Occurrence of chlorophyll precursors in leaves of cabbage heads--the case of natural etiolation.

In the interior of cabbage (Brassica oleracea) heads (kale, white cabbage, Brussels sprouts), natural leaf etiolation takes place due to a limited light access and chlorophyll biosynthesis is inhibited in a consequence. Instead, apart from carotenoids, whose biosynthesis is not light-dependent, chlorophyll precursors accumulate, mainly protochlorophyllide and to a smaller extent also chlorophyllides a and b. Protochlorophyllide was also detected in green, light-exposed leaves of heads of all the investigated cabbage varieties. Protochlorophyll was not found in the investigated leaves. The analysis of xanthophylls composition showed that the central leaves of kale and white cabbage heads contain relatively high amounts of trans-neoxanthin and lutein epoxide which are found only in trace amounts in green leaves. This is the first systematic study on natural occurrence of chlorophyll biosynthesis precursors in different cabbage varieties.

Quantitative determination of intact glucosinolates in broccoli, broccoli sprouts, Brussels sprouts, and cauliflower by high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry.

Many methods have been proposed to analyze glucosinolates, a class of phytochemicals whose breakdown products are thought to be responsible for an improvement in health; however, few are quantitative and many are time consuming. A selective and sensitive quantitative method for direct determination of intact glucosinolates was developed using liquid chromatography coupled to electrospray ionization tandem mass spectrometry with selected reaction monitoring detection. Detection limits for glucoiberin, sinigrin, progoitrin, glucoerucin, and glucotropaeolin were 1.75, 1.38, 1.36, 0.6, and 0.63 pmol, respectively. Intraassay precision of the method was within 10% for each compound. The method was successfully applied to quantify 10 individual glucosinolates in broccoli, broccoli sprouts, Brussels sprouts, and cauliflower. The advantage of the proposed method includes analysis of individual intact glucosinolates rather than the conversion to desulfoglucosinolates, an increased selectivity through the use of mass spectrometry, and a 10-fold improvement in detection sensitivity over conventionally used HPLC techniques.

Micro pulsed radio-frequency electroporation chips.

Electroporation (EP) is one of the most important physical methods in biotechnology, which employs electrical pulses to transiently permeabilize cell membranes. In this study, a new micro pulsed radio-frequency electroporation cell (microPREP) chip was fabricated using a lift-off technique and SU-8 photolithography. The biological tests were carried out using three different plant protoplasts (cabbage, spinach and oil rape) on the micro EP chip and a pulsed RF electric field was applied to the microchip. The variations of fluorescent intensity and cell viability as functions of the electric pulse amplitude and duration time during the electroporation process were studied in detail at the single-cell level. Using such chip design and test method, one can easily optimize the efficiency and cell viability. Also, a large amount of statistical data can be quickly obtained. Finally, results of this parametric study were presented in the "phase diagram", from which the critical electric field for inducing single-cell electroporation under different conditions can be clearly determined.
