[A computer-based microscope as the foundation for automated prescreening systems].

Abstract

The prototype of an automated screening system for cytological preparations is under development. The conditions concerning the handling of such system require the development of a special microscope with certain adjustment functions to be automated. The main specifications of this microscope are: --1 Splitting the light path into two synchronously usable channels for low and high magnification; whereby both images are projected onto two tv-cameras --Additional light path for visual observation --Automated focusing via movement of the microscope objective --Automated supply of immersion oil --Positioning and loading unit for slides --Microprocessor control --Closed and dust-protected design. Besides the mentioned specifications different microscopical observation methods, i. e. transmitted light bright field, incident fluorescence, are possible. The system can be used for all samples which are prepared on standard slides.

Automated detection of the factor V Leiden mutation using the LCx microparticle enzyme immunoassay.

The factor V Leiden mutation, a G-->A transition at position 1691 in exon 10 of the gene that codes for factor V, produces an Arg506Gln substitution and is the most common genetic risk factor for venous thrombosis. We have developed a rapid, sensitive, and specific method to detect the factor V Leiden mutation in genomic DNA from whole blood by PCR amplification and microparticle enzyme immunoassay detection using the Abbott LCx instrument. We compared this automated method with the standard procedure using restriction endonuclease digestion of PCR products followed by gel electrophoresis in blinded experiments. In 130 patients (from Veterans Affairs medical centers) with deep venous thromboses, including 24 heterozygotes with the factor V Leiden mutation, there was complete agreement between the two methods. The assay was also able to distinguish heterozygotes from homozygotes. This method, which carries a low potential for cross-contamination of samples, should be a useful routine test for the factor V Leiden mutation in clinical laboratories with sufficient demand for molecular diagnostic assays using the LCx instrument.

Automated flow cytometry compared with an automated dipstick reader for urinalysis.

Recently, the Sysmex UF-100 flow cytometer was developed to automate urinalysis. We compared UF-100 test results with those of an automated dipstick reader. A cross-check of UF-100, dipstick, and microscopic sediment data was performed in 1001 urine samples. Good agreements (P <0.001) were obtained between UF-100 and dipstick data for erythrocytes (r = 0.636) and leukocytes (r = 0.785). Even in urine with low conductivity, the UF-100 could detect lysed erythrocytes. The UF-100 bacterial count was higher among nitrite-positive urine samples (P <0.0001) and was positively correlated with the UF-100 leukocyte count (r = 0.745; P <0.001). In stored urine (24 h), bacterial counts increased, whereas the forward light scatter of leukocytes decreased (P <0.01). Casts and yeast cells reported by the UF-100 should be confirmed by microscopic review because false positives occurred. We suggest that a computer-assisted cross-check of UF-100 and dipstick data allows a clinically acceptable sieving system to reduce the workload of microscopic sediment urinalysis.

On-line coupling of microdialysis sampling with liquid chromatography for the determination of peptide and non-peptide leukotrienes.

An automated on-line sampling method was developed using microdialysis as the simultaneous sampling and sample pre-treatment technique. The extraction fraction values of microdialysis probes sampling different eicosanoids were investigated. The impact of cyclodextrins in the perfusion liquid used for sampling hydrophobic eicosanoids in biological systems was also studied. The total time for one analysis was 7.6 min allowing seven measurements per hour for monitoring kinetic changes in biological systems.
