Limits to exercise performance: some ideas from comparative studies.

Abstract

Examples of exercise performance and metabolic scope in non-human mammalian species are considered from the point of view of problems and questions which may provide insights into evolutionary processes influencing adaptations to muscular activity. Consideration of both aerobic and anaerobic performance is required. Some recent approaches, notably that concerned with the concept of symmorphosis, the integration of design for the accommodation of variations in activity, show promise of new ways for comparative investigations of the adjustments to exercise.

Some controls on oligosaccharide utilization by yeasts: the physiological basis of the Kluyver effect.

Many yeasts can aerobically catabolize exogenously supplied glycosides that are hydrolysed in the cytosol, but few do so anaerobically. This is so, even for yeasts that use one or more of the component hexoses anaerobically. The phenomenon, called the Kluyver effect, appears to be brought about by a combination of the following four factors: (i) fast transport of the glycosides into the cells involves proton symport and seems to require aerobiosis, so, under anaerobic conditions, the glycosides enter the cells much more slowly. This is probably because there is less ATP produced anaerobically than aerobically and, consequently, insufficient to supply the proton pump optimally, which is necessary to maintain proton symport; (ii) in addition, anaerobically, the transport carrier may have a lower substrate affinity; (iii) glycosidases generally have low substrate affinities; and (iv) the consequence of (i), (ii) and (iii) is a lowering of glycolytic flux and this deactivates pyruvate decarboxylase.

Characterization of components of the anaerobic ribonucleotide reductase system from Escherichia coli.

Anaerobic growth of Escherichia coli induces an oxygen-sensitive ribonucleoside triphosphate reductase system, different from the aerobic ribonucleoside diphosphate reductase (EC 1.17.4.1) of aerobic E. coli and higher organisms (Fontecave, M., Eliasson, R., and Reichard, P. (1989) Proc. Natl. Acad. Sci. U.S.A. 86, 2147-2151). We have now purified and characterized two proteins from the anaerobic system, provisionally named dA1 and dA3. dA3 is the actual ribonucleoside triphosphate reductase; dA1 has an auxiliary function. From gel filtration, dA1 and dA3 have apparent molecular masses of 27 and 145 kDa, respectively. In denaturing gel electrophoresis, dA3 gives two bands of closely related polypeptides with apparent molecular masses of 77 (beta 1) and 74 (beta 2) kDa. Immunological and structural evidence suggests that beta 2 is a degradation product of beta 1 and that the active enzyme is a dimer of beta 1. dA1 activity coincides on denaturing gels with a band of 29 kDa and thus appears to be a monomer. The reaction requires, in addition, an extract from E. coli heated for 30 min at 100 degrees C. Potassium is one required component, but one or several others remain unidentified and are provisionally designated fraction RT. With dA3, dA1, RT, and potassium ions, CTP reduction shows absolute requirements for S-adenosylmethionine, NADPH (with NADH as a less active substitute), dithiothreitol, and magnesium ions, and is strongly stimulated by ATP, probably acting as an allosteric effector. Micromolar concentrations of several chelators inhibit CTP reduction completely, suggesting the involvement of (a) transition metal(s).

Oscillatory metabolism of Saccharomyces cerevisiae in continuous culture.

Short-period (40-50 min) synchronized metabolic oscillation was found in a continuous culture of yeast Saccharomyces cerevisiae under aerobic conditions at low-dilution rates. During oscillation, many parameters changed cyclically, such as dissolved oxygen concentration, respiration rate, ethanol and acetate concentrations in the culture, glycogen, ATP, NADH, pyruvate and acetate concentrations in the cells. These changes were considered to be associated with glycogen metabolism. When glycogen was degraded, the respiro-fermentative phase was observed, in which ethanol was produced and the respiration rate decreased. In this phase, the levels of intracellular pyruvate and acetate became minimum, ATP became high and intracellular pH at its lowest level. When glycogen metabolism changed from degradation to accumulation, the respiratory phase started, during which ethanol was re-assimilated from the culture and the respiration rate increased. Intracellular pyruvate and acetate became maximum, ATP decreased and the intracellular pH appeared high. These findings may indicate new aspects of the control mechanism of glycogen metabolism and how respiration and ethanol fermentation are regulated together under aerobic conditions.
