Suppression of IgE antibody formation in mice: requirement for T-T lymphocyte interaction.

Norman SWR mice injected with syngeneic spleen cells from ovalbumin (Ov)-primed mice were unable to make IgE anti-Ov antibodies when challenged with alum-pertussis-Ov. Immune T lymphocytes were shown to be responsible for the inhibitory effects of adoptively transferred spleen cells. Treatment of recipient mice with mild x-irradiation or with cyclophosphamide 2 or 3 days before cell transfer resulted in abrogation of the suppressor effect of immune cells. The injection of T lymphocytes into x-irradiated milce restored the suppressive effect of immune cells. It thus appears that immune T cells provide the stimulus for activation of suppressor T cells of the host. Although the generation of suppression is antigen-specific, the expression of suppression appears to be nonspecific.

Delayed type hypersensitivity to bovine serum albumin and to lipid-conjugated bovine serum albumin in mice.

Delayed type hypersensitivity (DTH) to bovine serum albumin (BSA) and to lipid-conjugated BSA were studied comparatively. Unlike the case of BSA with which no DTH can be detected with native antigen, injection of butyric-conjugated BSA (Bu-BSA) in sensitized mice provokes a typical DTH for an early and limited period. Alum-precipitated Bu-BSA (Al-Bu-BSA) provokes from the beginning a stronger DTH which persists a much longer period.

Preferential synthesis of IgE reaginic antibodies in rats immunized with alum-adsorbed antigens.

The reaginic antibody response to alum-precipitated ovalbumin (OA) and the dialyzed water-soluble extracts of ragweed (DWSR) and Alternaria tenuis (DWST) in several strains of rats appeared to be wholly an IgE response. There was no evidence of a heat-stable (IgGa) antibody to OA, DWSR and DWST in the sera of the rats immunized with these antigens suspended in alum. Wistar-Furth and Lew inbred and hooded outbred rats produced comparable amounts of reaginic antibody after immunization with DWST, but BN inbred rats failed to generate a reaginic response to this antigen. The amount of antigen-induced histamine release from rat peritoneal mast cells did not always correlate with the level of circulating IgE-specific antibody.

The quantitative histochemistry of ribonucleic acid using gallocyanin.

A method for the cytophotometric estimation of ribonucleic acid in tissue sections using gallocyanin-chrome alum is described. The dye obeys Beer's law in gelatin sections. The effect of deoxyribonuclease on the staining of ribonucleic acid is also investigated. The results indicate that this method is of value in the quantitation of ribonucleic acid.

Physico-chemical investigation of AlPO4 for the manufacture of antacids. I. Effect of precipitation and processing conditions on physico-chemical properties of AlPO4.

Aluminium phosphate was precipitated from AlCl3-H3PO4 and NaOH, NH3H2O and NaHCO3--Na2CO3 mixture up to different pH values. The precipitates were dried at room temperature, 40 degrees and 105 degrees and their neutralizing properties (rate of neutralization and acid-consuming capacity) against 0.l N HCl were investigated. It was found that these properties depend on the nature of the reagents used for the formation of AlPO4, the pH and also the temperature of drying. The best antacid properties shows the AlPO4 preperation obtained from AlCl3-H3PO4 and NaHCO3-Na2CO3. For comparison, the therapeutic preparate "Aluphos" was also investigated.

Prolonged immunotherapy with alum-precipitated pyridine ragweed extracts-a clinical and immunological study.

In 18 ragweed hay fever patients who received injections of alum-precipitated pyridine (Allpyral) ragweed extracts in maintenance doses of between 6000 and 8000 P.N.U. at intervals of between two and four months over a two-year period, the prolonged interval did not seem to influence either the clinical response or the tendency to reaction, or result in significant change in immunological tolerance as determined by antibody studies, which included fluorescent antibody assays, as well as hemagglutination and RAST technics.

[Acetlysalicylic acid, protective antacid effect and lesions of the gastric mucosa. Effect of acetylsalicylic acid and an antacid drug (Gastropulgit Tabs) on the transmural electric potential in the human stomach].

Antacids are able to prevent acetylsalicylic acid-induced functional and morphological changes of the gastric mucosa. In order to test whether a new antacid in chewing-tablet form (Gastropulgit Tabs) might be able to protect the gastric mucosa from acetylsalicylic acid-induced functional changes similarly to liquid antacids the effect of this antacid on acetylsalicylic acid-induced changes of transmural gastric potential difference was measured in healthy volunteers. The decrease of transmural potential differences induced by 640 mg acetylsalicylic acid could be prevented by simultaneous addition of 2 tablets of Gastropulgit Tabs. Thus the antacid in tablet form is able-like liquid antacids-to protect the gastric mucosa against acetylsalicylic acid-induced functional changes.

Absorbances of hematoxylin solutions at varying pH levels.

An alum hematoxylin was prepared (0.5 g hematoxylin, 8 g potassium alum, distilled water 477.5 ml and Na10(3) solution, 1 mg/ml, 22.5 ml) and divided into 50 ml aliquots. The pH of these aliquots was adjusted with 2 1/2% NaOH or 10% H2SO4. As soon as the pH of a sample was adjusted the absorbance from 400 to 700 nm was recorded with a Cary 15 spectrophotometer using specially designed cuvettes and undiluted samples. The following day the absorbances at peak were measured on a Coleman Jr. spectrophotometer and three days later a variety of tissues was stained with each solution. The pH and peak of absorbance of the aliquots were pH = 2.0 450 NM, 2.5 505, 2.6 507, 2.7 515, 2.8 520, 2.9 530, 3.0 540, 3.1 550, 3.3 560, 3.5 560. In the stained material in the intensity of nuclear staining was about the same at all pH levels but non-specific staining was greatest in slides stained at pH = .5 and virtually absent in slides stained at pH = 2.0. At intermediate pH levels the amount of non-specific staining was intermediate.

Immunosuppression in murine malaria. II. The primary response to bovine serum albumin.

The primary antibody response to alumadsorbed bovine serum albumin was depressed in CBA mice infected with Plasmodium yoelii yoelii. Responses initiated within approximately 3 weeks of this malaria infection were reduced in quantity, but not in avidity. Responses initated later were normal. A depressed primary response was also seen in infected compared to control splenectomized mice; this was accompanied by impaired priming for a secondary response.
