Group B was fed with high fat low calcium diet(0.08% calcium), Group C was fed with high fat normal calcium diet(0.5% calcium), Group D and E was fed with high fat high calcium diet(1% and 1.5% calcium respectively), Group F was fed with high fat high calcium diet (1.5% calcium) supplemented with vitamin D(600 IU%). Reverse transcription polymerase chain reaction (RT-PCR) was used to measure the level of UCP3 mRNA. The results showed that the body weight and fat content in group C were significantly higher than those in group A. However, the body weight gain in groups of D, E, and F was less than those in group C. The serum insulin in group C increased significantly compared with that in groups of A, D, E and F(P < 0.05), but there was no significant difference in blood glucose among all groups. Compared with group A, the level of serum leptin increased significantly in group C(P < 0.05) and increased slightly in groups D, E and F (P > 0.05). The level of serum FT3 in groups D, E and F increased significantly compared with that of group A, B and C(P < 0.05), but there was no significant difference in FT4 among all groups(P > 0.05). Compared with group A, the expression of UCP3 mRNA decreased in skeletal muscle in group C, while UCP3 mRNA increased in groups D, E and F compared with that of group C. In conclusion, dietary calcium may not only reduce the body weight gain induced by high fat diet, and improve hyperinsulinemia, but also may enhance the expression of UCP3 mRNA in skeletal muscle by increasing the serum level of leptin and FT3 which may play the role in the prevention of obesity.

[Comparison of blood hormone level between DIO-R and DIO rats].

UNLABELLED: To study the blood hormone level between diet-induced obesity-resistant (DIO-R) rats and diet-induced obesity(DIO) rats in blood hormone. METHODS: Fifty male Sprague-dawley (SD) rats were randomly divided into control group and high-fat group and they were fed with basic diet and high-fat diet respectively for 13 weeks. DIO-R and DIO rats were selected according to their body weight, then the intake of diet and body fat contents, the level of serum insulin, serum leptin and plasma NPY were determined by radioimmunoassay. The results showed that the weight, diet intake and body fat of DIO-R rats were significantly lower than those of DIO rats(P < 0.05). The level of serum insulin and plasma NPY of DIO-R rats were lower than those of DIO rats(P < 0.05). The level of serum leptin of rats in high-fat diet group was higher than that of the rat in basic diet group, but there was no significant difference between DIO-R and DIO rats(P > 0.05). CONCLUSION: High fat diets can induce SD rat to develop obesity and obesity-resistance. The balance of insulin-leptin-NPY feedback may play a partial of role in resisting diet-induced obesity of rats.

[Clinical analysis of gastroesophageal reflux induced cough].

OBJECTIVE: To investigate the clinical features, diagnosis and management of gastroesophageal reflux induced cough (GERC). METHODS: The continuous ambulatory esophageal pH measurement was performed for 24 hour in 41 cases with chronic cough who had normal chest roentgenographic presentation, negative histamine provocation test and no abnormality of nasal cavity. The symptom association probability (SAP) was adopted to analyze the correlation of cough with gastroesophageal reflux. The anti-reflux treatment was given for 12 weeks to those patients with Demeester score >or= 14.72 and/or SAP >or= 75%. RESULTS: Twenty six out of 41 cases were diagnosed as patients with gastroesophageal reflux. The cough was completely cured in 12 cases after antireflux treatment. The SAP of cough in this cured group (proximal probe 0.75 +/- 0.21, distal probe 0.91 +/- 0.12, respectively) was significantly higher than that of patients who failed in response to antireflux treatment (proximal probe 0.36 +/- 0.31, distal probe 0.47 +/- 0.30, respectively, P < 0.05). CONCLUSION: The gastroesophageal reflux may be an independent factor which could induce chronic cough. Not only the frequency and severity of gastroesophageal reflux but also the SAP were helpful in making the diagnosis of GERC. Satisfied effect was achieved after sufficient anti-reflux treatment in patients with GERC.

