Identification of some novel AHAS inhibitors via molecular docking and virtual screening approach.

Acetohydroxyacid synthase (AHAS; EC 2.2.1.6) catalyzes the first common step in branched-chain amino acid biosynthesis. This enzyme is an important target for the design of environmental-benign herbicides. Based on the crystal structure of AHAS/sulfonylurea complex, we have carried out computational screening of the ACD-3D database in order to look for novel non-sulfonylurea inhibitors of AHAS for the first time. Three novel compounds were found to inhibit plant AHAS in vitro among 14 procured compounds. One compound showed promising activity in vivo for rape root growth inhibition bioassay. This research provided useful clues for further design and discovery of AHAS inhibitors.

[Apoptosis in myocardial cells after macleaya cordata total alkaloids poisoning in rats].

OBJECTIVE: To observe pathological changes and apoptosis in rats myocardial cells after Macleaya cordata total alkaloids poisoning, and to provide some references for Macleaya cordata total alkaloids poisoning detection. METHODS: An experimental model of Macleaya cordata total alkaloids poisoning was established, and the technology of TUNEL staining was used.The results were analyzed by computer image analysis competitive system. RESULTS: Quantities of apoptosis in myocardial cells in poisoning groups were much more than those in the control groups at different tages (P<0.01). In addition the quantities of apoptosis were different after different poisoning duration. CONCLUSION: Although clinical symptoms was not obvious and could not be detected by poison analysis. Pathological changes induced by Macleaya cordata total alkaloids could be found through the apoptosis detection.

[Surgical treatment for 217 patients with bladder transitional carcinoma].

BACKGROUND & OBJECTIVE: Bladder transitional cell carcinoma (TCC) is the most common cancer among urogenital neoplasms. Surgical operation is the most effective therapy for TCC. This study was to explore surgical treatment for TCC and its clinical significance. METHODS: Clinical data, including disease-free survival, recurrence and death, of 217 TCC patients who had received different surgical procedures were retrospectively analyzed. RESULT: After a median follow-up of 30 months, 195 patients were free of disease, recurrence was observed for 56 times. Fourteen patients died: 13 of them died of progressive disease, and 1 died of renal failure. None death occurred in patients with stage T1 or grade I tumor. The overall 2-year survival rate was 89.6%. The survival rate and disease-freely survival rate were negatively correlated to stage and grade of the tumors. The prognosis of stage T1 and T4 patients had no correlation to surgical procedures. For stage T2 and T3 tumors, the prognosis of the patients who received radical cystectomy was significantly better than that of the patients who received organ sparing procedure. CONCLUSIONS: The organ sparing treatment is safe for patients with early stage and well differentiated TCC. Radical cystectomy should be done in time for patients with advanced and poorly differentiated TCC.

[Nephron-sparing surgery for localized kidney neoplasms--a report of 25 cases].

BACKGROUND & OBJECTIVE: Nephron-sparing surgery (NSS) was initially used in treating bilateral renal cell carcinoma (RCC) or RCC in a solitary functioning kidney with good effectiveness. The satisfied long-term outcome was also obtained in patients with unilateral localized RCC and a normal contralateral kidney after NSS. This paper was to report our experiences in treating localized renal tumor with NSS. METHODS: From Nov. 1999 to Dec. 2004, 25 patients were diagnosed with localized RCC preoperatively, and treated with NSS. During operation, renal circulation was temporarily interrupted in 8 patients, surface cooling of the kidney with ice slush was performed in 3 patients, frozen section pathology was performed in 16 patients. RESULTS: The operation was technically successful in all cases. The median operating time was 129 min (ranged 65-205 min). The mean blood loss was 310 ml (ranged 100-800 ml) with 1 case of blood transfusion. Except for 1 case of urinary fistula which was cured conservatively, there was no other postoperative complication. Intraoperative pathology confirmed that the surgical margin of all cases were negative. Of the 25 cases, 19 were renal clear cell carcinoma, 2 were renal granular cell carcinoma, 4 were hamartoma. The pathologic stage was pT1N0M0 in 21 patients. The mean follow-up time was 22 months (ranged 6-55 months). No local tumor recurred, and all patients survived disease-freely. CONCLUSIONS: NSS is effective in treating localized RCC. We recommend NSS to patients with unilateral localized, but not center, kidney tumor less than 4 cm and a normal contralateral kidney.

