[IR studies on deuterated polyester polyurethaneurea elastomers].

Abstract

Segmented polyester polyurethaneurea (PUU) based on 4,4'-disphenylmethane diisocyanate (MDI), ethylene diamine (ED) and polybutylene adipate (PBA) elastomers and their deuterated polymers were studied by means of IR spectroscopy. The results indicated that IR bands of hydrogen bonded N-H vibrations involved in PUU were greatly affected by deuteration. The bands which were sensitive to deuteration were those near 3,332, 3,190, 1,600, 1,539, 1,317, 1,257 and 1,230 cm-1, respectively. In addition, it was also found that PUU samples with different hard segment contents and polyester soft segment molecule weights were similarly affected by deuteration in degree.

Effects of organic solvents on the scission energy of rodlike micelles.

The linear viscoelastic response of erucyl bis(hydroxyethyl) methylammonium chloride with added KCl has been studied as a function of temperature and nonpolar solvent addition. The plateau modulus is independent of temperature from 25 to 40 degrees C, in contrast to previous studies with salicylate counterions that showed a plateau modulus increasing with temperature over this range. The average micelle length, L, predicted by the model of Cates, depends experimentally on Escis/kBT, where Escis is the scission energy of the chain and kBT is the Boltzmann constant times the absolute temperature. With ethanol addition, the calculated average contour length, L, decreases by a factor of 4 as ethanol concentration varies from 0 to 1.3 M. This corresponds to an apparent energy for scission, Escis, decrease from 81 +/- 8 to 74 +/- 7 kJ/mol. On the other hand, only 80 mM of hexane is required to cause a decrease in Escis to the same level, and for hexane addition levels above 70 mM a disruption in the plateau modulus indicates the disruption of the rodlike structure. The correspondence between the effect of temperature and the effect of solvent addition allows the development of "solvent/temperature" superposition rules to predict the rheology of these viscoelastic fluids at elevated temperatures.

Viscoelastic properties of single polysaccharide molecules determined by analysis of thermally driven oscillations of an atomic force microscope cantilever.

We report on single molecule measurements of the viscoelastic properties of the polysaccharide dextran using a new approach which involves acquiring the power spectral density of the thermal noise of an atomic force microscope cantilever while holding the single molecule of interest under force-clamp conditions. The attractiveness of this approach when compared with techniques which use forced oscillations under constant loading rate conditions is that it is a near-equilibrium measure of mechanical response which provides a more relevant probe of thermally driven biomolecular dynamics. Using a simple harmonic oscillator model of the cantilever-molecule system and by subtracting the response of the free cantilever taking into account hydrodynamic effects, the effective damping zetamol and elastic constant kmol of a single molecule are obtained. The molecular elasticity measured by this new technique shows a dependence on applied force that reflects the chair-boat conformational transition of the pyranose rings of the dextran molecule which is in good agreement with values obtained directly from the gradient of a conventional constant loading rate force-extension curve. The molecular damping is also seen to follow a nontrivial dependence on loading which we suggest indicates that it is internal friction and not work done on the solvent that is the dominant dissipative process.

Measuring masses of single bacterial whole cells with a quadrupole ion trap.

A novel method has been developed to precisely measure the masses of single bacterial whole cells using a quadrupole ion trap as an electrodynamic balance. The bacterial cells were introduced into the ion trap by matrix-assisted laser desorption/ionization, confined in space by audio frequency ac fields, and detected by elastic light scattering. Mass measurement accuracy approaching 0.1% was achieved for Escherichia coli K-12 with a mass distribution of +/-3% from 60 repetitive measurements of the particles and their clusters. This is the first high-precision mass measurement reported for any intact microorganisms with masses greater than 1 x 1010 Da. The method opens new avenues for high-precision mass measurement of single microbial particles and offers an alternative approach for rapid identification of microorganisms by mass spectrometry.