[Vegetation state and soil enzyme activities of copper tailing yard on Tongguan mountain].

From the open investigation and laboratory analysis, this paper studied the vegetation state and soil enzyme activities of copper tailing yard of Tongguan mountain. The results showed that there were 34 species of natural colonized plants on copper tailing yard, subordinated to 16 families and 33 genera, and regard herbs as principle, and many for 1-2 years old. The main families were compositae (10 species), gramineae (9 species) and legumineceae (2 species). Hippochaete ramosissimum, which belonged to equisetaceae, had and significant dominant. There were some microcoenses such as Hippochaete ramosissimum + Imperata cylindraca community, Cynodon dactylon + Imperata cylindraca community and Phragmites australis community. But, the vegetation on copper tailing yard was distributed in spot piece and scattered mainly with single species. The activities of three soil enzymes had a stronger sensitivity to the vegetation state, and their relativity to the vegetation state was in order of urease > sucrase > catalase. It's suggested that unrease activity could be used as an indicator index for the reclamation of wasteland.

[A study on the expression of TGF-beta 1/T beta R II in laryngeal carcinoma].

OBJECTIVE: To explore the expression of TGF-beta 1/T beta R II in laryngeal carcinoma. METHOD: Immunohistochemical study using SP kit in 53 cases. RESULT: The expression of TGF-beta 1 was decreased in carcinoma tissues when compared with peri-cancer controls (P < 0.05). There wasn't difference in T beta R II expression when compared with peri cancer controls (P > 0.05). But it was correlated with the stage, differentiation and lymph node metastasis of laryngeal carcinoma. CONCLUSION: TGF-beta 1/T beta R II may involve in the histogenesis and development of laryngeal carcinoma. The decreased expression of TGF-beta 1/T beta R II may serve as an important molecular marker of laryngeal carcinoma.

[Tumor necrosis factor-alpha, caspase-3 expression and hepatocyte apoptosis in fulminanting hepatic failure].

OBJECTIVE: To study the role of tumor necrosis factor-alpha (TNFalpha) and caspase-3 expression on hepatocyte apoptosis in experimental model of fulminant hepatic failure (FHF). METHODS: Mouse experimental model of FHF was induced by lipopolysaccharide (LPS) and D-galactosamine (D-GalN). Serum TNFalpha level and TNFalpha mRNA expression in liver were tested by ELISA and reverse transcriptase PCR (RT-PCR) method, respectively. The expression of caspase-3 in liver tissue was determined by in situ hybridization. Hepatocyte apoptosis was examined by DNA agarose gel electrophoresis and TUNEL method. TNFalpha, caspase-3 and hepatocyte apoptosis were observed in different stages after drug administration. In addition, we observed the changes of above variables after pretreatment with anti-TNFalpha IgG1. RESULTS: TNFalpha mRNA expression in liver increased significantly (0.91 +/- 0.75, that of normal control was 0.32 +/- 0.10) 2 to 4 hours after administration of LPS and D-GalN. The level of serum TNFalpha increased [(320.50 +/- 86.57) ng/L; that of normal control was (16.66 +/- 7.01) ng/L] and caspase-3 expressed to a slight extent, too. Typical manifestation of hepatocyte apoptosis appeared 8h after drug administration. 8 hours after drug administration the level of serum ALT and total bilirubin (TBil) remarkably increased [(560.66 +/- 60.20) U/L and (163.66 +/- 34.51) micro mol/L, respectively, that of normal control was (23.56 +/- 8.03) U/L and (14.90 +/- 4.80) micro mol/L, respectively] and the expression of caspase-3 reached the peak. 12 hours after drug administration, hepatocyte apoptosis and necrosis coexisted, and the level of serum ALT and TBil reached a peak [(668.30 +/- 78.69) U/L and (203.17 +/- 19.92) micro mol/L, respectively] whereas the expression of caspase-3 already decreased. Hepatocyte apoptosis and liver injury and the expression of caspase-3 could be blocked by antagonizing TNFalpha. CONCLUSIONS: TNFalpha played an important role in hepatocyte apoptosis and liver injury in fulminant hepatic failure. The hepatocyte apoptosis induced by TNFalpha is correlated with the activation of caspase-3. Hepatocyte apoptosis occurs earlier than necrosis.

