A logical stepwise approach to laser diffraction particle size distribution analysis methods development and validation.

Abstract

In this study, a logical, stepwise, efficient approach was used to develop and validate particle size distribution analysis methods for 58 different pharmaceutical bulk powders in a timely fashion. Image analysis was used to determine particle morphology and laser diffraction particle size distribution analysis was used to evaluate the dispersion medium, dispersion concentration, sonication time, and dispersion stability. Ruggedness validation was performed, by two different analysis, on different days, with different instruments on two preparations each of two different lots of material. It was determined that if the relative standard deviation (RSD) of the median volume diameters (d50) of the four preparations for each lot was below 20%, the method was suitably rugged for use in a quality control setting. Data for methyldopa, metoprolol tartrate, and metronidazole are presented as typical method validation results for three different modes of analysis. Data at three points (d10, d50, and d90) on the distributions were tabulated and evaluated for all 58 methods validated. The median volume diameter (d50) was found to be adequate for method validation. The approach rapidly generated valid, reproducible particle size distribution analysis methodology.

Fluorouracil prodrugs for the treatment of proliferative vitreoretinopathy: formulation in silicone oil and in vitro release of fluorouracil.

Three new N(1)-alkylcarbonyl-5-fluorouracil derivatives that are prodrugs of 5-fluorouracil (FU), one of them being a co-drug FU-retinoic acid (RA), were studied as potentially effective drugs against postsurgical proliferative vitreoretinopathy (PVR). The stability of N(1)-octenoylFU (3), N(1)-lauroylFU (2), and N(1)-retinoylFU (4) in aqueous medium, their solubility in silicone oil (SiO), the kinetics of FU release in an in vitro system were determined. Compound 3 is very rapidly soluble in SiO. Its saturation concentration, reached after 6h, is 233 +/- 13 microg g(-1) SiO. Compound 2 is not very soluble in SiO but its kinetic of solubilization is fast. Its saturation concentration, reached after 2 days, is 27 +/- 2 microg g(-1) SiO. Compound 4 is poorly soluble in SiO. A concentration plateau, with a mean value of 4 microg g(-1) SiO, is reached after 4 days. The addition in SiO of 5% of a perfluorinated perhydrogenated alkene greatly improves the solubilization of compound 4. Two different types of FU release are observed. For compound 3, the release is fast and is achieved after 1 day. For compounds 2 and 4, the release is slower and is ended at 10 and 27 days, respectively. The solubility of the prodrugs in SiO is not correlated with their lipophilicity, whereas the release rate of FU decreased with increased lipophilicity of the prodrug. The most promising prodrug is compound 4 that slowly releases two active drugs (FU and RA) with a t (1/2 release) of 5.8 days. It might be interesting for the treatment of PVR. However, an in vivo study on an animal model of PVR is necessary to prove the efficacy of this formulation and to study its toxicity.

Chemistry challenges in lead optimization: silicon isosteres in drug discovery.

During the lead optimization phase of drug discovery projects, the factors contributing to subsequent failure might include poor portfolio decision-making and a sub-optimal intellectual property (IP) position. The pharmaceutical industry has an ongoing need for new, safe medicines with a genuine biomedical benefit, a clean IP position and commercial viability. Inherent drug-like properties and chemical tractability are also essential for the smooth development of such agents. The introduction of bioisosteres, to improve the properties of a molecule and obtain new classes of compounds without prior art in the patent literature, is a key strategy used by medicinal chemists during the lead optimization process. Sila-substitution (C/Si exchange) of existing drugs is an approach to search for new drug-like candidates that have beneficial biological properties and a clear IP position. Some of the fundamental differences between carbon and silicon can lead to marked alterations in the physicochemical and biological properties of the silicon-containing analogues and the resulting benefits can be exploited in the drug design process.

Correlation of characteristics of gel extrusion module (GEM) tablet formulation and drug dissolution rate.

The purpose of the study was to characterize the swelling properties of controlled release tablet formulations with compositional and processing differences. A correlation was also established between the drug dissolution rate from the controlled release gel extrusion module (GEM) tablet and the swelling properties of the core tablet. The GEM tablet consisted of a core tablet of water swellable Carbopol polymer, a neutralizing agent, drug, and excipients. The tablet was subsequently coated with a rigid, water impermeable membrane. A number of holes were then drilled by laser through the impermeable membrane. Dynamic mechanical analysis (DMA), Perkin Elmer DMA7, was used to characterize the swelling properties. During the swelling measurements, the measuring probe and sample were completely submerged in the surrounding medium. The results showed that the formulation containing potassium phosphate dibasic as a neutralizing agent had the highest swelling rate. Correspondingly, this formulation had the highest drug dissolution rate over the same time period. The processing difference included wet granulation and dry roller compaction. The compositional differences included different neutralizing agents, binders or Carbopol polymers. The results showed a linear relationship (r(2)=0.954) between the swelling rates of the core tablets and the drug dissolution rates of GEM tablets. No correlation was found between the drug dissolution rates and either the maximum extent of tablet swelling or the time needed to reach the maximum extent of swelling. The results demonstrated that DMA could be used to support both formulation and process development to determine the effects of different compositions and manufacturing processes on drug dissolution rates for swelling controlled release devices.
