Quantitative determination of the anticancer agent tubeimoside I in rat plasma by liquid chromatography coupled with mass spectrometry.

Tubeimoside I is an important component isolated from Bolbostemma paniculatum. Tubeimoside I has been demonstrated to possess many pharmacological activities, including anti-inflammatory, antitumor, and antitumor-promoting effects. The purpose of the present study was to examine in vivo pharmacokinetics and bioavailability of tubeimoside I in rats by using a liquid chromatography coupled with mass spectrometry quantitative detection method (LC/MS). The plasma samples were deproteinated, evaporated and reconstituted in 100 microl methanol prior to analysis. The separation was performed by Waters Symmetry C18 reversed-phase column (3.5 microm, 150 mm x 2.1mm, Waters Inc., USA) and a SB-C18 guard column (5 microm, 20 mm x 4.0mm). The mobile phase was a mixture of acetonitrile and water containing 5 microM NaAc (60:40, v/v). The method was validated within the concentration range 20-5000 ng/ml, and the calibration curves were linear with correlation coefficients >0.999. The lowest limit of quantitation (LLOQ) for tubeimoside I was 20 ng/ml in 0.1 ml rat plasma. The intra-assay accuracy and precision ranged from 92.4 to 104.9% and from 5.8 to 10.5%, respectively, while inter-assay accuracy and precision ranged from 94.2 to 95.0% and from 5.1 to 8.8%, respectively. The method was further applied to assess pharmacokinetics and oral bioavailability of tubeimoside I after intravenous and oral administration to rats. The oral bioavailability of tubeimoside I is only 0.23%, which indicates that tubeimoside I has poor absorption or undergoes acid-induced degradation. Practical utility of this new LC/MS method was confirmed in pilot pharmacokinetic studies in rats following both intravenous and oral administration.

Gastroprotective effect of a traditional Chinese herbal drug "Baishouwu" on experimental gastric lesions in rats.

"Baishouwu" is an appellative name of dried root tubers from three Asclepiadaceae plants: Cynanchum auriculatum Royle ex Wight, Cynanchum bungei Decne and Cynoctonum wilfordii Maxim. In order to establish the pharmacological basis for the ethnomedicinal use of Baishouwu in gastric disorders, this study examined the effects of ethanol extracts and fractions from root tubers of Cynanchum auriculatum, Cynanchum bungei and Cynoctonum wilfordii on ethanol-, indomethacin-induced gastric lesions and histamine-induced gastric acid secretion in rats. Plant materials were collected from various areas of China. Oral administration of ethanol extract and chloroform fraction of Cynoctonum wilfordii collected from Changbai Cordillera at doses of 150 and 68 mg/kg, respectively, significantly inhibited the development of ethanol- and indomethacin-induced gastric lesions and also caused significant decrease of gastric acid secretion after histamine-induced gastric lesion. Oral administrations of ethanol extract and chloroform fraction of Cynanchum auriculatum collected from Binhai at the doses of 300 and 69 mg/kg, respectively, significantly inhibited ethanol- and indomethacin-induced gastric lesions. This study demonstrates the gastroprotective property of Baishouwu for the first time.

Benzophenanthridine alkaloids from Zanthoxylum nitidum (Roxb.) DC, and their analgesic and anti-inflammatory activities.

Seven benzophenanthridine alkaloids, 1-7, were isolated from the roots of Zanthoxylum nitidum. Among them, two novel alkaloids, named (R)-8-[(R)-1-hydroxyethyl]dihydrochelerythrine (1) and 8-methoxynorchelerythrine (2), were structurally identified as new compounds on the basis of the spectroscopic analysis. Bioactivity evaluation showed that nitidine (3), dihydrochelerythrine (4), oxyavicine (5), 8-methoxychelerythrine (6), and 8-hydroxydihydrochelerythrine (7) exhibit comparable analgesic and anti-inflammatory effects as hydrocortisone.

ent-Labdane diterpenoids from Andrographis paniculata.

Six new ent-labdane diterpenoids, 3-O-beta-D-glucopyranosyl-14,19-dideoxyandrographolide (1), 14-deoxy-17-hydroxyandrographolide (2), 19-O-[beta-D-apiofuranosyl(1-->2)-beta-D-glucopyranoyl]-3,14-dideoxyandrographolide (3), 3-O-beta-d-glucopyranosylandrographolide (4), 12S-hydroxyandrographolide (5), and andrographatoside (6), together with 17 known analogues, were isolated from the aerial parts of Andrographis paniculata. The structures of 1-6 were determined by spectroscopic data analysis. All compounds isolated were evaluated for their inhibitory activity against several bacterial and fungal strains.

Saponins from Polygala japonica and their effects on a forced swimming test in mice.

Five new triterpenoid saponins, polygalasaponins E (1), F (2), G (3), H (4), and J (5), along with eight known ones (6-13), were isolated from the aerial parts of Polygala japonica. Their structures were established by chemical and spectroscopic means. Forced swimming tests on mice showed that saponins 1 and 4 significantly reduce the immobility status by 58.1% and 51.3% at a dosage of 100 mg/kg administrated orally once daily for 5 days, respectively.

[Chemical constituents in root of Zanthoxylum nitidum].

