Correlation between beta- and alpha-adrenergic receptor concentrations in human placenta.

alpha 2- and beta-adrenergic receptors in human placental membranes have been investigated using the radioligands [3H]-RX 821002 and [3H]-dihydroalprenolol, respectively. The specific binding of the alpha 2-adrenoceptor antagonist RX 821002 confirms the presence of an alpha 2-adrenoceptor in the human placenta, which has been characterized previously with [3H]-rauwolscine. The major finding presented here is a correlation between the alpha 2- and beta-adrenergic receptor concentrations (r = 0.765) in the human placenta at term. It is suggested that the alpha 2/beta adrenoceptor balance may play an important role in regulation of the vascular bed of the placenta. Determination of the alpha 2/beta ratio may help towards an understanding of the contractility of the placental vascular muscles.

[Biochemical requirements in tocolysis].

By a specific bond of 3H-dihydroalprenolol the number (Bmax) and characteristic (Kd) of sympathetic beta-receptors in cell membranes of the myometrium and lymphocytes of peripheral blood was assessed. The authors examined six tissue specimens taken during planned termination of pregnancy during the 38th-40th week by primary Caesarean section parallel with a peripheral blood sample. In healthy women even at the end of pregnancy the myometrium contains ample amounts of specific beta-receptors. As compared with the myometrium, the lymphocytes of the peripheral blood contain beta-adrenergic receptors of the same type as the myometrium but in much smaller amounts. So far it was not possible to show a correlation between the number of sympathetic beta-receptors in the membranes of the myometrium and of lymphocytes in the peripheral blood.

Glucocorticoids increase beta-adrenoceptors on human intact lymphocytes in vitro.

A modification of beta-adrenoceptor binding has been observed in different human and animal tissues after glucocorticoid administration. In this study we observed that hydrocortisone increased human intact lymphocyte beta-adrenoceptors labelled with 3H-dyhydroalprenolol in vitro, in physiological and pharmacological concentrations. In saturation experiments hydrocortisone (10(-6)M) significantly raised beta-adrenoceptor Bmax and KD. Intact living lymphocytes seem to be a useful model for further investigation in man of the regulation of beta-adrenoceptors by glucocorticoids in physiological conditions.
