Pharmacokinetic study with N-Ile1-Thr2-63-desulfato-r-hirudin in rabbits by means of bioassay.

AIM: To study the pharmacokinetic (PK) properties in rabbits treated with N-Ile(1)-Thr(2)-63-desulfato-r-hirudin (rH) newly developed in China by means of bioassay in order to provide preclinical experiment basis for its development as a novel anticoagulant agent. METHODS: rH plasma concentration was determined using bioassay based on ex vivo antithrombin activity of rH. Normal rabbits received iv rH 4.0, 2.0 and 1.0 mg/kg or sc rH 2.0 mg/kg, respectively. The rabbits with acute severe renal failure were given iv rH 2.0 mg/kg. RESULTS: The bioassay described in this paper met requirements for study of PK in rabbits. The major PK parameters after iv dosing were as follows: t(1/2beta) 58.4-59 min. V(d) 0.09-0.12 L/kg, CL 0.0035-0.0040 L/(kg.min); AUC were proportional to the doses, t(1/2) and CL did not change significantly with the doses. The sc bioavailability reached 94%. The rabbits suffering from acute severe renal failure presented 11-fold longer t(1/2beta) and 13-fold greater AUC than normal healthy rabbits. CONCLUSION: rH exhibited rapid elimination, distribution was only limited to extracellular space and good absorption from sc site. The excretion of rH by kidneys played a very important role in the elimination of rH. The PK of rH could be described by the two- and one-compartment model after iv and sc dosing, respectively, and followed linear kinetics.

[Expression of ETV5 and MMP-7 in early stage cervical squamous cell carcinoma and its role in lymphatic metastasis].

BACKGROUND & OBJECTIVE: Transcription factor Ets variant gene 5 (ETV5) participates in tumor metastasis, and matrix metalloproteinase-7 (MMP-7) also plays an important role in tumor invasion and metastasis. This study was to investigate the expression of ETV5 and MMP-7 in cervical squamous cell carcinoma, and explore the roles of ETV5 and MMP-7 in invasion and metastasis of early stage cervical squamous cell carcinoma. METHODS: Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of ETV5 and MMP-7 mRNA in 86 samples of cervical squamous cell carcinoma and 21 samples of normal cervix, and immunohistochemistry was used to detect the expression of MMP-7 protein in these samples. RESULTS: The mRNA levels of ETV5 and MMP-7 were significantly higher in cervical cancer tissues than in normal cervix tissues (0.507+/-0.073 vs. 0.201+/-0.057, P<0.01; 0.488+/-0.078 vs. 0.305+/-0.064, P<0.01), and were significantly higher in nodal metastasis group than in no nodal metastasis group (0.538+/-0.063 vs. 0.496+/-0.073, P<0.05; 0.542+/-0.066 vs. 0.468+/-0.072, P<0.01). The protein level of MMP-7 was significantly higher in cervical cancer tissues than in normal cervix tissues (3.826+/-1.427 vs. 1.714+/-0.383, P<0.01), and was significantly higher in nodal metastasis group than in no nodal metastasis group (4.935+/-1.614 vs. 2.623+/-1.086, P<0.01). The positive rate of MMP-7 protein was significantly higher in lymph node metastasis group than in no lymph node metastasis group (65.21% vs. 36.51%, P<0.01). ETV5 expression was positively correlated to MMP-7 expression (r=0.221, P<0.05). CONCLUSION: ETV5 and MMP-7 are highly expressed in metastatic cervical squamous cell carcinoma, and may be served as markers in estimating the invasive and metastatic potential of cervical squamous cell carcinoma.

[Expression of NMDA receptor-1 induced by NMDA in the brain of neonatal SD rats].

