[Decrease of singlet oxygen chemiluminescence by the presence of carnosine].

Abstract

To test antiradical medicines effect the chemical production of singlet oxygen (NaClO + H2O2) was investigated. The quantity singlet oxygen chemiluminescence was decreased in the presence of Japanese Catalin and Chine Baineiting, antirheumatic Voltaren and less strong Finish Catachrome and Carnosine. American Quinax does not possess such an effect. One of the possible starting mechanisms causing different diseases (atherosclerosis, cataract etc.) is destruction of biomembranes by active forms of oxygen.

Synthesis and study of a new adenine-acridine tandem, inhibitor of exonuclease III.

A new heterodimer adenine-chain-acridine containing a mixed amido-guanidinium linker chain was synthesized. To achieve the synthesis a new method of introduction of aminoalkyl chain at position 9 of adenine was designed. The heterodimer interacts specifically with the abasic sites in DNA and inhibits the major base excision repair enzyme in Escherichia coli, Exonuclease III.

Lucigenin: redox potential in aqueous media and redox cycling with O-(2) production.

The use of lucigenin luminescence as a measure of ¿O-(2) has been questioned because lucigenin has been shown to be capable of mediating the production of O-(2). This being the case, lucigenin can signal the presence of O-(2) even in systems not producing it in the absence of lucigenin. The reduction potential of lucigenin should be in accord with its ability to mediate O-(2) production; but it has not heretofore been measured in aqueous media. The problems facing such measurement are the insolubility of the divalently reduced form, which deposits on the electrode, and the slow conformational transition that follows the second electron transfer and which interferes with reversibility. We have now used rapid scan cyclic voltammetry to determine that the reduction potential for lucigenin is -0.14 +/- 0.02 V versus the normal hydrogen electrode. This value applies to both the first and the second electron transfers to lucigenin and it is in accord with the facile mediation of O-(2) production by this compound.

Acridines

Inhibition of cryptosporidium parvum in vitro by 9-(alkylthio)acridine derivatives.

The synthesis of several acridinic thioethers is described. Compounds prepared were tested in vitro as potential drugs against the opportunistic infection known as cryptosporidiosis. With a view to predict activity, the quantitative structure-activity relationships were investigated. Correlations between experimental data and either log P or pKa are discussed.
