Temporary salvage of ischemic myocardium by the protease inhibitor bis[ethyl(2R,3R)-3-[(S)-methyl-1-[4-(2,3,4-trimethoxyphenyl-methyl) piperazin-1-ylcarbonyl]butyl-carbonyl]oxiran-2-carbo xylate]sulfate.

The administration (i.v., 20, 40 and 60 mg/kg) of the thiol protease inhibitor bis[ethyl(2R, 3R)-3-[(S)-methyl-1-[4-(2,3,4-trimethoxyphenyl-methyl) piperazin-1-ylcarbonyl] butyl-carbonyl] oxiran-2-carboxylate]sulfate (NCO-700), before and/or after the coronary ligation significantly reduced the necrotic mass in the rabbit left ventricle and also prevented the creatine phosphokinase loss in the ischemic myocardium up to 3 h and not at 6 h after the ligation. The activities of both calcium-activated neutral protease and cathepsin B in the subendo- and subepicardial layers of the ischemic, marginal or control myocardium of dog were inhibited by the NCO-700 administration (20 mg/kg i.v.) after the coronary ligation. A hemodynamic study using heart-lung preparation of the dog demonstrated a dose-dependent coronary dilatation with weak and transient negative ino- and chronotropic effects. These data suggested that NCO-700 sensitive protease(s) is(are) included in the step of myocardial cell degradation and that NCO-700 temporarily salvages it, as is important for the recent PTCR (percutaneous transluminal coronary reperfusion) therapy.

Study of the antiaggregating activity of new phenacyl-piperazine derivatives.

The antiplatelet activity of twelve new derivatives of phenacyl-piperazine was studied. Eight of them proved to be inactive by Born's method. The other four compound partially inhibited the aggregating effects induced by adrenaline and ADP, but did not prevent the effects produced by collagen. Although it was not possible to establish any relationship between chemical structure and antiplatelet activity, it looks as if there is a certain relationship between the analgesic and the antiplatelet effect. The compounds tested did not usually modify bleeding time.

Effects of 1-[2-ethoxy-2-(3'-pyridyl)ethyl]-4-(2'-methoxyphenyl)piperazine (IP-66), prazosin, yohimbine and dihydroergotoxine mesylate on the vasoconstriction of rat mesenteric artery induced by electric stimulation and exogenous norepinephrine.

The effect of (1-[2-ethoxy-2-(3'-pyridyl)ethyl]-4-(2'-methoxy-phenyl)piperazine (IP-66) on the contraction of rat mesenteric artery induced by electric stimulation (ESV) or by injection of norepinephrine (NESV) was compared with the effects of prazosin, yohimbine and dihydroergotoxine mesylate (DHEM). Interference of propranolol on the effects of the 4 alpha-blockers was also considered. Prazosin has proved to be the strongest antagonist of alpha 1-receptors towards which, however, it showed a certain bond latency. In particular conditions related to increased perfusion time and concentrations prazosin also seemed to lose selectivity towards alpha 1-receptors. It was observed that yohimbine is a weak postsynaptic antagonist whereas DHEM showed scarce selectivity. IP-66 proved to be a strong postsynaptic antagonist with negligible presynaptic effect. It was not as strong as prazosin but its bond with alpha 1-receptors appeared more rapid and steady. Results obtained with prazosin or DHEM and propranolol seemed to suggest an equilibrium between beta 2- and alpha 1-receptors shifted towards the latter.

An investigation into the roles of synaptic vesicular Mg++-ATPase in neurotransmitter release, using benzhydryl piperazines.

A series of benzhydryl piperazines was found to inhibit synaptic vesicular Mg++-ATPase. These compounds were also found to increase basal and evoked release of noradrenaline from synaptosomes. A comparison was made between the concentrations effective in inhibiting the enzyme and promoting noradrenaline release. In general, as the degree of Mg++-ATPase inhibition increased, noradrenaline release was increased. The relevance of these findings to a possible role of Mg++-ATPase in noradrenaline release is discussed.

In vitro comparison of pre- and postsynaptic effects of dihydroergotoxine mesylate and 1-[2-ethoxy-2-(3'-pyridyl)ethyl]-4-(2'-methoxy-phenyl) piperazine (IP-66).

The effects caused on the intestine mesenteric artery preparation by 1-[2-ethoxy-2-(3'-pyridyl)ethyl]-4-(2'-methoxy-phenyl) piperazine (IP-66), a compound having a prevailing alpha-blocking action, and by dihydroergotoxine mesylate (DHEM) were compared in normal conditions and in propranolol-induced beta-receptors block. The two drugs behaved in a completely different way both on vasoconstrictor response (delta p) and on noradrenaline (norepinephrine, NA) outflow induced by electrical stimulation of periarterial nerves. In fact IP-66 inhibited delta p much more than DHEM did but, unlike the latter, it increased NA outflow only at the lowest concentration (8.7 X 10(-9) mol/l) whereas it reduced it at the highest one (2.9 X 10(-7) mol/l). Moreover the effect of IP-6--but not that of DHEM--on NA outflow was completely abolished by propranolol. The hypothesis is put forward that IP-66 may show a strong antagonism towards postsynaptic alpha-receptors and a slight agonism towards presynaptic beta-receptors.