OBJECTIVE: To determine the regulation of the expression of NMDA receptor-1 induced by NMDA in the brain of neonatal SD rats. METHODS: Neonatal SD rats (n=90) were randomly divided into normal control (n=6) and NMDA injected group (subdivided into 10 nmol-0 min, 15 min, 30 min, 1 h, 2 h, 4 h groups, and 10, 20, 50 nmol groups, each n=6). NMDA fluorescent inmmunohistological staining and TTC (2,3,5-triphenyltetrazoliun chloride) staining techniques were used. RESULTS: At 30 min after the injection of 10 nmol NMDA, a few NR1 positive cells could be observed along the injection tract. At 1 h after the injection, NR1 positive cells in large quantity could be observed in the hippocampal CA1 region and paraventricular thalamus of the ipsilateral hemisphere. The number and location of positive cells at 2 h and 4 h after the injection were not much different from that at 1 h after the injection. At 2 h after injection, stronger NR1 expression was observed in the 50 nmol injection group. In addition, slight crinkle of the cell wall with mild condensation of the nuclei was also observed in the 50 nmol injection group. At 2 h after the injection, no abnormality was observed in 10, 20, or 50 nmol group after TTC staining. CONCLUSION: The NR1 induced by NMDA is expressed in a time-dependent and dose-dependent pattern after a short period of "delay", providing a possible "therapeutic window" for using NMDA receptor antagonist to treat diseases relating to the NMDA receptor activation.

Influence factors for functional improvement after olfactory ensheathing cell transplantation for chronic spinal cord injury.

OBJECTIVE: To explore the influence factors for the functional improvement after the fetal olfactory ensheathing cell (OEC) transplantation for chronic spinal cord injury(SCI). METHODS: The olfactory bulbs were harvested and trypsinized down to single fetal OEC. They were cultured for 12-17 days prepared for use. From November 2001 to December 2003, a total of 300 patients volunteered for the fetal OEC transplantation, among whom 222 suffered from complete chronic SCI and 78 suffered from incomplete chronic SCI. The procedures were performed on the patients with a disease course ranging from 6 months to 31 years (average 3.1 years) after their injuries. The fetal OEC was transplanted by the form of injections into the spinal cord at the upper and lower ends of the injury site. All the patients were assessed by the ASIA standard before the transplantation and 2-8 weeks after the transplantation. The influence factors including age, sex, duration after the injury, and injury degrees and levels were compared with those in the functional improvement after fetal OEC transplantation. RESULTS: The partially-improved neurological functions assessed by the ASIA standard were indicated by the motor scores increasing from 39.1 +/- 20.6 to 45.9 +/- 20.3 (P < 0.001), the light touch scores from 51.7 +/- 24.9 to 63.4 +/- 23.0 (P < 0.001), and the pin prick scores from 53.0 +/- 24.2 to 65.3 +/- 22.7 (P < 0.001). There was no significant difference in the functional improvement of the motor, light touch, and pin brick when compared with the age, sex, duration after the injury, and the injury degrees and levels. The motor scores and light touch scores at the cervical level were higher than the scores at the thoracic level. CONCLUSION: The fetal OEC transplantation can partially improve the neurological functions quickly in treatment of the chronic spinal cord injury. All the influence factors except the motor scores and light touch scores, which were higher at the cervical level than at thoracic level, have no impact on the functional improvement after the fetal OEC transplantation.

[Synthesis and immunosuppressive activity of new artemisinin derivatives containing polyethylene glycol group].

AIM: To search for new artemisinin derivatives with higher immunosuppressive activity. METHODS: Two kinds of new artemisinin derivatives containing polyethylene glycol group were synthesized from dihydroartemisinin via condensation and esterification. These compounds were assayed for their inhibitory activity on ConA-induced T cell proliferation and LPS-induced B cell proliferation. RESULTS: Twenty three new compounds (2a - 2f, 3a - 3d, 4a - 4f, 6a, 6b and 7a - 7g) were synthesized and identified by 1H NMR and elemental analysis. CONCLUSION: These compounds had immunosuppressive activity in vitro. Among them, the symmetrical substituted compound 2 and 6 had higher activity than mono-substituted compound 3, 4 and 7. Especially, compounds 2a - 2f remarkably exhibited higher inhibition in comparison with artemisinin and artesunate.

A multicentral randomized control study on clinical acupuncture treatment of Bell's palsy.

