Oncorna-like viruses in baboon prostate tissue.

Abstract

Oncorna-like virus particles were observed in prostate tissue of 2 of 11 baboons inoculated 3 years previously with a chemical carcinogen. There were no indications, however, of any relationship between the carcinogen and the development of virus particles. Biopsy specimens from the animals showed no evidence of neoplasia, and electron microscopic examination suggested that this virus should be categorized as a B-type oncornavirus.

Isolation of two regions on rat chromosomes 5 and 18 affecting mammary cancer susceptibility.

We previously mapped several quantitative trait loci (QTLs) controlling DMBA-induced mammary tumor development in female rats derived from a SPRD-Cu3 (susceptible strain) x WKY (resistant strain) cross. Two of these QTLs were assigned to chromosomes 5 and 18. In the present study, we generated and characterized congenic strains in which a segment of WKY chromosomes 5 or 18 was introduced in the SPRD-Cu3 genetic background, thereby physically demonstrating that each of these two chromosomes controls mammary tumor multiplicity. The chromosome 5 QTL (Mcstm1) accounts for 7 tumors per animal (versus a total of 11 tumors per SPRD-Cu3 rat). The chromosome 18 QTL (Mcstm2) accounts for 3 tumors per animal and is the first chemically-induced mammary cancer susceptibility locus assigned to this chromosome. In addition, the Mcstm1 region was shown to also controls tumor latency. These loci thus play a major role in chemically-induced mammary tumor development. QTLs controlling chemically-induced or estrogen-induced mammary tumor development have independently been identified on chromosomes 5 and 18, using susceptible strains others than SPRD-Cu3. Therefore the haplotype structure of the relevant chromosome regions was analyzed in the different strains. Some chromosome regions were found to be highly mosaic (haplotype blocks < 1 Mb), while one region showed an apparently conserved haplotype block of 7.5 Mb. This analysis points to limited regions that could harbor the causative genes and also indicates that at least Mcstm2 is a novel QTL.

involvement of microsomal epoxide hydrolase enzyme in ovotoxicity caused by 7,12-dimethylbenz[a]anthracene.

Ovarian follicle disruption in mice caused by 7,12-dimethylbenz[a]anthracene (DMBA) is attributed to its bioactivation by CYP1B1 to a 3,4-epoxide which is then hydrolyzed to form a 3,4-diol by microsomal epoxide hydrolase (mEH). Further epoxidation by CYP1A1 or 1B1 forms the ultimate ovotoxicant, DMBA-3,4-diol-1,2-epoxide. Studies suggest that the mouse ovary expresses these enzymes, and thus, may be capable of bioactivating DMBA to its ovotoxic metabolite. The present study was designed to evaluate the role of ovarian mEH in DMBA-induced ovotoxicity using a novel neonatal mouse ovarian culture system. Ovaries from postnatal day (PND) 4 B6C3F(1) mice were incubated with DMBA (12.5 nM-1 microM) for various lengths of time. Following incubation, ovaries were histologically evaluated or assessed for mEH protein or mRNA. Following 15 days of incubation, DMBA reduced (p < 0.05) healthy follicles at concentrations >or= 12.5 nM. At 1 microM DMBA, follicle loss and increased mEH protein were measured (p < 0.05) by 6 h. mRNA encoding mEH markedly increased after 2 days of incubation, and this increase preceded accelerated follicle loss at 4 days. Furthermore, follicle loss induced by DMBA was prevented when cyclohexene oxide (2mM), an mEH inhibitor, was added to DMBA incubations. These studies suggest that the PND4 mouse ovary is capable of bioactivating DMBA to its ovotoxic form, and that ovarian mEH enzyme activity is likely involved. Furthermore, these observations support the use of a novel ovarian culture system to study ovary-specific metabolism of xenobiotic chemicals.

Suppression of cell proliferation, induction of apoptosis and cell cycle arrest: chemopreventive activity of vanadium in vivo and in vitro.

In the present study, the authors evaluated the anticancer mechanism of vanadium, a dietary micronutrient and an important pharmacological agent, on a defined model of chemically induced rat mammary carcinogenesis in vivo and on human breast cancer cell line MCF7 in vitro. Female Sprague-Dawley rats were treated with 7,12-dimethylbenz(alpha)anthracene (0.5 mg/100 g body weight) by a single tail vein injection in an oil emulsion to induce mammary preneoplasia. Vanadium (ammonium monovanadate) at a concentration of 0.5 ppm (4.27 micromol/l) was supplemented in drinking water and given ad libitum to the experimental groups for 24 weeks. Histological finding showed substantial repair of hyperplastic lesions. There was a significant reduction in incidence, multiplicity (34%, p < 0.01), size of palpable mammary tumors and delay in mean latency period of tumor appearance. Immunohistochemical analysis in vivo indicated a decrease in cell proliferation (24.68% p < 0.05) and an increase among the TUNEL-positive apoptotic cells along with strong expressions of p53 and Bax, and downregulation of Bcl2 proteins in the mammary tissue of vanadium-treated animals. Further, MCF7 cells were cultured in minimal essential medium and were treated with 100, 175 and 250 microM of vanadium (ammonium monovanadate) for 36 hr. Exposure of MCF7 cells to vanadium led to induction of apoptosis in a dose-dependent manner. It was found further that vanadium treatment brought about a prominent cell cycle arrest and chromosomal condensation, leading to apoptosis (42.62%, p < 0.05). Results of both the in vivo and in vitro study demonstrate that vanadium has the potential to be developed into an anti-breast cancer drug in the near future.
