To evaluate separation efficiency and concentrate purity, each of 50 donors underwent platelet (PLT) collection on both the Cobe Spectra and Fresenius AS 104 with the constancy of processed blood volume, ACD/blood ratio 1:10 and a blood flow of 50 ml/min. For the Fresenius AS 104 two protocols - V 4.11 and V 4.4 - were evaluated. Both systems provided high quality PLT collection: PLT yield 3.91 +/- 0.74 x 10(11) (Cobe); 4.16 +/- 1.12 x 10(11) (V 4.11); 4.66 +/- 0.89 x 10(11) (V 4.4). The mean cell contamination was low with all procedures, resulting in less WBC contamination with the Fresenius AS 104, whereby the higher efficiency obtained by the protocol version V 4.4 correlated with an increased of WBC contamination.
The time has come when a traditional hematologist of a developing country needs to change his frame of mind from time consuming, error prone, not so precise manual methodologies to economical, safe and precise automated procedures. More important is his role as a guide and teacher for his juniors who are exposed to automated laboratories from the beginning of their residency. At this juncture, one needs to be thorough with the principles of instrumentation, different models available, their merits and demerits and how and where these fit with the requirements of the laboratory. The haematologists also must be aware that automation has its own problems, limitations, disadvantages and interfering elements. The article discusses the present state of art.
Acid elution methods have been extensively used for the identification of fetal red cells in pregnant women with fetomaternal hemorrhage. A number of methods have been described: Kleihauer-Betke, Boehringer Mannheim, Fetaldex, Nierhaus-Betke modification and Sanguansermsri amido black B. A comparative evaluation of these methods was performed in this study. The technique of Sanguansermsri using amido black B was found to be more advantageous than the others in terms of simplicity, accuracy, precision and time taken.