[Genomic imprinting of insulin-like growth factor II in prostate cancer and its clinical significance].

BACKGROUND & OBJECTIVE: The incidence of prostate cancer is increasing rapidly in China, and most patients are advanced cases when they were confirmed. Advanced prostate cancer is very difficult to control, although the patients could get transient recovery after total androgen blockade. Insulin-like growth factor II (IGF-II) can improve cell proliferation and inhibit cell apoptosis. Loss of imprinting (LOI) of IGF-II, or activation of the normally silent and maternally inherited allele, was discovered in some types of cancer. Our study was to explore the genomic imprinting of IGF-II in prostate cancer and its correlation to disease progression. METHODS: LOI of IGF-II in 41 specimens of prostate cancer, 27 specimens of benign prostate hyperplasia, and 13 specimens of normal prostate tissue was detected by polymerase chain reaction-based restrictive fragment length polymorphism (PCR-RFLP) analysis. RESULTS: Rates of heterozygote of IGF-II DNA were 70.7% (29/41) in prostate cancer group, 55.5% (15/27) in benign prostate hyperplasia group, and 61.5% (8/13) in normal prostate tissue group. In the specimens with IGF-II DNA heterozygote, the occurrence rate of LOI of IGF-II was significantly higher in prostate cancer than in benign prostate hyperplasia and normal prostate tissue (58.6% vs. 13.3% and 12.5%, P<0.05). LOI of IGF-II had no correlation to patients'age, serum level of prostate-specific antigen (PSA), presence of bone metastasis, and cell differentiation before endocrinotherapy. Received total androgen blockade, the 1-year progress-freely survival rate was significantly lower in the patients with LOI of IGF-II than in the patients without LOI of IGF-II (70% vs. 100%, P=0.039). CONCLUSION: LOI of IGF-II occurred frequently in advanced prostate cancer, and maybe correlated to progression of prostate cancer after total androgen blockade.

[The expression of GFAP after brain concussion in rats].

OBJECTIVE: To study the expression of GFAP and pathologic changes after rats brain concussion, so that to provide evidence on brain concussion for forensic identification. METHODS: Forty-five SD rats were divided into 3, 6, 12, 24 h and 2, 4, 7, 10 d and normal control groups in terms of different wounding time after brain concussion model established, and the expression of GFAP after rats brain concussion were then observed by using SP immunohistochemical method. RESULTS: In normal control brain, low-level GFAP expressions could be observed. After six hours' brain concussion, GFAP positive cells increased obviously. The trend reached to the peak at 7d, partly declined at 10d, then decreased gradually. CONCLUSION: Brain concussion induced the expression of GFAP. The detection of GFAP could be useful for diagnosis of brain concussion on forensic pathology, and could be a reference index for timing of injury after brain concussion.

[Optimized culture medium and fermentation conditions for lipid production by Rhodosporidium toruloides].

Culture medium and fermentation conditions for lipid production by Rhodosporidium toruloides were optimized with single factor and uniform design experiment. The best medium recipe was found with 70 g/L glucose, 0.1 g/L (NH4)2SO4, 0.75 g/L yeast extract, 1.5 g/L MgSO4. 7H2O, 0.4g/L KH2PO4, sterilized at 121 degrees C for 15 min, and then supplemented with ZnSO4 1.91 x 10(-6) mmol/L, CaCl2 1.50 mmol/L, MnCl2 1.22 x 10(-4) mmol/L and CuSO4 1.00 x 10(-4) mmol/L. The optimal fermentation conditions were as follows: 50 mL of medium (pH 6.0) in 250 mL Erlenmeyer flask with 10% inoculum (28h) under orbital shaking at 200 r/min for 120h at 30 degrees C. Under these conditions, yeast biomass accumulated lipids up to 76.1%.

[Thymus CT scan and thymoma associated antibodies in myasthenia gravis with thymoma].

