Two phenanthraquinones from Dendrobium moniliforme.

Two phenanthraquinones were isolated from the stems of Dendrobium moniliforme. They were identified by interpretation of spectral data as 2,6-dimethoxy-1,4,5,8-phenanthradiquinone and 5-hydroxy-3,7-dimethoxy-1,4-phenanthraquinone, named moniliformin (1) and denbinobin (2), respectively. This is the first report of compound 1, which possesses a novel 1,4,5,8-diquinone moiety in the phenanthraquinone skeleton. Compound 2 showed potent antiinflammatory effects in vitro.

New antiviral cassane furanoditerpenes from Caesalpinia minax.

A bioassay-guided study led to the isolation of five new cassane furanoditerpenes, designated as caesalmin C (1), D (2), E (3), F (4), and G (5), along with stigmasterol (6) from the seeds of Caesalpinia minax. The (1)H and (13)C NMR spectra were completely assigned by using a combination of 2D NMR analyses. The structures of all five furanoditerpenes were confirmed by X-ray analyses. The structure of 6 was verified by X-ray analysis for the first time. The bioassay results showed that the anti-Para3 virus activity of tetracyclic furanoditerpenoids 1-4 is more potent than that of the furanoditerpenoid lactone 5, which is in turn better than 6. As the major components of the plant possess significant potent activity, it may be feasible to develop new antiviral agents from this source.

Antimalarial (+)-trans-hexahydrodibenzopyran derivatives from Machaerium multiflorum.

Bioassay-guided fractionation of Machaerium multiflorum yielded the hitherto unreported (+)-trans-hexahydrodibenzopyrans machaeriol A (1) and machaeriol B (2), as well as the known guaiane sesquiterpene (-)-kessane. Structure elucidation was based on (1)H and (13)C NMR data, mainly 2D NMR (1)H-(1)H COSY, (1)H-(13)C HMQC, (1)H-(13)C HMBC, and (1)H-(1)H NOESY experiments. This is the first report of the hexahydrodibenzopyrans from a higher plant other than the genus Cannabis. The cannabimimetic activity was thus evaluated by radioligand binding assay for cannabinoid receptor CB1, which indicated, notably, that both 1 and 2 were inactive. In addition, the cross reactivity of 1 and 2 toward antibodies designed for urinary metabolites of cannabinoids was evaluated with the EMIT and On Line cannabinoids assays. Both compounds showed no response at 100 000 ng/mL in both assays. Machaeriol B (2) demonstrated in vitro antimalarial activity (IC(50) = 120 ng/mL) against Plasmodium falciparum W-2 clone.

The structures of compounds 1-11 were established on the basis of extensive 1D and 2D NMR spectroscopic data interpretation. All compounds exhibited cytotoxicity activity against the A2780 ovarian cancer cell line, but none of the six compounds selected for testing in multiple cell lines showed significant selectivity.