OBJECTIVE: To confirm the clinical therapeutic effects of acupuncture and moxibustion on Bell's palsy. METHODS: 480 cases from 4 hospitals were enrolled for this study, among whom 439 cases completed the whole course of the study. The patients were randomly divided into the following 3 groups, a control group(treated with prednisone, vitamin B1, vitamin B12 and dibazol), an acu-moxibustion group (treated with filiform needle plus moxibustion), and a basic treatment plus acu-moxibustion group (treated with oral medicine like those in the control group plus acupuncture, and with moxibustion like in the acu-moxibustion group). The whole treatment course lasted 4 weeks. The therapeutic effects were evaluated according to the symptoms and signs, House-Brackmann grading scale and facial disability indexes(FDI). RESULTS: All the 4 centers (hospitals) completed this study well, with no statistically significant difference found among the 4 centers in therapeutic effects. The patients with different conditions were well distributed in the 3 groups, thus the basic general data were comparable (P > 0.05). The therapeutic effects of the two treatment groups were better than the control group (respectively P < 0.05 and P < 0.01), and it was the best in the acu-moxibustion group (P < 0.01). CONCLUSION: Acupuncture and moxibustion may exert definite therapeutic effects on Bell's palsy, better than that of the basic treatment group or the basic treatment plus acu-moxibustion group.

[Physiological metabolism and protective enzyme activity of Equisetum ramosissimum under Cu stress].

The study with pot culture experiment showed that Equisetum ramosissirmunm did not appear obvious poisoning symptoms when treated with low concentration Cu (500 mg x kg(-1)), while serious injuries were found when treated with high concentration Cu (1000 to approximately 3000 mg x kg(-1)), which reflected in the severe damage of cell membrane and cytoarchitecture as well as the structure and function of main organelles, and the significant decrease of the contents of leaf chlorophyll a and b and stem soluble monosaccharose. The cell membrane osmolarity and the average MDA content of the plant exposed to heavy copper pollution was 1 to approximately 2 and 1 to approximately 3 times greater than the control, respectively. It could be concluded that high concentration Cu disturbed the physiological metabolism, and critically threatened the normal growth of E. ramosissimum. The activities of protective enzyme, especially of SOD and POD, were enhanced with increasing Cu concentration, and had a positive correlation with Cu concentration (rPOD = 0.978, rSOD = 0.926, P < 0.05).

[Design and application of biological rhythm automatic monitor and data analysis system].

OBJECTIVE: To design and develop a biological rhythm monitor and data analysis system for studying biological rhythm in animals under different conditions. METHODS: The system was a distributed digital control system consisting of a computer and zeitgeber generators. Acquisition of data on animal activity and generation of zeitgeber were modularized to meet the multiple requirements of experiment. System functions and animal adaptability to this system were detected by monitoring the rhythm of mouse activities under different light-dark cycles. RESULT: The animal experiment showed that the functions of this system, including data acquisition, data communication, light regulation and data analysis, were qualified for application. There was no apparent malfunction. CONCLUSION: The system is safe, reliable and easy to operate. The system could be a very useful system to study biological rhythm in animals.

[Mutation analysis of FOXL2 in Chinese patients with blepharophimosis-ptosis-epicanthus inversus syndrome].

OBJECTIVE: To identify the genetic mutation in two Chinese families and 6 sporadic patients with belpharophimosis-ptosis-epicanthus inversus syndrome (BPES). METHODS: Polymorphisms of 5 satellite markers on 3q were analyzed and linkage analysis was performed using linkage software (MLINK) in all cases of two families. FOXL2 gene fragments were amplified by PCR and mutation was determined by sequencing DNA fragments in all patients. RESULTS: The BPES locus in the pedigrees was mapped to 3q23, a 9.88 cM interval between markers D3S3696 and D3S1744. The maximum lod scores were 2.11 (theta = 0.00) at D3S1549 and D3S3586 and 1.51 (theta = 0.00) at D3S1764. By direct sequencing FOXL2 gene, two sporadic cases had a 30-bp in frame duplication 909 - 938 dup 30 and one sporadic case showed a nucleotide insertion 1041 - 1042 ins C. However, it was unable to find any causal mutation of FOXL2 in two families with BPES. CONCLUSIONS: The gene responsible for BPES in two Chinese families was linked to D3S1549, D3S3586 and D3S1764. This is the first reported mutations of FOXL2 (909 - 938 dup 30 and 1041 - 1042 ins C) in Chinese sporadic cases. One of the mutations, in-frame 30-bp duplication (909 - 938 dup 30), is one of the most common mutation hotspots in the coding region of FOXL2. In BPES families without FOXL2 mutation, it cannot be excluded that the disorder is caused by a position effect in the surrounding region of FOXL2 gene or by other genes located at 3q23.