OBJECTIVE: To study the chemical constituents of Zanthoxylum nitidum. METHOD: Column chromatography on Silica gel and Sephadex LH - 20, and recrystallization were applied for the isolation and purification of the constituents. The structures were elucidated on the basis of spectral analysis, chemical evidences and by comparison with the data reported in literature. RESULT: From the CHCl3 fraction and n-butanol fraction of the EtOH extract of the roots of Z. nitidum, 10 compounds were isolated and identified as 2, 4-dihydroxypyrimidine (1), syringic acid (2) , 2, 6-dimethoxy-1, 4-benzoquinone (3) , 4-hydroxybenzoic acid (4), ethylparaben (5), (Z)-3-(2, 3, 4-trimethoxyphenyl) acrylic acid (6), 5, 6, 7-trimethoxycoumarin (7), stigmast-9 (11) -en-3-ol (8), daucosterol (9), beta-sitosterol (10). CONCLUSION: Compounds 1-9 were isolated and identified from the roots of Z. nitidum for the first time. Furthermore, we note here the first isolation of compound 6 as a natural product.

Lancifodilactone G: a unique nortriterpenoid isolated from Schisandra lancifolia and its anti-HIV activity.

[structure: see text]. Lancifodilactone G (1), a novel, highly oxygenated nortriterpenoid featuring a partial enol structure and a spirocyclic moiety, was isolated from the medicinal plant Schisandra lancifolia. Its structure and stereochemistry were determined from extensive one- and two-dimensional NMR and mass spectral data, coupled with single-crystal X-ray analysis. Compound 1 exerted minimal cytotoxicity against C8166 cells (CC50 > 200 microg/mL) and showed anti-HIV activity with EC50 = 95.47 +/- 14.19 microg/mL and a selectivity index in the range of 1.82-2.46.

Structure characterization and possible biogenesis of three new families of nortriterpenoids: schisanartane, schiartane, and 18-norschiartane.

Four new, highly oxygenated nortriterpenoids with unique schisanartane skeletons, micrandilactones D-G (1-4), have been isolated from the leaves and stems of Schisandra micrantha, and their structures have been elucidated on the basis of extensive spectral studies. The postulated biogenetic sequences of sixteen highly oxygenated nortriterpenoids and bisnortriterpenoids with new skeletons from three Schisandra species are discussed and have been compared from a chemotaxonomic standpoint.

Cytotoxic ent-kaurane diterpenoids from Isodon eriocalyx.

Five new ent-kaurane diterpenoids, epi-maoecrystal N (1), eriocalyxin G (2), maoecrystal W (3), maoecrystal X (4), and maoecrystal Y (5), along with 22 known ones, were isolated from Isodon eriocalyx (Dunn.) Hara., and their structures were determined by spectroscopic methods. All diterpenoids, except for 3 and 13, were evaluated for inhibition of the K562, T-24, Me180, QGY-7701, and BIU87 cell lines (Table 2).

Five new triterpene glycosides from Lysimachia foenum-graecum and evaluation of their effect on the arachidonic acid metabolizing enzyme.

Five new oleanane-type triterpene saponins, named foenumosides A ( 1), B ( 2), C ( 3), D ( 4) and E ( 5), were isolated from the aerial parts of Lysimachia foenum-graecum Hance. Their structures were identified on the basis of 1D and 2D NMR techniques, including H-H COSY, HMQC, HMBC, HMQC-TOCSY, ROESY experiments as well as chemical methods. We have evaluated the cytotoxity of 1 - 5 against rat and human polymorphonuclear leukocytes and the effect of 5 on the arachidonic acid metabolizing enzyme. All compounds showed a high degree of toxicity except for compound 5, while 5 notably reduced the production of leukotriene B (4) (LTB (4)) from rat peritoneal leukocytes with an IC (50) value of 74 microM without inhibiting human elastase. Compound 5 also reduced the production of 12-HHTrE and 12-HETE by 14 % and 50 % as a measurement for cyclooxygenase-1 and 12-lipoxygenase inhibition at 100 microM.

Maoecrystal V, cytotoxic diterpenoid with a novel C19 skeleton from Isodon eriocalyx (Dunn.) hara.

Maoecrystal V (1), a novel C(19) diterpenoid possessing a unique 6,7-seco-6-nor-15(8-->9)-abeo-5,8-epoxy-ent-kaurane skeleton, was isolated from the leaves of a Chinese medicinal herb, Isodon eriocalyx. Its structure was determined by comprehensive NMR and MS spectroscopic analysis and confirmed by single-crystal X-ray diffraction study. Compound 1 showed remarkable inhibitory activity toward HeLa cells with IC(50) = 0.02 microg/mL (cis-platin: IC(50) = 0.99 microg/mL).

Coriatone and corianlactone, two novel sesquiterpenes from Coriaria nepalensis.

Both coriatone (1). a novel highly oxygenated picrotoxane-type sesquiterpene, and corianlactone (2). with an unprecedented sesquiterpene basic skeleton, named coriane, were isolated from Coriaria nepalensis Wall. The structures of 1 and 2 were determined by analysis of their two-dimensional NMR data, and the structure of 2 was confirmed by X-ray analysis. Compounds 1 and 2 showed no remarkable inhibitory activity toward K(562) cells. They are cytotoxic with IC(50) > 50 microg/mL (cis-platinim: IC(50) = 0.49 microg/mL).

Ent-kaurane diterpenoids from Isodon rubescens var. rubescens.

Five new ent-kaurane diterpenoids xindongnins H-L (1-5), together with five known ones, xindongnins A and B (6, 7), melissoidesins G (8), dawoensin A (9), and glabcensin V (10) were isolated from Isodon rubescens var. rubescens. Their structures were elucidated by spectroscopic methods including extensive 2D NMR techniques.