[Application of support vector machines to classification of blood cells].

The support vector machine (SVM) is a new learning technique based on the statistical learning theory. It was originally developed for two-class classification. In this paper, the SVM approach is extended to multi-class classification problems, a hierarchical SVM is applied to classify blood cells in different maturation stages from bone marrow. Based on stepwise decomposition, a hierarchical clustering method is presented to construct the architecture of the hierarchical (tree-like) SVM, then the optimal control parameters of SVM are determined by some criterion for each discriminant step. To verify the performances of classifiers, the SVM method is compared with three classical classifiers using 3-fold cross validation. The preliminary results indicate that the proposed method avoids the curse of dimensionality and has greater generalization. Thus, the method can improve the classification correctness for blood cells from bone marrow.

Influence of patients' age on functional recovery after transplantation of olfactory ensheathing cells into injured spinal cord injury.

OBJECTIVE: To evaluate the restoration of function after spinal cord injury (SCI) in patients of different ages who have underwent intraspinal transplantation of olfactory ensheathing cells (OECs). METHODS: One hundred and seventy-one SCI patients were included in this study. Of them, 139 were male and 32 were female, with age ranging from 2 to 64 years (mean, 34.9 years). In all SCI patients the lesions were injected at the time of operation with OECs. According to their ages, the patients were divided into 5 groups: </= 20 years group (n = 9), 21 - 30 years group (n = 54), 31 - 40 years group (n = 60), 41 - 50 years group (n = 34) and > 51 years group (n = 14). The spinal cord function was assessed based on the American Spinal Injury Association (ASIA) Classification System before and 2 - 8 weeks after OECs transplantation. One-way ANOVA and q test were used for statistical analysis, and the data were expressed as mean +/- SD. RESULTS: After surgery, the motor scores increased by 5.2 +/- 4.8, 8.6 +/- 8.0, 8.3 +/- 8.8, 5.7 +/- 7.3 and 8.2 +/- 7.6 in 5 age groups respectively (F = 1.009, P = 0.404); light touch scores increased by 13.9 +/- 8.1, 15.5 +/- 14.3, 12.0 +/- 14.4, 14.1 +/- 18.5 and 24.8 +/- 25.3 respectively (F = 1.837, P = 0.124); and pin prick scores increased by 11.1 +/- 7.9, 17.2 +/- 14.3, 13.2 +/- 11.8, 13.6 +/- 13.9 and 25.4 +/- 24.3 respectively (F = 2.651, P = 0.035). Restoration of pin prick in > 51 years group was better than other age groups except 21 - 30 years group. CONCLUSION: OECs transplantation can improve the neurological function of spinal cord of SCI patients regardless of their ages. Further research into the long-term outcomes of the treatment will be required.

[Relation of body mass index and waist circumference with clustering of other risk factors for cardiovascular disease].

