Synthesis of [O-methyl-11C]1-(2-chlorophenyl)-5-(4-methoxyphenyl)-4-methyl-1H-pyrazole-3-carboxylic acid piperidin-1-ylamide: a potential PET ligand for CB1 receptors.

Synthesis and in vitro evaluation of [O-methyl-(11)C]1-(2-chlorophenyl)-5-(4-methoxyphenyl)-4-methyl-1H-pyrazole-3-carboxylic acid piperidin-1-ylamide ([(11)C]-1), a potential imaging agent for CB(1) receptors using PET is described. 1-(2-Chlorophenyl)-5-(4-hydroxyphenyl)-4-methyl-1H-pyrazole-3-carboxylic acid piperidin-1-ylamide (5), the precursor for radiolabeling, was synthesized from 4-OTBDPS-propiophenone (2) in five steps with 30% overall yield. The reaction of alcohol 5 with [(11)C]MeOTf at 60 degrees C afforded [(11)C]-1 with an average radiochemical yield of 14.5% (EOS) and >2000 Ci/mmol specific activity. The radiotracer was found to selectively label CB(1) receptors in slide-mounted sections of postmortem human brain containing prefrontal cortex as demonstrated by in vitro autoradiography using phosphor imaging.

Applied jasmonates accumulate extracellularly in tomato, but intracellularly in barley.

Jasmonic acid (JA) and its derivatives are well-characterized signaling molecules in plant defense and development, but the site of their localization within plant tissue is entirely unknown. To address the question whether applied JA accumulates extracellularly or intracellularly, leaves of tomato and barley were fed with 14C-labeled JA and the label was localized in cryofixed and lyophilized leaf tissues by microautoradiography. In tomato the radioactivity was detectable within the apoplast, but no label was found within the mesophyll cells. By contrast, in barley leaf tissues, radioactivity was detected within the mesophyll cells suggesting a cellular uptake of exogenously applied JA. JA, applied to leaves of both plants as in the labeling experiments, led in all leaf cells to the expression of JA-inducible genes indicating that the perception is completed by JA signal transduction.

Laboratory analyses of 137Cs uptake by sunflower, reed and poplar.

The 137Cs uptake by three plant species (Phragmites australis L., Heliantus annus L., Populus simonii L.) was analyzed in a hydroponic medium (14 MBql(-1); 0.5 mM CsCl) during cultivation. The radioactivity disappearance from the medium was measured after 2, 4, 8, 16 and 32 days of cultivation. Radioactivity distribution within the plant was determined by autoradiography. We did not find differences between uptake of radioactive and stable caesium isotopes. Relations between the uptake of 137Cs and concentration of potassium and ammonium ions in medium were also tested. The highest uptake of radiocaesium by sunflower was obtained for medium with 1 mM K2SO4 (14.2%) and in case of ammonium ions for concentration ratio 6 mM NH4Cl : 3 mM NH4NO3 (13.2%). The obtained results make it possible to compare the capability and rate of 137Cs phytoremediation of different plant species.
