Swa: a subdivision.

Abstract

For some time, anomalous serological reactions have been observed when the same anti-Swa sera are tested against red cells from different individuals reported as Sw(a+). A comparative collaborative study using the same collection of Sw(a+) cells and anti-Swa sera was undertaken by 4 reference laboratories, and it was found that Swa represents a heterogeneous group of antigens that can be subdivided into two categories. Both categories, Sw(a+) 700:41 and Sw(a+) 700:-41, were shown to be inherited.

Origins of the Indo-Europeans: genetic evidence.

Two theories of the origins of the Indo-Europeans currently compete. M. Gimbutas believes that early Indo-Europeans entered southeastern Europe from the Pontic Steppes starting ca. 4500 B.C. and spread from there. C. Renfrew equates early Indo-Europeans with early farmers who entered southeastern Europe from Asia Minor ca. 7000 BC and spread through the continent. We tested genetic distance matrices for each of 25 systems in numerous Indo-European-speaking samples from Europe. To match each of these matrices, we created other distance matrices representing geography, language, time since origin of agriculture, Gimbutas' model, and Renfrew's model. The correlation between genetics and language is significant. Geography, when held constant, produces a markedly lower, yet still highly significant partial correlation between genetics and language, showing that more remains to be explained. However, none of the remaining three distances--time since origin of agriculture, Gimbutas' model, or Renfrew's model--reduces the partial correlation further. Thus, neither of the two theories appears able to explain the origin of the Indo-Europeans as gauged by the genetics-language correlation.

[Genetic traits in the area of Bodrogköz].

In 1984 a late malaria endemic area, called Bodrogköz was studied. This was a reexamination of the population genetic work performed by Walter, Nemeskéri. In six villages of Bodrogköz 328 persons were tested for AB0, Rh blood groups, haptoglobins, haemoglobin concentration, haematocrit, erythrocyte amount, the MCV, the MCH and the G-6-PD were analyzed. The quantitative determination of HbF and HbA2, red cell osmotic resistance and thalassemia were measured as well. Thalassemia heterozygote carriers and an increased level of HbF were revealed. The frequency of G-6-PD deficiency was 0.39%. In Bodrogköz the frequencies of AB0, Rh and haptoglobin types were similar in the present and all previous studies. The background of this similarity might be the genetic similarity between two following generations. On the basis of these facts, the Hb0 Arab and partially DNA work we suggested an alternative hypothesis that these mutant genes got into Bodrogköz by the rather later migration than with ancient Hungarian people during the period of conquest of Hungary.

Freeze-thaw effects on the detection of blood group substances in detergent extracts by enzyme-linked immunosorbent assays (ELISA).

The sensitivity of an enzyme-linked immunosorbent assay (ELISA) for human blood group antigens extracted from blood stains with the surfactant, n-octyl-beta-D-glucopyranoside (OBG), at concentrations below the critical micelle concentration can be increased by the introduction of a single freeze-thaw step. The ELISA signals increase from 3- to 4-fold for OBG extracts of 80 nl bloodstains. The ELISA signal enhancement occurs irrespective of the age of the bloodstains, at least for bloodstains up to 1 year old. The origin of the effect has been investigated and its is demonstrated that the freeze-thawing cycle increases the extent of adsorption of the blood group determinants in OBG-solubilized complexes onto microtitre plates. Gel filtration has been used to analyse the composition of OBG extracts of bloodstains in terms of the carriers of the blood group substances, protein and phospholipid in fresh and freeze-thawed extracts. It was found that freeze-thawing alters the distribution of blood group active material in the lipid-protein OBG complexes leading to a greater proportion of blood group active material in higher molecular weight complexes. The freeze-thaw effect is eliminated on the addition of a cryoprotectant, such as glycerol, and the factors which contribute to changes in the microstructure of OBG extracts on freeze-thawing are discussed.
