Aromatic compound glucosides, alkyl glucoside and glucide from the fruit of anise.

From the polar portion of the methanolic extract of the fruit of anise (Pimpinella anisum L.), which has been used as a spice and medicine since antiquity, four aromatic compound glucosides, an alkyl glucoside and a glucide were isolated together with 24 known compounds. The structures of the new compounds were clarified as (E)-3-hydroxyanethole beta-D-glucopyranoside, (E)-1'-(2-hydroxy-5-methoxyphenyl)propane beta-D-glucopyranoside, 3-hydroxyestragole beta-D-glucopyranoside, methyl syringate 4-O-beta-D-glucopyranoside, hexane-1,5-diol 1-O-beta-D-glucopyranoside and 1-deoxy-L-erythritol 3-O-beta-D-glucopyranoside by spectral investigation.

Antibody response of Echinococcus granulosus infected mice: recognition of glucidic and peptidic epitopes and lack of avidity maturation.

The antibody response was followed weekly during 68 weeks in 17 Balb/c mice intraperitoneally (i.p.) infected with 2000 Echinococcus granulosus protoscoleces (PSC) and in three mice i.p. immunized with 2000 dead PSC. Antibodies against hydatid cyst fluid (HCFA) and its peptidic (periodate-resistant) and carbohydrate (periodate-sensitive) epitopes were titrated by ELISA. Avidity and the antigen recognition pattern of antibodies were also analysed during infection and immunization by ELISA and immunoblot, respectively. The antibody response of infected mice showed quantitative and qualitative variations during infection, since both titre as well as recognition of peptide and carbohydrate epitopes in HCFA depended on time post infection. No avidity maturation was evident during the course of infection. Sera from infected mice recognized the 38 kDa subunit of Ag5 but did not react with the 8 kDa subunit of AgB. On the contrary, the antibodies response of immunized mice showed only one peak of antibodies that recognized both peptidic and carbohydrate epitopes of HCFA. In addition, sera from these mice recognized mainly 60 and 110 kDa bands. Our results suggest that: a) avidity and antigen recognition patterns of antibodies in mice treated with live PSC are different from those treated with dead PSC; b) antibodies against HCFA glucidic or peptidic epitopes appear at different times post infection.

Rats treated with oleoyl-oestrone maintain glucidic homeostasis: comparisons with a pair-fed model.

To determine whether or not the weight (and fat) loss induced by oleoyl-oestrone treatment results only as a consequence of decreased food intake, we compared treated animals with a pair-fed model. To this end, Wistar female rats received daily oral gavages of 10 mumol/kg per d oleoyl-oestrone in sunflower oil, or vehicle alone for 10 or 20 d. A second group of rats received the gavage of sunflower oil and the same amount of food ingested as the oleoyl-oestrone-treated animals (pair-fed group). Rats treated with oleoyl-oestrone maintained glucidic metabolism homeostasis despite a marked decrease in adipose tissue weight (P<0.001). Pair-fed rats exhibited a different pattern, comparable to short-term starvation, with greatly decreased glycogen stores (P<0.0001). The most significant effects were detected in the 10 d period groups. Oleoyl-oestrone affected the activity of the ponderostat system not only by decreasing appetite but also by modifying energy partition: treated animals maintained their glucose and energy homeostasis despite decreased food intake and the massive depletion of lipid stores.

[Effect of prolonged glucidic starvation on the contents of starch and tannins of Blechnum brasiliense L. gametophytes].

Some Blechnum brasiliense gametophytes have been maintained in darkness, in a purely mineral medium, for the purpose of investigating the effect of prolonged glucidic starving on the amount of tannins and starch that they contain. During the first two months the tannin contents undergoes a fast decrease. The starch variations are different : a transitory increase simultaneous with the fall in tannin; a decrease after the disappearance of 3/4 of the tannin contents; and lastly, a new amylogenese which precedes the prothallus necrosis. It seems that the gametophytes, in order to survive, degrade first their tannoidic reserves, and they utilize the starch that they contain only with difficulty. This behaviour can be explained by the distribution of the two metabolites; by the acid pH of the medium, which is not very propitious to the phosphorylases activity and lastly by the presence of chlorogenic acids, which are inhibitors of those enzymes.

Effects of 2200-m hypoxia on glucidic metabolism.

Rats exposed for different intervals (1, 3 and 6 hours, the first day, and then 6 hours a day for 9 days) to a 2200-m hypoxia presented metabolic changes demonstrating an anaerobic glucose utilization. Thus, in accordance with other authors, at different depressurization values the liver homogenate had increased lactate and pyruvate concentrations, decreased NAD and NADH2 concentrations and a decreased ATP formation, demonstrating the inefficient glucide metabolization. These modifications were evident in the first hours of depressurization, but did not occur under acclimatization during the 9 days of exposure. The LDH concentration remained practically unchanged, which might be explained by its explained by its excessive presence in relation to the normal exercise solicitation. A urinary hypercompensatory alkalinization was also recorded.

Glucidic and lipidic metabolic changes in rats induced by irradiation and the effect of adrenalectomy.

In experiments on X-irradiated rats (1000 R) the hepatic glycogen, total lipids, phospholipids content, and plasma glucose, cholesterol and beta-lipoprotein concentration were determined in intact and adrenalectomized animals. It was confirmed that irradiation produces a hepatic glycogen and blood glucose increased concentration. The glucidic metabolic response on irradiation is diminished by adrenalectomy. The adrenalectomy-induced modifications in the lipid metabolism of irradiated rats are more inconstant, which corresponds with its relative independence from glucocorticoid hormones.
