Echographic imaging with dynamically focused insonification.

Abstract

This work describes a real-time echographic imaging device with lateral scanning and a new process for dynamic focusing. While, in reception, a time varying focus is a well-established process, the system that we propose is able to perform dynamic focusing also in transmission. This is carried out by emitting several acoustic pulses focused in different regions. This process, which we call dynamic focused insonification, gives good results and can be used either alone or in connection with dynamic focusing in reception.

New low-contrast resolution phantoms for computed tomography.

Computed tomography (CT) has been established as a major imaging modality in diagnostic radiology. Accordingly, acceptance testing and quality control of CT scanners is of great importance. While most procedures and phantoms for testing are widely accepted, there is still discussion and uncertainty about low-contrast (LC) sensitivity. In our opinion this unsatisfactory situation is caused at least in part by the lack of suitable phantoms for LC resolution measurements. We investigated the commonly used phantoms for LC detectability, the Catphan, and for LC resolution, the ATS phantom. While the Catphan showed stable object contrasts, the ATS phantom's measured contrast exhibited a strong dependence on temperature and x-ray quality. Based on newly developed polyurethane resin materials, we designed and tested a LC resolution phantom with several different contrast steps. The object contrasts showed no dependence on temperature and beam quality. The new LC resolution phantom proved to be very suitable for measuring a scanner's low-contrast sensitivity in the image plane, one of the most important image quality parameters. To assess LC resolution in three dimensions we designed an additional phantom with rows of spherical objects. A first prototype was evaluated in a multicenter study. The setup proved to be very helpful to quantify the in-plane and axial LC sensitivity of spiral CT scan modes.

Automated recognition of patterns characteristic of subcellular structures in fluorescence microscopy images.

Methods for numerical description and subsequent classification of cellular protein localization patterns are described. Images representing the localization patterns of 4 proteins and DNA were obtained using fluorescence microscopy and divided into distinct training and test sets. The images were processed to remove out-of-focus and background fluorescence and 2 sets of numeric features were generated: Zernike moments and Haralick texture features. These feature sets were used as inputs to either a classification tree or a neural network. Classifier performance (the average percent of each type of image correctly classified) on previously unseen images ranged from 63% for a classification tree using Zernike moments to 88% for a backpropagation neural network using a combination of features from the 2 feature sets. These results demonstrate the feasibility of applying pattern recognition methods to subcellular localization patterns, enabling sets of previously unseen images from a single class to be classified with an expected accuracy greater than 99%. This will provide not only a new automated way to describe proteins, based on localization rather than sequence, but also has potential application in the automation of microscope functions and in the field of gene discovery.

Learning about truth and biases through experience: section surface corrugation, protein denaturation, and staining.

By developing and investigating the use of thin resin sections for electron microscopy, we discovered that many of the current biases became invalid: 1) No evidence could be produced that the involved organic liquids produce the protein denaturation responsible for loss of antigenicity and lowered resolution. For immunolabeling, "water-soluble" resins are not per se to be preferred over less polar and more hydrophobic ones. 2) The relief formed by the corrugated section surface enables access to the antigenic sites. The depth of relief is determined by the strength of copolymerization and depends on temperature, the chemical nature of resin and biological matter, and, thus, also on surface modification through fixatives. The stronger the relief, the better the immunolabeling and the less the image resolution. Strong copolymerization favors flatter relief, but also a hiding of antigenic sites by thin layers of resin. 3) Heavy metal stain remains the main culprit for low resolution of sections when compared to results obtained with other preparation methods.
