Hypoxic alterations of tenocytes in degenerative tendinopathy.

The authors examined 34 tendons under the electron microscope, excised within 48 h after rupture of the tendons. The tendons did not exhibit any signs of inflammation or lipomatosis, but fine-structurally marked hypoxic alterations in the tenocytes could be seen. The degenerative (hypoxic tendinopathy appears in three phases, and the hypoxic lesions of the tendon occur at multiple foci or diffusely, and the various phases can exist simultaneously.

[Changes of the tissue CO2 and O2 tension in the healing process of crushed muscles (author's transl)].

Silastic tonometer was implanted into the space between soleus and gastrocnemius muscles of the rat. When tissue O2 and CO2 tensions returned to normal, the muscles were crushed with the skin closed. The O2 tension in the muscles was found to decrease from 44 HGMM to 10-15 hgmm following the crush, and kept it until the 4th day. After the 5th day pO2 begins to increase on the 10th day it will reach the O2 tension of the intact muscle, and keeps increasing after the 10th day. Afterwards the muscular O2 tension begins to decrease, parallel with the development of granulation tissue and will return to the original rate on the 20th day post injury. Histological picture shows that regeneration has been completed by this time too.

[The role of non-progressive myopathies in the spontaneous rupture of tendons (author's transl)].

A series of non-progressive myopathies have been found (central core disease, centronuclear myopathy, myofibrillar dysproportion) when muscles of patients with spontaneous tendon ruptures were examined. Pathological muscle actions, uneven pulling force on the tendons, disturbances in the nervous feed-back between tendons and muscles and the role of myopathies in the alteration of resting and working muscle tone by affecting the type--I. muscle fibers are thought to be pathogenic factors in the spontaneous tendon ruptures associated with non-progressive myopathies.

Experiences about the application of gelatin-resorcinol-formaldehyde tissue adhesive in experimental ruptured liver injury.

Ruptured liver injury was produced in dogs. For arresting the bleeding and for the tissue unifying GRF adhesive was used instead of the traditional wound treatment. The experiences obtained during the wound healing were positive. On the 4th week the adhesive was well markedly emerged from the liver surface, but on the 20th postoperative week the adhesive could not be noticed even by microscopy. The necrosis spread over 10-15 cell lines has nearly disappeared by the end of the 15th week. By this time the granulation tissue filling up the wound transformed into native connective tissue.

Histochemical and ultrastructural study of adult human tendon.

The authors have studied the enzymhistochemical and ultrastructural pictures of tenocytes of adult human tendons. High succinate dehydrogenase, cytochrome oxidase, TPN-diaphorase, lactate dehydrogenase and glucose-6-phosphate dehydrogenase activity were found, as indicated both oxidativ, anaerobic and pentose-phosphate shung activity. Phosphorylase and glutamate dehydrogenase activity was medial, lipase and alcaline phosphatase activity was slight. In tenocytes well developed rough endoplasmic reticulum and GOLGI apparatus, large amount of free ribosomes were found.

Myofibrillar proteins, enzyme activity and fibre types in human skeletal muscle.

The myofibrillar proteins and relative LC-3 content of myofibrils prepared from five different, left and right muscles of the human forearm were studied by electrophoresis in 10% SDS-polyacrylamide gels followed by densitometry. The specific K-activated ATPase activity of the myofibrillar myosin of the myofibrils was determined and the distribution of the fibre types was analyzed by histochemical methods. The qualitative pattern of the myofibrillar proteins was found to be identical in the different muscles; in the other parameters, however, a variation in a narrow range was observed. The relative LC-3 content, the ATPase activity and the type II fibre content of the right side muscles were always higher than those of the corresponding left side muscles.

[Changes in the skeletal musculature due to tendon rupture].

Alterations of the sceletal muscles after spontaneous rupture of the tendon were studied. It was established that after the rupture of the tendon in the muscle activity of the succinedehydrogenase, myofibrillar ATP-ase, creatinphosphokinase and aldolase markedly decreases. After the rupture of the tendon alteration of the proportion of various types of fibers may also be observed i.e. the rate of the fibres of type I considerably decreases. Electron microscopic findings indicate that alterations of the musculature are diffuse first of all elements of contractility are damaged: atrophy and rupture of them can be seen. Fibres of normal ultrastructure were not found at all, which indicates that both types of fibres I and II are equally damaged.

Effect of experimental hypoxia on the ubiquinone level of skeletal muscle in rabbits.

Rabbits were kept for 2 and 4 hours in a chamber containing a gas mixture of 92% N2 and 8% O2. The ubiquinone level in the musculus quadriceps femoris of the animals was determined. Rabbits after 4 hours hypoxia were retaken into normal conditions for 2 hours before killing. Biochemical findings revealed a significant decrease in the ubiquinone activity during hypoxia independently from duration of hypoxia. 2 hours after hypoxia the ubiquinone activity became normal. The slight alterations biochemically determined can not be followed unambiguously by histochemical methods, but the microscopic studies have revealed that the red fibres of mixed musculus quadriceps femoris are responsible for the quantitative changes.

[Specific fibre compositon of human hand and arm muscles].

The authors examined the fiber composition of the finger, hand, and arm muscles. It is established that the type of fiber composition is not influenced by the mechanical employment of the hand- and arm muscles. The fiber composition of the finger muscles, however, is different in the two hands, in that the amount of fiber type II is significantly higher than that of the fiber type I in the dominant hand. In case of the non-dominant hand the fiber type I is predominant.
