Abstract

In this work we collect the results of the variation of the variable "rechange of the wind with the exterior" in the three possibilities of a bedroom: --close window, semi close and totally open. And we unite the relation that we already know between the prevail of acariens (Dermatophagoides) and the relative humidity of the wind in a Bilbao city's house.

Interfering with DNA repair pathways to enhance CRISPR-Cas9-mediated homology-directed repair in a chelicerate genetic model.

The two-spotted spider mite, Tetranychus urticae, is a major pest and an emerging genetic model. Recent CRISPR-Cas9 advances, especially the SYNCAS method for maternal delivery of Cas9 ribonucleoproteins, have enabled precise genome editing in this and other difficult-to-transform arthropods. Yet SYNCAS-mediated knockins vary in efficiency, possibly due to competition between DNA repair pathways, whose mechanisms in T. urticae and other chelicerates remain unknown. Here, we provide the first functional analysis of double-strand break repair in a chelicerate. Loss of DNA polymerase theta (Polθ) redirects repair almost entirely toward homology-directed repair, whereas absence of Ligase IV has no detectable impact. Using a reporter assay targeting phytoene desaturase, we demonstrate that Polθ-deficient strains enhance incorporation of repair templates, even when mutations are distant from the cut site. Also, insertion of larger fragments is improved. Finally, disrupting Polθ imposes only a modest fitness cost, highlighting its value for future genome engineering in this species.

Phylogeny of parasitiform mites (Acari) based on rRNA.

Acari (mites and ticks) form one the most diverse lineages of arthropods, but basal relationships in the group are still poorly understood. The current study addresses this issue for one of its two main lineages, the order Parasitiformes. Relationships are examined at the subordinal and infraordinal level using complete 18S and partial 28S nuclear rRNA sequence data. Most currently recognized lineages are recovered with good support, suggesting that nuclear rRNA, and specifically 18S rRNA, is very well suited for analyzing relationships at this level in this lineage. These results were found despite quite variable rates of sequence evolution, with rates "ratcheting up" from relatively low in most non-mite arachnid lineages, to intermediate in Pseudoscorpiones, the mite order Acariformes, and the parasitiform suborders Opilioacarida, Holothyrida, and Ixodida, to high in the parasitiform suborder Mesostigmata. The most species rich mesostigmatid infraorder, Dermanyssina, shows huge distances to the outgroups, but remarkably low within-group divergence in nuclear rRNA. This suggests the possibility of a relatively recent origin of this lineage.

Microhabitat selection in the simple oribatid community dwelling in epilithic moss cover (Acari: Oribatida).

The moss cover of a roof was studied as the model of a simple habitat divided into microhabitats by the members of a community of saprophagous mites. This community consisted of two species of oribatid mites: Scutovertex minutus and Trichoribates trimaculatus. They were extracted from moss onto moist paper, and subsequently, their mobility, responses to moisture fluctuation, and food selection were tested in laboratory experiments. For the nutritional biology, the microanatomy of their alimentary tract was examined according to the system of histological characteristics formulated in the laboratory of the author (type of food, digestive activity of gut walls, etc.). The paraplast sections of the mites were stained by Masson triple stain for these purposes. Moreover, the enzymological tests (chitinase and cellulase activities) were performed to detail the digestive processes. Such an approach was applied to field-sampled specimens as well as to those in the laboratory experiments. These above-mentioned data were discussed with respect to microhabitat selection, vertical and horizontal distribution, and dispersal ability of these two oribatid mites sharing this habitat. These two species differ in several characteristics from each other and these differences resulted in their different microhabitat choices and role in the habitat as a whole.