[Comparison of the antilipemic effect of bezafibrate with that of piperazine-sultosilate (A 585). A crossed double-blind study].

In 20 patients suffering from primary hyperlipoproteinaemia the action of sultosilic acid piperazine salt (A-585) was compared with bezafribrate. In a double-blind cross-over study parameters of the lipoprotein metabolism, as well as of fibrinolysis and of platelet function were examined. Both drugs significantly diminished total cholesterol, triglycerides, beta- and pre-beta cholesterol whilst alpha-cholesterol increased. Moreover, both drugs caused a significant shortening of the euglobulin lysis time and a diminution of platelet adhesiveness. In patients under oral anticoagulants the thrombotest levels were not influenced by A-585, but were depressed by bezafibrate. A slightly elevated gamma-GT normalized during bezafibrate therapy but was not influenced by A-585.

Comparative effects of p-chloroamphetamine and 1-(p-chlorophenyl)piperazine on 5-hydroxyindole concentration in rat brain.

1-(p-Chlorophenyl)piperazine (PCPP), a structural analog of p-chloroamphetamine (PCA), increased serotonin and decreased 5-hydroxyindoleacetic acid (5-HIAA) concentration in rat brain at 6 hours. At one day and one week, serotonin and 5-HIAA concentrations were normal. In contrast, PCA lowered serotonin and 5-HIAA at all three times. These different effects on 5-hydroxyindole concentrations in brain were not due to differences in brain levels of the two compounds, since these were approximately equal. PCPP lacks the ability of PCA to deplete brain 5-hydroxyindoles.

Controlled comparative study of the efficacy of pyrantel pamoate and a combined regimen of piperazine citrate and bephenium hydroxynaphthoate in the treatment of intestinal nemathelminthiases.

Pyrantel pamoate at a dose of 10 mg/kg body weight daily for 2 or 3 days was found to be more effective against Ascaris than both a regimen of bephenium hydroxynaphthoate (2.5 g base) and piperazine citrate (2-3 g) given daily for 2 days and a placebo-treated control group. The cure rates for pyrantel were about 95% compared to 90 and 20% for the other two groups respectively. None of the drug schedules was effective against the Trichuris. The bephenium/piperazine regimen was superior to both the 2-day and 3-day courses of pyrantel pamoate against the hookworm with aggregate cure rates of 58-60% for pyrantel pamoate and 68-85% for bephenium/piperazine. The results were better than those reported earlier in the series when a single dose of pyrantel pamoate (10 mg/kg of body weight) was used. Pyrantel pamoate was well tolerated and the drug merits further studies at higher doses, particularly against the hookworm.

A serotonergic component in the central action of 1-(o-methoxyphenyl)-piperazine.

1-(o-methoxyphenyl)-piperazine (o-MPP) is regarded as an antagonist of the dopamine receptor. Its effects were studied for the hind limb flexor reflex in the spinal rat, the body temperature in rats maintained at ambient temperature of 28 degrees C and stereotypy induced in rats by a low dose of apomorphine (0.5 mg/kg). o-MPP (2-12.5 mg/kg) stimulated the flexor reflex and, in higher doses (10-40 mg/kg), elevated the body temperature of heat-adapted rats. Both effects were inhibited by central serotonin receptor blocking agents--cyproheptadine or metergoline. Low doses of o-MPP (below 5 mg/kg) did not affect stereotypy induced by apomorphine; higher doses (5-25 mg/kg) attenuated the stereotypy in a dose-dependent manner. The results obtained suggest that o-MPP may facilitate the central serotonergic transmission similarly as several other phenylpiperazines. The role of this effect in the "antidopamine" action of o-MPP is discussed.

A comparative pharmacological study of trazodone, etoperidone and 1-(m-chlorophenyl)piperazine.

Since 1-(m-chlorophenyl)piperazine (mCPP) is a metabolite of trazodone (TRZ) and etoperidone (ETO), two atypical antidepressants, a pharmacological study was undertaken to establish the possible contribution of mCPP to the effects of the parent compounds. Behavioral effects of mCPP in rats consist in head shakes and other signs of serotoninergic stimulation; subtoxic doses also produce clonic convulsions and prostration. TRZ and ETO produce sedation and signs of alpha-adrenergic blockade; subtoxic doses produce tremors, clonic convulsions and prostration. Peripheral effects of norepinephrine (NE) and serotonin (5-HT) in rats are potentiated by mCPP and inhibited by TRZ and ETO. 5-hydroxytriptophan (5-HTP)-induced head twitches in mice are inhibited by TRZ and ETO and unaffected by mCPP. At similar doses mCPP, TRZ and ETO inhibit some nociceptive responses in rats and mice.

Treatment of Syphacia obvelata in the Syrian hamster (Mesocricetus auratus) with piperazine citrate.

The effective dose for eradication of Syphacia obvelata infestation in Syrian hamsters (Mesocricetus auratus) was determined and a colony of 2,278 hamsters housed in 751 cages were treated. Two treatment courses with 10 mg of piperazine citrate/ml of drinking water for 7 days, separated by 5 days without treatment, achieved eradication of parasites previously demonstrated by perianal fecal smears containing ova.