[A research on TGFBI gene mutations in Chinese families with corneal dystrophies].

OBJECTIVE: To identify what kind of TGFBI gene mutation happening to Chinese patients with corneal dystrophies. METHODS: Three Chinese families with stromal corneal dystrophies and one Chinese family with Thiel-Behnke corneal dystrophies were studied, of whom three were Han race and another was Mongolia race in China. All members of families were examined clinically and their genomic DNAs were extracted from blood leukocytes. Thirteen exons in TGFBI gene were amplified by polymerase chain reaction (PCR) and directly sequenced for molecular analysis. RESULTS: Mutations in TGFBI gene were detected from all the patients with corneal dystrophy, but not found in normal subjects of families. The mutation R555W was found and identified from the family with granular corneal dystrophy; R555Q from the family with Thiel-Behnke corneal dystrophy; and R124H from the other two families with Avellino corneal dystrophy. CONCLUSION: The above study results show that the amino acids R124 and R555, if their genetic codes result from the mutations, play an important role in the pathogenesis of autosomal dominant corneal dystrophy of Chinese patients, and the molecular genetic analysis can improve the accuracy of diagnosing corneal dystrophy. In China, the mutation R555Q found in the family with Thiel-Behnke corneal dystrophy is reported for the first time.

[Applying patial least-squares discriminant analysis on autofluorescence spectra to identify gastric cancer].

Measurement of fluorescence intensity was performed at excitation wavelength of 308 nm and emission wavelength in the range of 328-596 nm. The partial least-squares (PLS) method was used to analyze autofluorescence spectra of gastric cancer. The 42 normal samples and 42 cancer samples were taken from 42 gastric cancer patients. The normalized and centralized spectra of two kinds of samples showed similar but divergent patterns. PLS classification algorithm could differentiate cancer tissues from normal tissues with a sensitivity of 83.3%, a specificity of 95.2%, and a positive predictive value of 94.6%. We concluded therefore that the PLS method was a fast, effective choice for identification of gastric cancer.

[Effects of tacrolimus and cyclosporine on albumin secretion in cultured human hepatocyte].

OBJECTIVE: To investigate the effects of inflammation cytokines, (FK506) and cyclosporine (CSA) on albumin secretion, and the effects of FK506 and CSA on the IL-6 induced suppression of albumin synthesis in cultured human hepatocytes. METHODS: Human hepatoma cell lines (HepG2 cells) were separately cultured with IL-6, IL-2 and IL-10 (0 approximately 10 microg/L) and FK506, CSA (0 approximately 100 microg/L) for 48 h. In another experiment, HepG2 cells were stimulated with different doses of FK506 and CSA (0 approximately 10 microg/L) in the presence of IL-6 (5 microg/L) for 48 h. Albumin levels in the supernatant of all groups were measured by radioimmunoassay (RIA). The concentration of LDH secreted by cells stimulated with FK506 and CSA were detected with spectrophotometry. RESULTS: For cultured HepG2 cells, IL-6 significantly decreased albumin levels in a dose-dependent manner (P <0.01), and the maximal inhibition occurred at 5 microg/L. CSA mildly decreased albumin levels and a significant reduction in albumin production was first visible at 10 microg/ L (P <0.05). In contrast, IL-2, IL-10 and FK506 did not significantly influence albumin pro- duction (P > 0.05). FK506 obviously decreased LDH levels in the supernatant (P < 0.05) and attenuated IL-6 induced suppression of albumin synthesis (P < 0.01). But CSA slightly increased LDH concentration and could not block the IL-6 induced decrease of albumin synthesis (P > 0.05). CONCLUSION: IL-6 but not IL-2 and IL-10 suppressed the production of hepatic albumin in vitro. FK506 protected against the suppression of hepatic albumin synthesis caused by IL-6, suggesting its potential role in improving hypoalbuminaemia in immune glomerulonephritis.

[Effects of different cryoprotectants on post-thaw viability of porcine preadipocyte].

