Calculation on ultrafiltration for pullulan solution.

Abstract

Ultrafiltration is an important step in isolation and purification of pullulan. Here a new formula for ultrafilter calculation is derived from experiments about the relationship between the ultrafiltrate rate and the concentration of pullulan solution. It can be used for calculating and designing of ultrafilter under defined production scale and its related technical parameters.

Translation Co-factor PABP-interacting protein 11 moonlights as a transcriptional activator to modulate callose synthesis gene expression.

The development of rice fertility is a complex process, which is precisely regulated by numerous genes. In this study, we cloned and characterized OsPAIP11, a PABP-interacting protein that functions as an auxiliary factor in translation initiation. The ospaip11 exhibited multiple defects, including impaired callose synthesis, delayed tapetum apoptosis, and abnormal pollen wall development, which are essentially consistent with the phenotype of the allelic mutant dcet1. Subcellular localization analysis revealed that OsPAIP11 is localized in both the cytoplasm and nucleus. Interestingly, further investigation demonstrated that the RRM2 domain of OsPAIP11 exhibits transcriptional activation activity. Moreover, OsPAIP11 directly binds to the promoter of the callose synthesis-related genes GLUCAN SYNTHASE-LIKE 5 (OsGSL5) and OsGAMYB, thereby regulating their transcription and influencing callose biosynthesis during pollen development. Additionally, OsPAIP11 also interacts with the translation initiation factor and auxiliary factors. These findings suggest that OsPAIP11 modulates male fertility primarily by regulating the transcription of callose synthesis-related genes and may also participate in the translation process.

Molecular and crystal structure of (1----3)-alpha-D-glucan triacetate.

A crystal and molecular structure for GTA I, the low temperature polymorph of (1----3)-alpha-D-glucan triacetate, is proposed on the basis of X-ray diffraction analysis of well-oriented films, combined with stereochemical model refinement. The unit cell is monoclinic with parameters a = 30.17 A, b = 17.42 A, c (fibre axis) = 12.11 A, and beta = 90 degrees C. The probable space group is P2(1) with b axis unique. Six molecular chains pass through the unit cell with alternating polarity and with three independent chains comprising the asymmetric unit. The chain axes are located in a hexagonal packing arrangement. The chain backbone conformation is a left-handed, three-fold helix, but all nine O(6) acetyl groups of the asymmetric unit are in non-equivalent rotational positions. The most probable structure is indicated by X-ray residuals R = 0.261 and R" = 0.283, based on 62 reflection intensities (41 observed and 21 unobserved).

Mechanism of water-insoluble glucan synthesis in Streptococcus sobrinus.

Synthesis of water-insoluble glucan (IG) by 1,3-alpha-D-glucan synthase from Streptococcus sobrinus was examined using methylation analysis. The purified enzyme was incubated with sucrose and dextran T2000 (DT2000) for a given time and only IG was harvested by centrifugation. The remaining supernatant was incubated again, and IG was obtained. By repeating the above method using the residual supernatant, we obtained 5 varieties of IG precipitated in different periods. These IGs were methylated and examined using gas chromatograph mass spectrometry. The DT2000 water-insolubilized in the early reaction stage tended to have a highly ramified structure, with 1,3-alpha-D-glucan on a 1,6-alpha-D-glucan chain as the main chain. On the contrary, the DT2000 water-insolubilized in the late stage tended to have sparse side chains of 1,3-alpha-D-glucan which elongated with incubation. Specifically, the greater the number of side chains, the sooner the DT2000 was insolubilized. These results suggest that water-insolubilization of the water-soluble glucan not only depends on the increase of the ratio of 1,3-alpha-glucoside linkages to 1,6-alpha-glucoside linkages but also on the degree of branching of the 1,3,6-alpha-branched glucoside linkages.
