Nine rheumatoid factor assays compared.

Abstract

In this study we have investigated five quantitative and three semi-quantitative rheumatoid factor assays and the Rose-Waaler assay in 120 patients suffering from rheumatoid arthritis and in 76 with other systemic diseases. All tests measure the IgM anti-IgG antibodies. The correlations between the quantitative tests were all higher than 0.86 and much better than between the quantitative and semi-quantitative tests and the semi-quantitative tests themselves (r between 0.22 and 0.85). The within run and between run precision studies for the quantitative tests showed CV values lower than 16%. In spite of the standardisation on the WHO and the Center of Disease Control Reference Preparation we found important differences in patient results. From an analytical point of view, the quantitative assays for rheumatoid factors show certain advantages over the traditional haemagglutination tests.

Protection of mice against homologous or heterologous infections with antiserum mixture to the predominant variable antigen type repertoire of Trypanosoma evansi YNB stock.

The objective of this study was to test a hypothesis that the predominant variable antigen type (VAT) repertoire of a single stock of Trypanosoma evansi was limited and small. It was further assumed that six rabbits could produce all antibodies against the predominant VAT repertoire of a stock of T. evansi and the antiserum mixture from the six rabbits containing all the antibodies could completely protect mice against any homologous stock infections and partially protect mice against some heterologous stock infections. Mice were each intraperitoneally infected with 100 parasites of clone-derived and non-clone-derived populations of the YNB stock, Kazakhstan strain or Vietnam strain of T. evansi, and treated with the antiserum mixture when trypanosomes had been detected in the blood. All of the 10 mice infected with either non-clone-derived or clone-derived populations of the YNB stock survived, and some (4/10) of mice infected with the heterologous Kazakhstan strain survived, while all those (10/10) infected with the heterologous Vietnam strain died. These results support the hypothesis that the predominant VAT repertoire of a single stock of T. evansi was limited and small, and have important implications in the consideration of treating human trypanosomosis due to drug resistant strains with antiserum mixture.

Evaluation of four serological techniques to determine the seroprevalence of Neospora caninum in foxes (Vulpes vulpes) and coyotes (Canis latrans) on Prince Edward Island, Canada.

The objectives of this study were (1) to evaluate the performance and agreement of serological assays (ELISA, IFAT, Neospora caninum agglutination test and immunoblot) using reference sera and field sera from foxes and coyotes and (2) to estimate the N. caninum seroprevalence in foxes and coyotes on Prince Edward Island, Canada. With fox and coyote reference sera the test performance of the ELISA, IFAT and IB was excellent (100% sensitivity and specificity). NAT showed a low sensitivity (50%). Serum was collected from 201 coyotes and 271 foxes. The seroprevalence observed in the different assays ranged from 0.5 to 14.0% in coyotes and 1.1 to 34.8% in foxes. The seroprevalence, when taking more than one test positive as cut-off value was 3.3 and 1.1% for coyotes and foxes, respectively. From the N. caninum-positive group, all coyotes were older than 3 years. Agreement among assays (measured as prevalence-adjusted bias-adjusted kappa) using the field sera ranged from 0.17 to 0.97. Best agreement was observed between ELISA and IFAT, poor agreement was observed between NAT and the other assays. Positive agreement was moderate to poor among all assays utilized in this study. Although the seroprevalence observed was low, N. caninum antibodies are present in foxes and coyotes on Prince Edward Island (PEI) and their role in the N. caninum epidemiology needs further study.

Identification of the capsular polysaccharides in Staphylococcus aureus clinical isolates by PCR and agglutination tests.

Staphylococcus aureus is a major cause of nosocomial and community-acquired infections. The predominance of two capsular polysaccharides, types 5 and 8, on the surface of clinical isolates led to the development of a conjugate vaccine (StaphVAX) based on capsular polysaccharides types 5 and 8 conjugated to a carrier protein. We have studied the capsular phenotypes and genotypes of 195 isolates representative of all clinical syndromes that encompassed both hospital and community-acquired infections. These isolates were mainly detected in France between January 2001 and December 2004. In this population, most of clinical isolates (87%) expressed either capsular polysaccharide type 5 (42%) or 8 (45%), whereas 13% were nontypeable by the serotyping method with antibodies specific to capsular polysaccharide type 5 or 8. These 26 nontypeable strains were further serotyped and were demonstrated to express the cell wall surface antigen 336, a polyribitol phosphate N-acetylglucosamine, which resembles cell wall teichoic acid. Among methicillin-resistant Staphylococcus aureus (MRSA) strains, we found a predominance of serotype 5 for 64% of strains, whereas MSSA isolates were predominantly capsular serotype 8 (60%). All S. aureus clinical isolates included in the present study have been investigated by PCR method, demonstrating that all isolates carried either the cap5 or the cap8 locus.