OBJECTIVE: To investigate the clinical significance of the serum anti-titin, anti-ryanodine receptor (RyR) antibody level and thymus CT scan in the diagnosis of myasthenia gravis with thymoma. METHODS: Totally 32 patients with myasthenia gravis who had received thymectomy were included in the study. Abnormalities were shown under CT scan of thymus in all these patients. The relationships were studied among the pathological diagnosis, CT findings, and serum level of thymoma associated antibodies: anti-titin and anti-RyR antibodies. RESULTS: The pathological diagnosis of thymoma was made in 21 patients and thymus hyperplasia in 11 patients after operation. The sensitivity of CT scan in the diagnosis of thymomas was 90.5%, the specificity of serum thymoma associated antibodies in the diagnosis of thymoma was 100%. CONCLUSION: The thymoma-associated antibodies test is helpful in the differential diagnosis of thymomas and thymus hyperplasia; when combined with CT scan, it may achieve high sensitivity in the diagnosis of the thymoma.

Synthesis, crystal structure and herbicidal activity of mimics of intermediates of the KARI reaction.

Two mimics of the intermediate in the reaction catalyzed by ketol-acid reductoisomerase (KARI) were synthesized. Their structures were established on the basis of elemental analyses, IR, 1H NMR and GC/mass detector. The crystal structure of compound 2 was found to be a substituted dioxane, formed by the condensation of two molecules. The two compounds showed some herbicidal activity on the basis of tests using rape root and barnyard grass growth inhibition. However, the herbicidal effect was weaker in greenhouse tests.

Bioconversion of isoeugenol into vanillin by crude enzyme extracted from soybean.

Crude enzyme extracted from soybean was used to convert isoeugenol into vanillin. The effects of several factors on the bioconversion were studied. Conversion was affected by the amount of substrate and was also improved by the addition of absorbents, among which powdered activated carbon was the best. The effect of H2O2 concentration on the conversion was also studied. The optimum concentration of H2O2 was 1% (v/v). With 10 g/L of powdered activated carbon and 0.1% H2O2 added, vanillin reached a maximum concentration of 2.46 g/L after 36 h, corresponding to a molar yield of 13.3%.

[Clinical study of Chaiqin Chengqi Decoction in treating severe acute biliary pancreatitis].

OBJECTIVE: To study the therapeutic effects of Chaiqin Chengqi Decoction (CQCQD) in treating severe acute biliary pancreatitis. METHODS: Ninety patients with severe acute biliary pancreatitis were treated with CQCQD, and they were divided into two groups: early-treated group (54 patients treated with CQCQD within 3 days after the onset of severe acute biliary pancreatitis) and late-treated group (36 patients treated with CQCQD between 3 and 7 days after the onset of severe acute biliary pancreatitis). The complication incidence rate, operation rate, mortality rate and hospitalization period were examined. RESULTS: The incidence rates of encephalopathy, infection and gastrointestinal hemorrhage were lower in the early-treated group than those in the late-treated group (P<0.05). The hospitalization periods of the early- and late-treated groups were (24.9+/-18.4) days and (51.6+/-45.9) days respectively (P<0.05). The general mortality rate was 14.4%. The mortality rate of the early-treated group (7.4%) was significantly lower as compared with that of the late-treated group (25.0%) (P<0.05). The operation rate of the early-treated group (11.1%) was also significantly lower as compared with that of the late-treated group (27.8%) (P<0.05). CONCLUSION: Treating severe acute biliary pancreatitis with CQCQD in early stage may reduce the complication incidence rate, shorten the hospitalization period, and decrease the operation rate and mortality rate.

[Clinical study on severe acute pancreatitis associated with hypoalbuminemia in early stage].