OBJECTIVE: To assess the relationship of body mass index (BMI) and waist circumference (WC) with clustering of other risk factors for cardiovascular disease (CVD). METHODS: A total of 30 561 participants aged 35 - 59 from different parts of China were surveyed for risk factors of CVD in two independent cross-sectional studies carried out in 1992 - 1994 and 1998. Data were pooled to analyze clustering rate of risk factors for CVD and relative risk of their clustering at varied levels of BMI and WC. Clustering of other risk factors for CVD was defined as any participant who had any two or more risk factors, such as high blood pressure, high serum total cholesterol, low HDL-C, high fasting plasma glucose. Clustering rate of other risk factors for CVD at different levels of BMI and WC was estimated. RESULTS: Clustering rate of other risk factors for CVD significantly increased with rising of BMI and WC. In most of varied BMI groups, clustering rate of other risk factor increased with rise of WC in both men and women (P value for trend < 0.05), and in most of varied WC groups, clustering rate of other risk factor significantly increased with rise of BMI (P value for trend < 0.05). Clustering rate of other risk factors for CVD adjusted for age was 11.1% and 10.4% with BMI < 24.0 kg/m(2) and WC < 85/80 cm, 24.0% and 17.0% with BMI of 24 - 27.9 kg/m(2) and WC < 85/80 cm, 34.3% and 24.0% with BMI of 24 - 27.9 kg/m(2) and WC of 85 - 95.9/80 - 89.9 cm, 40.8% and 29.6% with BMI of 24 - 27.9 kg/m(2) and WC >/= 95/90 cm, 44.2% and 29.9% with BMI >/= 28 kg/m(2) and WC of 85 - 95.9/80 - 89.9 cm, and 54.7% and 35.4% with BMI >/= 28 kg/m(2) and WC >/= 95/90 cm, for men and women, respectively. CONCLUSIONS: BMI and WC were independently and positively associated with clustering rate of other risk factors for CVD. It is very important for health to keep both BMI and WC in normal level.

[A study of the link between upper and lower respiratory inflammation].

OBJECTIVE: To evaluate the correlation between upper and lower respiratory inflammation. METHODS: Thirty rats were randomly divided into allergic rhinitis group (AR), asthma group (AS), and control group (Con). The pulmonary function was measured by pulmonary function instrument of animal model, the infiltration of eosinophils and mast cells was detected by hematotoxylin-eosin staining (HE staining) and toluidine blue staining respectively, the expression of VCAM-I and IL-13 was examined by immunohistochemistry. ELISA was used to measure the concentration of IL-5 in the peripheral serum. RESULTS: The numbers of eosinophils and mast cells in nasal mucosa and lung tissue of AR were both significantly higher than those of Con, and similar results were observed between AS and Con. The forced expiratory volume in 0.3 second (FEV0.3)/forced vital capacity (FVC) of AR and AS was both significantly lower than that of Con. The numbers of VCAM-I and IL-13 positive vessels in lung tissue of AR were both significantly higher than those of Con, and similar results were observed between AS and Con. The concentration of IL-5 in the serum of AR and AS was both significantly higher than that of Con. The concentration of IL-5 in serum was positively correlated with the number of eosinophils in peripheral blood of AR and AS. CONCLUSION: The inflammation is similar between AR and AS, and repeatedly challenge upper respiratory tract can impact on lower respiratory response.

[Treatment of helical keloid with retroauricular skin expansion].

OBJECTIVE: To introduce a new technique for the treatment of helical keloid. METHODS: The procedure consisted of two steps. In the first step, a kidney-shaped expander of 50 ml was placed subcutaneously in the retroauricular area. Routine inflation with normal saline followed. In the second step, about two months afterwards, the expander was taken out and the helical keloid was excised. The expanded flap was advanced to cover the wound. RESULTS: The operation has been performed on 12 patients of 16 sites of helical keloid since 2000. Postoperative follow-up from 3 months to 2 years revealed satisfactory results. The reconstructed ear maintained a good contour. CONCLUSION: The technique creates a retroauricular flap rich in blood supply, which can be used to close the defect left by helical keloid excision. It provides a good treatment for the helical keloid replaced with normal skin from the mastoid region.

Hypocretin/Orexin excites hypocretin neurons via a local glutamate neuron-A potential mechanism for orchestrating the hypothalamic arousal system.

