Observing bacterial activity interferometrically.

It is shown that bacterial activity, even of slowly growing species, can be detected by precise interferometric measurements of refractive index changes of the culture medium. The bacteria-containing sample is kept in an isothermal block together with a reference liquid without bacteria. The biological activity is obtained from the difference of the index changes of these samples. Experiments were performed with Bacilo Calmette-Guérin. The order of magnitude of the observed total refractive index change was compatible with theoretical estimates based on the amount of available oxygen. An unexpected positive index change during the lag phase was observed, which might permit fast diagnostics in medical applications. This technique may provide cheap and quick tests of bacterial susceptibility with respect to antibiotics.

High-speed adaptive interferometer for optical coherence-domain reflectometry through turbid media.

Two-wave mixing in a dynamic holographic film acts as the adaptive beam combiner in a short-coherence interferometer that performs optical coherence-domain reflectometry (OCDR) through turbid media. This approach combines the high spatial resolution and sensitivity of coherence-domain reflectometry with photorefractive quantum-well-based adaptive homodyne detection. A depth resolution of 28 microm and penetration through 16 mean free paths in a turbid medium have been obtained in this adaptive OCDR application.

Delay and dispersion characteristics of a frequency-domain optical delay line for scanning interferometry.

The reflective frequency-domain optical delay line employing a diffraction grating, a lens, and a tiltable mirror has emerged as a device particularly suitable for interferometry and optical coherence tomography. The device is comprehensively described, both theoretically and experimentally, in the context of interferometry. The variations of phase and group delay produced by the device as well as its dispersive properties are described and demonstrated experimentally.