OBJECTIVE: To investigate the occurring mechanism and clinical characteristics of severe acute pancreatitis (SAP) associated with hypoalbuminemia in early stage and its influence on prognosis of SAP and the preventive and therapeutic management of this disease. METHODS: One hundred and thirty-eight cases diagnosed as SAP complicated by hypoalbuminemia in early stage were accepted in our hospital from August 1, 2003 to December 31, 2004, and they were divided into 2 groups according to the level of plasma albumin: mild hypoalbuminemia (30 to 35 g/L) group and severe hypoalbuminemia (<30 g/L) group. The complications in the early stage, related parameters, and the incidence rate of infection and mortality in the later stage were evaluated respectively. RESULTS: The incidence rates of renal dysfunction, shock, cardiovascular failure and gastrointestinal hemorrhage, the score of acute physiology and chronic health evaluation II (APACHE II ) and the frequencies of pulse and breath in the severe hypoalbuminemia group were all higher than those in the mild hypoalbuminemia group (P<0.05 or P<0.01). The differences of incidence rate of hepatic failure and the scores of Ranson and Balthazar CT between these two groups had no statistical significance (P>0.05). The incidence rate of infection and the mortality in the severe hypoalbuminemia group were higher than those in the mild hypoalbuminemia group (P<0.01) in the later stage of SAP. CONCLUSION: Hypoalbuminemia in the early stage can accelerate the deterioration in pathophysiology of SAP. The lower level of the plasma albumin is in the early stage, the more complications and the higher incidence rate of infection and mortality will be in the later stage. To relieve the extent of systemic inflammatory response syndrome (SIRS) and abundant supplement of albumin, amino acid and lipid in time may be crucial to prevent the occurrence and deterioration of hypoalbuminemia.

[Initial results of intra-arterial chemotherapy for poorly differentiated bladder transitional cell carcinoma].

BACKGROUND & OBJECTIVE: Bladder cancer is the most common disease among urogenital tumors, and poorly differentiated bladder transitional cell carcinoma (BTCC) tends to recur, progress, and metastasize. This paper was to report our experiences in intra-arterial chemotherapy as adjuvant and palliative therapy for poorly differentiated BTCC. METHODS: Twenty-four patients with BTCC of grade 3 were treated with intra-arterial chemotherapy of GC regimen (gemcitabine plus cisplatin). Among them, 21 had post-operative adjuvant intra-arterial chemotherapy for 3 cycles, 3 advanced cases had palliative intra-arterial chemotherapy for 6 cycles. RESULTS: All patients were followed-up for 6-20 months. The mean follow-up was 12 months for 21 patients received adjuvant treatment, 1 developed pelvic metastasis, the others survived without evidenced tumors. Of 3 advanced cases, 1 with CR survived disease-freely for 8 months; 1 with PR survived progression-freely for 6 months; 1 with PR died of tumor relapse 13 months after chemotherapy. No serious complication was observed after intra-arterial chemotherapy. CONCLUSIONS: Intra-arterial chemotherapy is effective in managing poorly differentiated BTCC.

[Quality control methods for recombinant human endostatin].

AIM: To establish the quality control methods for recombinant human endostatin. METHODS: Biological activity was determined by endothelial cell migration assays. Peptide mapping was tested by trypsin digestion and RP-HPLC. Purity was determined by non-reduced SDS-PAGE and RP-HPLC. Other tests including molecular weight, isoelectrical point, etc. were done according to the National Requirements for Biological Products (2000). RESULTS: The method of bioassay was established and used for determining activity of endostatin. Specific activity of the three batchs of drug substance was 1.45 x 10(6), 1.57 x 10(6) and 2.73 x 10(6) u.mg-1 proteins. Peptide mappings of the three batches of drug substance were completely identified. Both purity results of the products tested by SDS-PAGE and RP-HPLC were more than 99%. CONCLUSION: The established methods can effectively control the quality of recombinant human endostatin.

Cloning and Distribution of rRP58, A Novel Neuronal Gene.

EST (AW055733) is a 3'-cDNA fragment specially expressed in rat brain. It is homologous to human and mouse RP58 gene, which is a transcriptional repressor gene. Primers were designed based on these two genes, and then two transcripts of rRP58 gene were got from male SD rat using RT-PCR method. The rRP58 protein is a C(2)H(2) type zinc finger protein, containing POZ domain at N-terminal and zinc finger domain (ZFD) at C-terminal. In addition, there is a highly acidic region between POZ and ZFD. POZ and ZFD were cloned, expressed and purified. Then, the antibodies to POZ and ZFD were prepared, and used to analyze distribution of alternative transcripts of rRP58.