Comparative anthelminthic efficacy and safety of Caesalpinia crista seed and piperazine adipate in chickens with artificially induced Ascaridia galli infection.

The antiascarid activity of Caesalpinia crista Linn. seeds, popularly known as Karanjwa, was evaluated in chickens of the Fumi breed, suffering from artificially induced Ascaridia galli infection. Eggs per gram (EPG) counts were determined in the droppings of chickens prior and after treatment with powdered C. crista at doses of 30, 40 and 50 mg/kg of body weight along with its extracts in water and methanol in amounts representing 50 mg/kg of crude powder. The crude drug at the dose rates of 40 and 50 mg/kg and its methanol extract induced a significant (P < 0.001) effect on post-treatment days 10 and 15 while the 30 mg/kg dose was efficacious (P < 0.05) on day 15 only. However, the aqueous extract did not show significant results. These results suggest that a 50 mg/kg dose of C. crista seed powder, its equivalent methanolic extract and piperazine (200 mg/kg) are equieffective in treating the ascarid infection of poultry. The crude C. crista powder appears to be potent and safer than its methanol extract on the basis of the side effects observed.

4-(2'-Methoxy-phenyl)-1-[2'-(n-2"-pyridinyl)-p-iodobenzamido]-ethyl- piperazine ([125I]p-MPPI) as a new selective radioligand of serotonin-1A sites in rat brain: in vitro binding and autoradiographic studies.

Binding characteristics of a radioiodinated serotonin-1A (5-HT1A) receptor antagonist, 4-(2'-methoxy-phenyl)-1-[2'-(n-2"-pyridinyl)-p- iodobenzamido]-ethyl-piperazine ([125I]p-MPPI) were evaluated using in vitro homogenate binding and autoradiographic techniques in rat brains. [125I]p-MPPI displayed a Kd value of 0.32 +/- 0.04 nM (in the presence of MgCl2) and a Bmax value of 315 +/- 60 fmol/mg of protein in rat hippocampal homogenates. The number of 5-HT1A receptors labeled by [125I]p-MPPI was 40% higher than that labeled by trans-8-hydroxy-2-(N-n-propyl-N-3'-iodo-2'- propenyl)aminotetralin ([125I]R(+)8-OH-PIPAT) (225 +/- 47 fmol/mg of protein), a radioiodinated 5-HT1A agonist. The magnesium ion showed an inhibitory effect on [125I]p-MPPI binding but increased the specific binding of [125I]R(+)8-OH-PIPAT. A significant increase in Bmax values in the presence of guanyl nucleotides was observed for [125I]p-MPPI (control, 307 +/- 35 fmol/mg of protein; with GTP, 345 +/- 30 fmol/mg of protein; with guanylyl-imidodiphosphate, 362 +/- 35 fmol/mg of protein); however, both guanyl nucleotides significantly reduced the Bmax values measured by [125I]R(+)8-OH-PIPAT (control, 213 +/- 50 fmol/mg of protein; with GTP, 133 +/- 20 fmol/mg of protein; with guanylyl-imidodiphosphate, 108 +/- 20 fmol/mg of protein). The binding characteristics of [125I]p-MPPI for 5-HT1A receptors suggest that p-MPPI is an antagonist for 5-HT1A receptors. In vitro autoradiographic studies in rat brain sections with [125I]p-MPPI showed specific labeling of areas rich in 5-HT1A receptors and the regional distribution closely matched those labeled by [125I]R(+)8-OH-PIPAT.(ABSTRACT TRUNCATED AT 250 WORDS)

Structure-activity relationship studies of CNS agents. Part 14: Structural requirements for the 5-HT1A and 5-HT2A receptor selectivity of simple 1-(2-pyrimidinyl)piperazine derivatives.

The 5-HT1A and 5-HT2A receptor affinity of model 1-(2-pyrimidinyl)-piperazine derivatives 15-21 and 23-32 has been determined. 2-(N-Methylpiperazino)-4,6-di(2-thienyl)pyrimidine 26 is a new, highly active and selective 5-HT2A receptor ligand. The topography of a molecule and the stereoelectronic effects of the thiophene rings are the major factors responsible for the high affinity and selectivity of 26 towards 5-HT2A sites.

Pharmacokinetics of 14C-N-N'-dicyclopropyl-methyl-piperazine-dichlorhydrate. 3rd communication: whole-body autoradiographic study in Cynomolgus monkeys after oral administration.

A distribution study was carried out on Cynomolgus monkeys (Macaca fascicularis) dosed orally with 14C-labelled N-N'-dicyclopropyl-methyl-piperazine-dichlor-hydrate 14C-Ino 2628-CZ and sacrificed 2, 8 and 24 h after dosage, using whole-body autoradiography. It is demonstrated that radioactivity is found in all the body; high intakes occur in melanin containing tissues, in blood-forming organs, and in accessory genital glands. Moreover, radioactivity crosses the blood-brain barrier and is mainly localized in hippocampus, caudate, putamen and frontal cortex.
