Aldehyde methacrylates derived from hydroxybenzaldehydes.

Abstract

Three crystalline aldehyde methacrylates with low melting points were synthesized from the readily available, isomeric hydroxybenzaldehydes and 2-bromoethyl methacrylate. These monomers can be purified by recrystallization and liquified by admixture in various proportions to obtain polymerizable liquids having workable viscosities at room temperature. These monomers may be used alone or as blends with other methacrylates since they are miscible and copolymerizable with the usual dental monomers. Also, they should be studied with other functional methacrylates designed to promote adhesion via the mineral phase to determine if this synergistic approach can improve the adhesion of dental resins to dentin. These aldehyde methacrylates, their mixtures, polymers and copolymers merit evaluation as adhesion-promoting agents for proteinaceous substrates such as bone and dentin.

Inhibitors of calpain activation (PD150606 and E-64) and renal ischemia-reperfusion injury.

Calpain activation has been implicated in the development of ischemia-reperfusion (I-R) injury. Here we investigate the effects of two inhibitors of calpain activity, PD150606 and E-64, on the renal dysfunction and injury caused by I-R of rat kidneys in vivo. Male Wistar rats were administered PD150606 or E-64 (3mg/kg i.p.) or vehicle (10%, v/v, DMSO) 30min prior to I-R. Rats were subjected to bilateral renal ischemia (45min) followed by reperfusion (6h). Serum and urinary biochemical indicators of renal dysfunction and injury were measured; serum creatinine (for glomerular dysfunction), fractional excretion of Na(+) (FE(Na), for tubular dysfunction) and urinary N-acetyl-beta-d-glucosaminidase (NAG, for tubular injury). Additionally, kidney tissues were used for histological analysis of renal injury, immunohistochemical analysis of intercellular adhesion molecule-1 (ICAM-1) expression and nitrotyrosine formation. Renal myeloperoxidase (MPO) activity (for polymorphonuclear leukocyte infiltration) and malondialdehyde (MDA) levels (for tissue lipid peroxidation) were determined. Both PD150606 and E-64 significantly reduced the increases in serum creatinine, FE(Na) and NAG caused by renal I-R, indicating attenuation of renal dysfunction and injury and reduced histological evidence of renal damage caused by I-R. Both PD150606 and E-64 markedly reduced the evidence of oxidative stress (ICAM-1 expression, MPO activity, MDA levels) and nitrosative stress (nitrotyrosine formation) in rat kidneys subjected to I-R. These findings provide the first evidence that calpain inhibitors can reduce the renal dysfunction and injury caused by I-R of the kidney and may be useful in enhancing the tolerance of the kidney against renal injury associated with aortovascular surgery or renal transplantation.

2-Haloacrylate reductase, a novel enzyme of the medium chain dehydrogenase/reductase superfamily that catalyzes the reduction of a carbon-carbon double bond of unsaturated organohalogen compounds.

A soil bacterium, Burkholderia sp. WS, grows on 2-chloroacrylate as the sole carbon source. To identify the enzymes metabolizing 2-chloroacrylate, we carried out comparative two-dimensional gel electrophoresis of the proteins from 2-chloroacrylate- and lactate-grown bacterial cells. As a result, we found that a protein named CAA43 was inducibly synthesized when the cells were grown on 2-chloroacrylate. The CAA43 gene was cloned and shown to encode a protein of 333 amino acid residues (M(r) 35,788) that shared a significant sequence similarity with NADPH-dependent quinone oxidoreductase from Escherichia coli (38.2% identity). CAA43 was overproduced in E. coli and purified to homogeneity. The purified protein catalyzed the NADPH-dependent reduction of the carbon-carbon double bond of 2-chloroacrylate to produce (S)-2-chloropropionate, which is probably further metabolized to (R)-lactate by (S)-2-haloacid dehalogenase in Burkholderia sp. WS. NADH did not serve as a reductant. Despite the sequence similarity to quinone oxidoreductases, CAA43 did not act on 1,4-benzoquinone and 1,4-naphthoquinone. 2-Chloroacrylate analogs, such as acrylate and methacrylate, were also inert as the substrates. In contrast, 2-bromoacrylate served as the substrate. Thus, we named this novel enzyme 2-haloacrylate reductase. This study revealed a new pathway for the degradation of unsaturated organohalogen compounds. It is also notable that the enzyme is useful for the production of (S)-2-chloropropionate, which is used for the industrial production of aryloxyphenoxypropionic acid herbicides.

Amphipathic and membrane-destabilizing properties of the cationic acrylate polymer Eudragit E100.

The cationic acrylate polymer Eudragit E100 (E100) produces a biphasic effect on the stability of casein micelles disrupting their internal structure. These results suggested that this polymer could have some amphipathic character. Therefore, in this study the polymer was characterized with respect to its interaction with different amphipathic systems (bile-acid micelles, lipoproteins and liposomes), cell membranes (red blood cells) and virus membranes (Herpes simplex type 2 virus). As with caseins, a biphasic effect was observed with bile acids with a precipitation phase at low polymer/bile acid ratio and a solubilization phase when the polymer concentration was increased. Upon interaction with human plasma, an important reduction in cholesterol and triglycerides was observed upon remotion of E100 by a rise in pH to 8.5 and centrifugation. In agreement with this finding, an important reduction in plasma lipoproteins was observed upon its treatment with E100 and further remotion by pH rise and centrifugation. However, the amount of the major protein components of human plasma and the activity of several enzymes and antibodies were not affected by their treatment with E100. The membrane-destabilizing properties of E100 were confirmed by its lytic activity on liposomes and red blood cells and by an important antiviral effect of E100 on Herpes simplex virus type 2. Altogether, these results show that, despite its water solubility and cationic character, E100 displays a significative amphipathic and membrane-destabilizing character with potential biotechnological applications.
