Tests of intestinal function.

Abstract

The most important tests for investigating the functions of the small bowel are faecal-fat analysis, the determination of xylose tolerance, vitamin-B12 absorption, and 14C-glycoholate transport, barium examination of the small bowel, and small bowel biopsy. With these tests, most disorders of digestion and of absorption can be adequately investigated and diagnosed.

Clostridium scindens: history and current outlook for a keystone species in the mammalian gut involved in bile acid and steroid metabolism.

Clostridium scindens is a keystone bacterial species in the mammalian gut that, while low in abundance, has a significant impact on bile acid and steroid metabolism. Numerous studies indicate that the two most studied strains of C. scindens (i.e. ATCC 35704 and VPI 12708) are important for a myriad of physiological processes in the host. We focus on both historical and current microbiological and molecular biology work on the Hylemon-Björkhem pathway and the steroid-17,20-desmolase pathway that were first discovered in C. scindens. Our most recent analysis now calls into question whether strains currently defined as C. scindens represent two separate taxonomic groups. Future directions include developing genetic tools to further explore the physiological role of bile acid and steroid metabolism by strains of C. scindens and the causal role of these pathways in host physiology and disease.

Evaluation of the lipophilicity of bile acids and their derivatives by thin-layer chromatography and principal component analysis.

The lipophilic character of bile acids and their glyco- and tauro-conjugates was studied. The classical R(Mo) values were measured by means of reversed thin-layer chromatography using a mixture of methanol-water as the solvent system and compared with the factors scores obtained by principal component analysis based also onto the TLC-retention data. The reliability of the factor scores values as lipophilic indices are shown by their high correlation with the classical R(Mo) values. In addition, a better correlation was observed between scores corresponding to the first principal components and the partition coefficients (log P) of bile acids. Finally, the "lipophilicity chart" described by the first two components has the effect of separating compounds from each other most effectively from the congeneric aspect point of view.

Design and synthesis of novel chiral dendritic species derived from bile acids.

Bile acids have been used for the first time as the building block for the construction of dendritic units. Orthogonally functionalized 7-deoxycholic and cholic acid derivatives were synthesized. The construction of a bile acid heptamer, a nonamer, and a decamer using the convergent strategy are described in detail. Chromatographic, spectral, and optical properties of these molecules have been investigated. Molecular modeling suggests that these molecules have globular shapes with nanometric dimensions.