To explore the optimal cryoprotectant for porcine preadipocyte cryppreservation, the primary pig preadipocytes were cryopreseved with DMSO, ethlyleneglycol(EG), PVP and DMSO+PVP cryoprotective agents (CPA) and without CPA, respectively. After 30d, the cells were recovered, viability of pig preadipocyte cryopreserved with different CPAs was evaluated by the Typlan exclusion, growth characteristic was analyzed with MTT, the differentiation potentials of pig preadipocytes were observed by morphology change from preadipocyte to mature adipocyte and Oil Red O staining. The results were as follows: four cryoprotectants all could protect the pig preadipocyte from frozen at a certain degree, and the efficiency was as follows: PVP > MSO+PVP > DMSO > EG > without cryoprotectant. The growth state and the lipogenic capability of the pig preadipocytes cryopreserved with PVP were similar to those unfreezing cells. Thus, we draw a conclusion: PVP is the optimal cryoprotectant for pig preadipocyte.

[Comparison of the difference of the magnetic force by cast post-keepers combined with three different metals].

PURPOSE: Three different metals were used to make cast post-keepers of magnetic attachment. The magnetic force was tested and analyzed. METHODS: The cast-post keepers were combined by gold alloy, Ni-Cr alloy and Ti metal respectively. 12 samples were divided into three groups, 4 in each. Root surface structures were manufactured by these samples. The values of the magnetic force of the magnetic attachment were tested and compared using ANOVA. RESULTS: The average value of the attractive force of the keeper combined by gold alloy was 4.84N, while that of the keeper combined by Ni-Cr alloy was 4.61N and the keeper combined by Ti was 4.12N. Significant difference was found between each two samples, P<0.01. CONCLUSION: The magnetic force of the keepers combined by three different metals decreased to certain degrees, the keeper combined by gold alloy decreased the least.

[Photochemical degradation of chlorothalonil in aqueous solution].

The study on the effects of light source, solution pH and temperature, and surfactant on the photochemical degradation of chlorothalonil showed that the half-life of chlorothalonil photodegradation under high pressure mercury lamp (HPML), UV lamp and sunlight was 22.4, 82.5 and 123.8 min, respectively. Under HPML and sunlight, chlorothalonil had a higher photolysis rate in alkaline solution than in neutral and acid solution. The photolysis rate increased with increasing solution temperature in the range of 10 degrees C - 40 degrees C, which was doubled when the temperature increased every 10 degrees C. Sodium laurylsulfonate (SDS), sodium dodecylbenzene sulfonate (SDBS), Tween 60 and Span 20 showed significant photosensitizing effects, while cetyltrimethylammonium bromide (CTAB) had significant photoquench effect on the photolysis of chlorothalonil.

[Effects of alendronate on bone mineral density, cytokines and indices of bone metabolism in postmenopausal osteoporotic patients].

OBJECTIVE: To observe the effects of alendronate on bone mineral density (BMD), cytokines and indices of bone metabolism in postmenopausal osteoporotic (PMO) patients. METHODS: 185 PMO patients, aged from 55 to 60 years, were randomized into three groups. All of them received a kind of calcium preparation, 1.0 g/d. Group A, 69 patients, received alendronate, 10 mg/d; group B, 66 patients, received tibolone 1.25 mg/d. The remaining patients, group C, received calcium preparation only. 20 women of 55 - 60 (57.4 +/- 3.5) years old were taken as normal controls. Dual-energy X-ray absorptiometry and measurement of a series of biochemical indices were carried out before and after medication at 24th and 48th week. RESULTS: After medication, BMD increased in alendronate and tibolone group. The increment in spine BMD was 2.53% and 3.65% (P < 0.05) and that in nondominant proximal femur BMD was 7.17% and 3.01% (P < 0.001). In the tibolone group, the levels of estradiol (E(2)) increased rapidly (P < 0.01), but those of IL-6, TNFalpha and type I collagen cross-linked N-telopeptides (NTX) decreased (P < 0.01). In the alendronate group, no change of levels of E(2) happened, while levels of alkaline phosphatase (ALP) and osteocalcin (BGP) increased (P < 0.05) and levels of NTX decreased (P < 0.05). There was no change of the levels of other parameters. In the calcium preparation group and control group, the levels of BMD, E(2), ALP, BGP and insulin-like growth factor I decreased (P < 0.05), while those of IL-6, TNFalpha and NTX increased (P < 0.05). CONCLUSION: Alendronate can significantly improve BMD as tibolone do. Thus it plays an important role in the treatment of osteoporosis. However calcium tablet can not prevent the loss of bone.