Neurons that release hypocretin/orexin modulate sleep, arousal, and energy homeostasis; the absence of hypocretin results in narcolepsy. Here we present data on the physiological characteristics of these cells, identified with GFP in transgenic mouse brain slices. Hypocretin-1 and -2 depolarized hypocretin neurons by 15mV and evoked an increase in spike frequency (+366% from a 1-3 Hz baseline). The mechanism for this appears to be hypocretin-mediated excitation of local glutamatergic neurons that regulate hypocretin neuron activity, in part by presynaptic facilitation of glutamate release. This represents a possible mechanism for orchestrating the output of the diffuse hypothalamic arousal system. No direct effect of hypocretin on membrane properties of hypocretin cells was detected. Norepinephrine and serotonin, transmitters of other arousal systems, decreased spike frequency and evoked outward currents, whereas acetylcholine and histamine had little effect.

Sequence and analysis of rice chromosome 4.

Rice is the principal food for over half of the population of the world. With its genome size of 430 megabase pairs (Mb), the cultivated rice species Oryza sativa is a model plant for genome research. Here we report the sequence analysis of chromosome 4 of O. sativa, one of the first two rice chromosomes to be sequenced completely. The finished sequence spans 34.6 Mb and represents 97.3% of the chromosome. In addition, we report the longest known sequence for a plant centromere, a completely sequenced contig of 1.16 Mb corresponding to the centromeric region of chromosome 4. We predict 4,658 protein coding genes and 70 transfer RNA genes. A total of 1,681 predicted genes match available unique rice expressed sequence tags. Transposable elements have a pronounced bias towards the euchromatic regions, indicating a close correlation of their distributions to genes along the chromosome. Comparative genome analysis between cultivated rice subspecies shows that there is an overall syntenic relationship between the chromosomes and divergence at the level of single-nucleotide polymorphisms and insertions and deletions. By contrast, there is little conservation in gene order between rice and Arabidopsis.

[Association between low-dose oral contraceptive use and stroke in Chinese women].

OBJECTIVE: To investigate the association between low-dose combined oral contraceptive (COC) widely distributed in China and the risk of hemorrhagic stroke so as to decrease the adverse reaction of COC. METHODS: A prospective cohort study was made among 44 408 women who took hormonal contraceptives (HCs) and 75 230 women who used in trauterine device (IUD) in 25 townships and toens in Rudong County and Taicang City, Jiangsu Province, to compare the incidence of stroke among them from 1 July 1997 to 30 June 2000. RESULTS: (1) The incidence of hemorrhagic stroke (age-and-county standardized rates) was 34.74 per 100 000 py in the HC cohort, 2.72 times higher than that in the IUD cohort (P < 0.01). The incidence of hemorrhagic stroke among women less than 45 years of age was remarkably higher in HC cohort than in the IUD cohort. The relative risk of incidence of hemorrhagic stroke was 2.17 (1.16, 4.06) among the HC users who had stopped HC use for more than 10 years, still significantly higher than among the IUD users. (2) The relative risk of incidence of hemorrhagic stroke among the women who used low-dose COCs during the survey was 3.60 (1.73, 7.53) times higher than that among the IUD users. The relative risk of incidence of hemorrhagic stroke was 3.09 (1.26, 7.57) among the women who had stopped low-dose COC use for less than five years, however, the risk of hemorrhagic stroke remarkably decreased after they stopped taking COC for more than 5 years. (3) No obvious increase in incidence of infarctional stroke was found among the women who took low-dose COC during the survey. (4) Hypertension might be the most important risk factor of hemorrhagic stroke among the low-dose COC users. CONCLUSION: There is an obvious increase in risk of hemorrhagic stroke among the Chines women who take low-dose of China-made COCs for a long time. The efffect of China-made COCs on pathogenesis of hemorrhagic stroke lasts sometime after the discontinuation of COC use.

Expression of MUM1/IRF4 correlates with clinical outcome in patients with B-cell chronic lymphocytic leukemia.

In this study, we evaluated the prognostic significance of multiple myeloma-1/interferon regulatory factor-4 (MUM1/IRF4) expression in B-cell chronic lymphocytic leukemia (B-CLL). Our results demonstrated that the absence of MUM1/IRF4 expression showed the highest relative risk among the factors analyzed in determining the probability for death in patients with B-CLL using univariate and multivariate Cox regression analysis. Patients without MUM1/IRF4 expression had significantly worse overall survival than did those with MUM1/IRF4 expression (52% cumulative survival, 63 months vs not reached, Kaplan-Meier survival analysis; P <.03, log-rank test). Patients with MUM1/IRF4 expression were more likely to have disease at low Rai stage and interstitial/nodular marrow involvement. Furthermore, only 1 of 11 patients with MUM1/IRF4 expression and interstitial/nodular marrow involvement died during a 100-month follow-up. Our results suggest that B-CLL with expression of MUM1/IRF4, indicative of postgerminal center origin, has a more favorable clinical course and that MUM1/IRF4 is an important prognostic marker in B-CLL.

Anti-nociceptive effect of neuropeptide Y in the nucleus accumbens of rats: an involvement of opioid receptors in the effect.

The present study investigated the effect of neuropeptide Y on nociception in the nucleus accumbens of rats. Intra-nucleus accumbens administration of neuropeptide Y induced dose-dependent increases in the hindpaw withdrawal latency (HWL) to thermal and mechanical stimulation in rats. There were no significant changes in the HWL to both stimulation during 60 min after the administration of NPY to outside of the nucleus accumbens. The anti-nociceptive effect of NPY was blocked by subsequent intra-nucleus accumbens injection of the Y1 receptor antagonist neuropeptide Y 28-36, indicating that Y1 receptor is involved in the neuropeptide Y-induced anti-nociception in the nucleus accumbens. Furthermore, the anti-nociceptive effect of neuropeptide Y was attenuated by intra-nucleus accumbens administration of the opioid antagonist naloxone, suggesting an involvement of the endogenous opioid system in the neuropeptide Y-induced anti-nociception in the nucleus accumbens of rats. Moreover, the neuropeptide Y-induced anti-nociception was attenuated by following intra-nucleus accumbens injection of the selective opioid antagonists nor-binaltorphimine and beta-funaltrexamine, but not by naltrindole, illustrating that mu- and kappa-opioid receptors, not the delta-opioid receptor, were involved in the neuropeptide Y-induced anti-nociception in the nucleus accumbens of rats.

Hsp110 over-expression increases the immunogenicity of the murine CT26 colon tumor.

Several studies have suggested a positive correlation between heat shock protein (hsp) expression and tumor immunogenicity. Independently, many studies have shown that hsp purified from tumors can be used as a tumor-specific vaccine. In this study, we have explored the connection between hsp expression and anti-tumor immunity by transducing murine CT26 colon carcinoma cells with the cDNA of a major hsp, i.e. hsp110. We have shown that over-expression of hsp110 has no effect on CT26 tumor cell growth in vitro, and does not inhibit their anchorage-independent growth capacity. However, in situ, hsp110 over-expressing CT26 tumor (CT26-hsp110) grew at a significantly reduced rate as compared to the wild-type CT26 tumor in immunocompetent mice. Moreover, immunization of mice with inactivated CT26-hsp110 cells significantly inhibited the growth of wild-type CT26 tumor. This immunity was associated with an increased frequency of tumor-specific T cells after vaccination. An in vivo antibody depletion assay demonstrated that inactivated CT26-hsp110 cells elicited anti-tumor responses involving CD8(+) T cells and natural killer (NK) cells, but not CD4(+) T cells. Lastly, the effect of the addition of granulocyte-macrophage colony stimulating factor (GM-CSF) to these vaccine formulations was determined. Mice immunized with irradiated CT26-hsp110 cells combined with GM-CSF-producing bystander cells revealed a complete inhibition of CT26 tumor growth, indicating a synergy between inactivated CT26-hsp110 vaccine activity and GM-CSF. These observations demonstrate that manipulation of hsp110 expression in tumors, specifically when combined with GM-CSF, represents a potentially powerful approach to cancer vaccine formulation.
